Glioblastoma (GBM) is a devastating brain tumor with limited treatment success, partly because in vitro models poorly mimic in vivo complexity. This study introduces a high-throughput 3D culture platform utilizing modular starPEG-glycosaminoglycan (GAG) hydrogels that enable independent control of extracellular matrix (ECM) cues: stiffness, cytokine affinity, matrix metalloproteinase-responsive remodeling, and cell adhesiveness via integrin-binding RGD peptides. This platform supports encapsulation of patient-derived GBM cells, recreates physiologically relevant tumor microenvironments in 384-

BACKGROUND: Serine-threonine kinase 10 (STK10) is a member of Ste20 family of serine/threonine kinases. Our previous study showed STK10 is expressed in platelets and regulates platelet function in arterial thrombosis. Whether it plays a role in venous thrombosis remains unclear.
OBJECTIVES: In this study, we aim to investigate the role of platelet STK10 in deep vein thrombosis (DVT) by using platelet-specific STK10 knockout mice.
METHODS: A DVT model was constructed via ligation of the inferior vena cava, and 24 hours later, an inferior vena cava sample was obtained for analysis of thrombi l

The myotendinous junction (MTJ) is a critical interface between muscle fibers and tendons, essential for force transmission between muscle and bone. Laminin-α2, a key extracellular matrix (ECM) component, is strongly enriched at this interface. Mutations in the LAMA2 gene cause LAMA2-related muscular dystrophy (LAMA2 MD), an early-onset severe congenital muscular dystrophy. Here, we examined the MTJ in dyW/dyW mice, a mouse model for LAMA2 MD. We find a strong disruption of MTJ morphology, including altered muscle fiber tips, collagen XXII mislocalization, and reduced muscle tendon interface.

Junken meat sheep display distinct coat-color phenotypes, including a white body with a black head and an entirely black coat at birth, but the transcriptional basis of variation across skin regions and phenotypic backgrounds remains unclear. We performed bulk RNA sequencing (RNA-seq) on nine skin samples from six 1-month-old lambs: black-haired head skin (BT), white-haired dorsal trunk skin (WT), and black-haired dorsal trunk skin from lambs born entirely black (B), with three biological replicates per group. The paired BT-WT comparison characterized within-individual regional differences, wh

Acute kidney injury (AKI) often leads to incomplete recovery of renal function, progressing to chronic kidney disease (CKD). Key pathological features in the AKI-CKD transition include microvascular rarefaction and fibrosis. However, the direct effects of activated fibroblasts on microvasculature and endothelial cells remain unclear. We constructed a single-cell RNA sequencing (scRNA-seq) database from unilateral ischemia reperfusion injury (uIRI) mouse model and identified five heterogeneous fibroblast subpopulations, with C0-Sema3dhi fibroblasts significantly increasing post-injury and corre

BACKGROUND: Biologically active glasses (BG) are widely studied as potential materials for bone tissue engineering, owing to their capacity to form hydroxyapatite (HA) layers when exposed to body fluids. However, the demerit of BG is the slow deposition rate of calcium ions on the surface, a key factor in increasing the conversion rate to mineralization. The purpose of this study is to introduce phosphoryl group recognized as a trigger for the nucleation processes of apatite formation on the surface of BG.
METHODS: BG was coated with poly (2-methacryloyloxyethyl phosphorylcholine) (PMB) and c

Wound healing in diabetic patients is severely compromised by persistent infection, impaired tissue integrity, and chronic inflammation. Here, a thermoresponsive magnetic hydrogel dressing (MHd; PNIPAm-PAm/Laponite/Fe3O4/TA) is developed to enable magnetically programmable, time-sequenced thermos-mechano-biochemical regulation for infected diabetic wounds. MHd integrates a temperature-sensitive polymer network, magnetic Fe3O4 nanoparticles, and the natural polyphenol tannic acid (TA), allowing external control under an alternating magnetic field (AMF). By temporally adjusting AMF intensity, MH

BACKGROUND: Fibrosis is a hallmark of organ failure observed after chronic epithelial injury and inflammation. The transforming growth factor beta (TGF-β) is the master regulator of fibrogenesis, so blockade of the TGF-β pathway is a potential treatment strategy for fibrosis; however, the therapeutic potential of pan-TGF-β blockade is limited by side effects.
METHODS: We generated SOF10, a humanized antibody that targets latent TGF-β1 and selectively blocks protease- and integrin αvβ8-mediated latent TGF-β1 activation. We conducted gene expression and histological analyses in nonalcoholic ste

To evaluate the impact of low-level laser-based photobiomodulation (PBM) on cellular homeostasis and oncology safety by examining viability and proliferation in gingival fibroblasts (HGFs), human dermal fibroblasts (HDFs), and MCF-7 breast cancer cells under different irradiation conditions, and by assessing the signaling-related protein expression in HGFs. The HGFs, HDFs, and MCF-7 cells were exposed to 808 nm diode laser irradiation at 2 and 3 J/cm², applied immediately or 24 h after seeding or continuous dose specifically for MCF-7. Cell viability and metabolic activity were assessed using

The precise intraoperative delineation of tumor margins and the controlled intratumoral release of chemotherapeutic agents remain major challenges in breast cancer therapy. Tumor microenvironment (TME)-responsive nanoplatforms provide a promising approach by utilizing the distinctive biochemical properties of tumors to achieve targeted imaging and treatment. Herein, we developed a glutathione (GSH)-activatable theranostic nanoprobe (DSM-cRGD) emitting fluorescence in the second near-infrared window (NIR-II). The probe is constructed from Nd-doped rare-earth nanoparticles coated with a manganes

Blood deficiency syndrome (BDS) is a systemic disorder characterized by hematopoietic dysfunction and immune dysregulation. Given the limitations of current therapies, such as single efficacy and adverse effects, there is an urgent need for multitarget therapeutic agents with systemic regulatory effects. In this study, a mouse model of BDS was established through chronic benzene inhalation. Using an integrated transcriptomics and proteomics approach, we systematically investigated the therapeutic mechanism of Danggui Buxue Decoction (DBD). The results demonstrated that DBD significantly restor

Tumor-associated macrophages (TAMs), particularly the M2-like subtype, contribute to an immunosuppressive tumor microenvironment and promote tumor progression. Melittin, a natural bioactive peptide derived from bee venom, has been reported to exert anticancer and immunomodulatory effects; however, the cellular targets and molecular mechanisms underlying its selective action on TAMs remain incompletely defined. This study aimed to investigate the immunomodulatory effects of melittin on TAM subsets and to identify the molecular determinant responsible for its selectivity toward M2-like macrophag

Cancer stem cell (CSC) models are essential for understanding tumor progression and evaluating targeted therapies. However, conventional approaches rely on sphere formation, destructive characterization, and re-culture steps, resulting in prolonged workflows and limited reproducibility (15-30 days). Herein, we report an engineered functional peptide hydrogel interface that seamlessly integrates rapid CSC induction with in situ pathway monitoring and drug evaluation. The self-assembled Fmoc-diphenylalanine (Fmoc-FF) hydrogel provides a 3D microenvironment, while co-assembly with Fmoc-RGD peptid

Idiopathic nephrotic syndrome (INS) is a podocyte disease triggered by immune-derived factors. Endothelial activation occurs in this context, but whether the activated endothelium contributes to podocyte injury is unknown. We tested the hypothesis that CD93, a protein primarily expressed in the endothelium, is a contributory factor of podocyte injury. We studied 460 patients with INS and 150 with other podocytopathies. CD93 was analyzed in kidney tissue, urine, and serum samples. We tested the efficacy of CD93 blockade in vitro and in vivo and investigated the relationship between soluble CD93

BACKGROUND: Pregnant women, as well as patients with endometriosis and some ovarian tumors, may experience a state of high estrogen levels. However, the role of high estrogen in platelet function and thrombosis remains controversial, which directly affects the prevention and clinical treatment of related diseases.
METHODS: First, we conducted a retrospective analysis of clinical data from 108 pregnant women. Then, the effects of high estrogen levels on thrombosis were examined using the FeCl3-induced mouse mesenteric artery thrombosis model, as well as mouse stroke and pulmonary embolism mode

Androgen receptor signaling inhibitors (ARSIs) have transformed the treatment of advanced prostate cancer, yet durable responses are limited by the emergence of therapy-resistant disease states, including neuroendocrine prostate cancer (NEPC) and double-negative prostate cancer (DNPC). Lineage plasticity has traditionally been studied from a tumor-cell-intrinsic perspective, but single-cell and spatial studies increasingly indicate that the tumor microenvironment, particularly tumor-associated macrophages (TAMs), may influence tumor-cell state transitions, immune exclusion, and therapeutic res

Axial spondyloarthritis (axSpA) is a chronic inflammatory disease characterized by complex immune dysregulation, with T cells playing a central role in its pathogenesis. In this review, we synthesize current knowledge on diverse T cell subsets in axSpA, their pathogenic mechanisms, and emerging therapeutic strategies targeting these cells. We highlight conventional αβ T cell receptor-expressing CD8+ T cells, CD4+ Th17 cells and Treg cells, and CD103+CD49a+ tissue-resident memory integrin-expressing T cells in axSpA initiation and progression. Innate-like T cell subsets, including γδ T cells, m

Soft tissue fibrosis and chronic pain involve a cycle linking extracellular matrix (ECM) stiffening, inflammation, and neural sensitization. This review develops a multiscale framework that spans from molecular mechanisms to organ-level physiology and focuses on the directly mechanosensitive Piezo1/2 channels and the mechano-responsive TRPV4 channel, drawing on their distinct molecular architectures to ground divergent gating properties. We examine how these channels convert membrane tension and integrin-mediated traction into Ca2+ and other ionic signals that, together with Ca2+-independent p

Crustacean shell waste from the seafood industry represents an underutilized food waste with the potential to produce chitosan and chitosan oligosaccharides (COS), with promising applications in bone regeneration. However, the molecular mechanisms of COS during osteogenic differentiation remain uncharacterized. In this study, chitosan was extracted from mud crab (Scylla olivacea) shell waste and COS was prepared from chitosan by acid hydrolysis and then characterized by FT-IR and FESEM, confirming the chitosan backbone and a degree of deacetylation (DD) exceeding 80%. The COS treatment in the

The enthesis links tendon/ligament to bone, forming a soft-hard interface characterized by unique cellular-scale compositional and biomechanical gradients. Enthesis injuries are common and increase with age, yet the lack of biomaterials that faithfully mimic its cellular-scale gradient mechanical environment hinders understanding of the mechanobiological basis of this vulnerability. We showed that the enthesis cellular-scale stiffness gradient progressively flattens with aging. This age-dependent mechanical remodeling diminishes chondrogenesis and may predispose the attachment site to degenera
