Previous studies have shown that the pineal gland of Roman high avoidance (RHA/Verh) rats is larger than that of Roman low avoidance rats (RLA/Verh). In this work we describe the synthesis, Ca+2 channel blockade capacity and antioxidant power of N3,N5-bis(2-(5-methoxy-1H-indol-3-yl)ethyl)-2,6-dimethyl-4-aryl-1,4-dihydropyridine-3,5-dicarboxamides 1-9, a number of multi-target small 1,4-dihydropyridines (DHP), designed by juxtaposition of melatonin and nimodipine. As a result, we have identified antioxidant DHP 7 (Ca2+ channel blockade: 55%, and 8.78 Trolox/Equivalents), the most balanced DHP analyzed here, for potential Alzheimer's disease therapy.

The nuclear orphan receptor RZRbeta is highly expressed in the rat pineal gland. Recent studies proposed that melatonin, the pineal hormone which regulates a wide variety of circadian-linked phenomena, may be the natural ligand of this receptor. These provocative reports prompted us to learn more about RZRbeta and how it might function in circadian physiology. Here we confirm high expression of this receptor in the pineal gland, and report that pineal RZRbeta expression exhibits a strong daily rhythm. Expression is under photoneural regulation and involves an adrenergic --> cAMP mechanism.
