Bridging Pancreatic Amyloidosis and Neurodegeneration: The Emerging Role of Amylin in Diabetic Dementia.

A hallmark of type 2 diabetes mellitus (T2DM) is the presence of abundant amyloid deposits composed of amyloid polypeptide (amylin) within the pancreatic islets of Langerhans. Given its high prevalence among diabetic individuals, human amylin fibrillization has long been considered a key pathogenic factor in T2DM. Co-secreted with insulin, amylin can misfold and aggregate, inducing β-cell toxicity, impairing insulin secretion, and accelerating disease progression. Emerging evidence also indicates that amylin accumulates in the brains of patients with Alzheimer's disease, where it may interact

AIM2 positively regulates B cell activation and function through the SNX9-PI3K-WASP axis.

AIM2, an inflammasome sensor, has been extensively investigated for its ability to induce pyroptosis in macrophages. However, its role in the adaptive immune system remains poorly studied, particularly in B cells. AIM2 knockout mice had decreased follicular (FO) and marginal zone (MZ) B cell subsets and impaired IgG3 switching. The activation of B cells enhanced the co-localization of AIM2 and BCR. Interestingly, AIM2 exerts dual regulatory effects on BCR signaling transduction by positively regulating the PI3K-AKT signaling axis and negatively regulating the BTK-NFκB signaling axis. Through i

Hyperuricemia in Chronic Kidney Disease: Emerging Pathophysiology and a Novel Therapeutic Strategy.

Hyperuricemia has been increasingly recognized as a modifiable contributor to chronic kidney disease (CKD) progression. Although the traditional classification of hyperuricemia distinguished between renal underexcretion and renal overload types, recent studies suggest that hyperuricemia in patients with CKD can result from heterogeneous excretory defects, including glomerular under-filtration and tubular over-reabsorption. These distinct phenotypes may drive divergent renal injury mechanisms. Experimental and clinical data reveal that monosodium urate crystals and soluble uric acid independent

Emerging molecular targets in deep vein thrombosis: from inflammation to coagulation.

Deep vein thrombosis (DVT), a prevalent vascular disorder driven by venous stasis, endothelial injury, and hypercoagulability, imposes a significant global health burden due to life-threatening complications like pulmonary embolism. Recent advances highlight inflammation as a pivotal contributor to DVT pathogenesis, intricately linked with coagulation through immunothrombosis. This review synthesizes emerging molecular targets bridging these pathways, focusing on neutrophil extracellular traps (NETs), peptidylarginine deiminase 4 (PAD4), P-selectin, high-mobility group box 1 (HMGB1), tissue fa

A Lipopolysaccharide-Directed Piezoelectric Sonosensitizer for Addressing Deep Periodontal Pocket Infections.

Complete removal of dental plaque biofilms and associated pathogens from deep periodontal pockets, coupled with subsequent inflammation resolution, constitutes a pivotal challenge in current periodontitis therapy. Here, a targeted bactericidal and gas-based anti-inflammatory strategy to regulate the periodontal microenvironment was established. The strategy aimed to eradicate deep-seated infections by modifying piezoelectric bimetallic Ti/Mn-MOFs nanosheets with phenylboronic acid (PBA) and l-arginine (l-arg). PBA mediated chemical targeting against periodontal pathogens via lipopolysaccharide

Targeting the DYNLL2-PAK1 axis inhibits caspase-11-dependent pyroptosis to alleviate sepsis.

Sepsis, a life-threatening syndrome caused by dysregulated host responses to infection, lacks effective therapeutic strategies due to its complex immune pathophysiology. Here, we identify Dynein Light Chain LC8-Type 2 (DYNLL2) as a critical risk gene driving sepsis progression through bioinformatics and machine learning analysis of clinical datasets. Elevated DYNLL2 expression correlates with poor prognosis and monocyte expansion in sepsis patients. Mechanistically, DYNLL2 interacts with p21-Activated Kinase 1 (PAK1) to regulate the endocytosis of Gram-negative bacterial outer membrane vesicle

Uncovering immune pathways for therapeutic targeting of hypertension.

INTRODUCTION: Hypertension is a major health problem worldwide, yet fewer than half of patients maintain adequate blood-pressure control, pointing to hidden pathogenic drivers and therapeutic gaps. Normal pressure regulation depends on seamless cross-talk among the kidney, vasculature, brain, and gut; once this dialogue falters, low-grade, T cell-centered inflammation sustains disease. AREAS COVERED: In the kidney, CD8 + T cells bearing the purinergic receptor P2X7sense extracellular ATP and hypertonicity, upregulate renal sodium transporters, and lock in salt. Perivascular adipose tissue mob

Molecular insights into α-Lactalbumin-Chlorogenic acid interaction: Peptide design and anti-inflammatory mechanisms.

In this study, we systematically explored the molecular interaction mechanism between α-lactalbumin (ALA) and chlorogenic acid (CA), and designed the functional peptide TQVYW based on key interfacial residues (THR33, GLN54, VAL99, TYR103, and TRP104) identified from ALA through molecular docking with CA. Furthermore, we elucidated the synergistic anti-inflammatory mechanism of the peptide TQVYW in combination with CA. The study showed that ALA binds CA via hydrogen bonds and hydrophobic interactions, forming a stable molten globule. Further studies showed that TQVYW-CA suppressed reactive oxyg

Inhibition of NLRP3 alleviates calcium oxalate crystal-induced renal fibrosis and crystal adhesion.

Kidney stones are gaining attention as one of the most common urological diseases. In this study, we first constructed a mouse model of calcium oxalate (CaOx) crystal deposition by intraperitoneal injection of glyoxalate (Gly) and found that the levels of NLRP3, CASP1 and ASC, which constitute the NLRP3 inflammasome, as well as the level of its downstream product, IL-1β, were elevated in the kidneys of the model group of mice, as determined by RNA-seq. We then examined NLRP3 expression via immunohistochemistry, immunofluorescence, qPCR, and Western blotting in human samples, calcium oxalate mo

Structure of the human TWIK-2 potassium channel and its inhibition by pimozide.

The potassium channel TWIK-2 is crucial for ATP-induced activation of the NLRP3 inflammasome in macrophages. The channel is a member of the two-pore domain potassium (K2P) channel superfamily and an emerging therapeutic target to mitigate severe inflammatory injury involving NLRP3 activation. We report the cryo-EM structure of human TWIK-2. In comparison to other K2P channels, the structure reveals an unusual "up" conformation of Tyr111 in the selectivity filter and a resulting SF1-P1 pocket behind the filter. Density for acyl chains is present in fenestrations within the transmembrane region

Hot phase episodes in arrhythmogenic cardiomyopathy: More than just a desmoplakin issue?

Arrhythmogenic cardiomyopathy (ACM) is a genetically determined myocardial disease characterized by myocyte loss, fibro-fatty replacement, and electrical instability. In a subset of patients, episodes of chest pain with troponin release and electrocardiographic abnormalities occur in the absence of ischemic causes. These events, commonly referred to as "hot phases" (HP), often mimic acute myocarditis and raise important diagnostic and prognostic considerations. Among ACM-related genes, desmoplakin (DSP) variants are most frequently associated with HP, although episodes have also been observed

Morroniside Ameliorates High-Fat and High-Fructose-Driven Chronic Kidney Disease by Motivating AMPK-TFEB Signal Activation to Accelerate Lipophagy and Inhibiting Inflammatory Response.

Studies have substantiated that dietary-fat- and fructose-overconsumption-caused lipid metabolism disorders can trigger renal lipotoxicity to drive the progression of chronic kidney disease (CKD). This study was conducted to evaluate the efficacy of morroniside, a natural active substance extracted from the fruit of Cornus officinalis, in inhibiting the progression of CKD in high-fat and high-fructose-fed mice. Our results showed histological changes such as fatty degeneration of renal tubular cells, tubular dilatation, glomerular fibrosis, and abnormal renal function in the kidneys of high-fa

Mineralocorticoid receptor antagonists promote renal immunosenescence.

Chronic kidney disease (CKD) is often associated with chronic inflammation, influenced by the activation of mineralocorticoid receptors (MR). This review focuses on changes in immune cells and explores the important role that MR antagonists (MRAs), especially the new nonsteroidal MRA, finerenone, play in alleviating renal and cardiac injury by affecting the transformation of stimulated immune cells. We found that MR can promote the transformation of macrophages to M1 pro-inflammatory phenotype through IL-14 receptor and mitogen-activated protein kinase (MAPK)-JNK. MR also activates helper T ce

Unraveling chemotherapy-evoked hepatic dysfunction: a deep dive into cyclophosphamide-related liver injury.

Cyclophosphamide (CPA) is an alkylating drug utilized in the treatment of several cancers and autoimmune illnesses. Liver injury is a serious adverse effect linked to the administration of CPA. Nonetheless, the mechanism behind this toxicity remains incompletely elucidated; mechanistic investigations have identified oxidative stress, inflammatory responses, and apoptosis as pivotal elements contributing to CPA-induced liver dysfunction. In addition, CPA triggers the production of reactive oxygen species that act as damage-associated molecular patterns that rapidly activate TLR4/MYD88/NF-κB and

Cilastatin Attenuates Acute Kidney Injury and Reduces Mortality in a Rat Model of Sepsis.

Sepsis is a life-threatening condition caused by an abnormal host response to infection, leading to organ dysfunction and potentially death. Acute kidney injury (AKI) is a critical complication of sepsis. Various pathways, especially signaling through Toll-like receptors (TLRs) and the nucleotide-binding oligomerization domain, leucine-rich repeat and pyrin domain-containing protein 3 (NLRP3) inflammasome, contribute to inflammation and tissue damage. Cilastatin, a renal dehydropeptidase I inhibitor, has shown promise in protecting against AKI induced by nephrotoxic drugs. This study assessed

GDF15 nanotherapy ameliorates NLRP3-associated redox imbalance and cardiac injury in sepsis.

Sepsis-induced cardiomyopathy (SICM) is a life-threatening complication of sepsis, characterized by acute cardiac dysfunction and high mortality. Despite its prevalence and severity, SICM lacks effective targeted therapies. Therefore, we developed macrophage membrane-coated PLGA nanoparticles (MGP) for the targeted delivery of recombinant human GDF15 (rhGDF15), aiming to enhance its therapeutic efficacy. In this study, MGP was designed to encapsulate rhGDF15. MGP displayed favorable stability and biocompatibility in vitro and significantly improved left ventricular function and contractility i

The circadian regulator PER1 inhibits osteoclastogenesis by activating inflammatory genes.

Disruption of circadian rhythms predisposes shift workers to many chronic conditions, including osteoporosis. However, the effects of disrupted circadian rhythms on bone remodeling remain largely unknown. Here, we show that one of the core circadian regulators PER1 inhibits osteoclastogenesis by upregulating genes involved in inflammation. The conditional knockout of Per1 in osteoclasts and related cells resulted in decreased bone mass in the femurs of mice, along with increased osteoclasts and decreased osteoblasts. Osteoclastogenesis was also promoted by Per1 depletion in vitro with 17 downr

Tailoring UPS-to-autophagy transition to restore RGC prosperity and functionality in a rodent traumatic optic neuropathy model.

Direct or indirect injury to the optic nerve can lead to traumatic optic neuropathy (TON), a pathological process eventually resulting in long-term vision loss. Experimentally, optic nerve crush (ONC) simulates TON by inducing retinal ganglion cell (RGC) apoptosis and vision loss. This study investigates the neuroprotective effect of a HECT domain-E3 ubiquitin ligase inhibitor, Compound 056, on retinal ganglion cells after traumatic injury. Experimental traumatic optic neuropathy (TON) was induced by mechanical compression of the rodent optic nerve. Compound 056 was administered (60 mg/kg) sub

Deciphering the role of cell signaling pathways in gout pathogenesis and the therapeutic potential of phytoconstituents in their modulation.

PURPOSE: Gout, a type of inflammatory arthritis, arises from the accumulation of monosodium urate crystals in joints, leading to severe pain and inflammation. While conventional treatments, such as uric acid-lowering agents and anti-inflammatory drugs, are effective, they are often associated with adverse effects. This review aims to explore the potential of phytoconstituents as alternative therapeutic agents for gout, focusing on their mechanisms of action and strategies to enhance their clinical efficacy. METHODS: A comprehensive literature review was conducted to analyze the role of phytoc
