Ubiquitination-dependent regulation of ferroptosis in ischemic heart and brain.

Ischemia-reperfusion (I/R) injury is a major cause of tissue damage after myocardial infarction and ischemic stroke. Ferroptosis is an iron-dependent form of regulated cell death (RCD) marked by phospholipid peroxidation, and it is an important contributor to I/R-related injury. Although oxidative stress and lipid peroxidation are common features of I/R injury, they do not fully explain why ferroptosis sensitivity increases during reperfusion. Recent studies have shown that ferroptosis is also influenced by the ubiquitin system. Changes in ubiquitination and deubiquitination regulate key prote

miR-940 suppresses ferroptosis by controlling expression of key regulatory genes.

Ferroptosis is a form of regulated cell death that is characterized by iron-dependent lipid peroxidation. This process is regulated by specific metabolites, the lipid composition of the cells, redox-active iron, and antioxidant mechanisms. Although numerous regulators have been identified over the past decade, exploring other mechanisms, particularly from non-coding genomic regions, can build a thorough understanding of the multifaceted regulatory processes underlying ferroptosis. MicroRNAs (miRNAs) play a crucial role in gene regulation and cellular functions. Through a CRISPR KO screen, we i

Environmental PFOS and 6:2 Cl-PFESA Reshape Ferroptosis Vulnerability in Liver Cancer.

Ferroptosis, an iron-driven form of programmed cell death characterized by lipid peroxidation and reactive oxygen species (ROS) accumulation, is essential for tissue homeostasis and tumor suppression, yet its regulation in hepatocellular carcinoma (HCC) under environmental pollutant exposure remains unclear. Here, using long-term in vitro and in vivo exposure models with environmentally relevant doses combined with multiomics analyses, we show that per- and polyfluoroalkyl substances (PFAS) exert antithetical effects in HCC and normal hepatocytes. Whereas PFAS promote ferroptosis in normal cel

Multiparametric CEST MRI for Time-Associated Characterization of Acute Ischemic Stroke: NOE (-3.5 ppm) Signal as a Potential Imaging Biomarker.

This study investigated time-dependent group-level changes in chemical exchange saturation transfer (CEST) signals in a mouse model and explored utility in identifying stroke onset time. Multiparametric magnetic resonance imaging (diffusion weighted imaging, arterial spin labeling, and CEST) was conducted on 12 normal and 26 ischemic mice. Imaging findings were validated by immunohistochemical analysis. CEST parameters in infarct and penumbra were compared between strokes within 4.5 h and over 4.5 h. Correlations, regression, and receiver operating characteristic curves evaluated performance.

Exploiting Metal-to-Metal Electron Transfer in a Ru(II) Polypyridine-Deferasirox Conjugate for Hypoxic Photodynamic Therapy.

The unique microenvironment of solid tumors, characterized by pathological hypoxia, remains a primary driver of treatment resistance and poor clinical outcomes in oncology. While photodynamic therapy has emerged as a promising treatment modality, its clinical efficacy is constrained by its dependence on molecular oxygen. Conventional photosensitizers typically operate by oxygen-dependent energy transfer, leading to a marked reduction or complete loss of therapeutic activity within the oxygen-depleted regions of solid tumors. Herein, we show that iron coordination can serve as a molecular switc

Glucolipid metabolic disorders and ferroptosis in diabetic ulcers: pathogenic crossroads and therapeutic opportunities.

Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has emerged as a key player in the pathogenesis of Diabetic Ulcers (DUs). Concurrently, Glucolipid Metabolic Disorders (GLMDs), driven by persistent hyperglycemia and lipotoxicity, constitute a core pathological basis for tissue damage and dysfunction of reparative cells. The interplay between ferroptosis and GLMDs represents a promising yet underexplored research area. This review synthesizes recent advances in understanding the molecular mechanisms underlying their interaction, focusing on how GLMDs dri

4-Hydroxynonenal, a Potential Biomarker for Lung Inflammatory Diseases.

Asthma, chronic obstructive pulmonary disease (COPD), and acute respiratory distress syndrome (ARDS) are the major lung inflammatory complications affecting the global population. Exposure to allergens, infections, smoking, and environmental pollutants could cause persistent oxidative stress and dysregulated immune responses, leading to lung inflammatory complications. Increased oxidative stress can lead to lipid peroxidation and the formation of toxic lipid aldehydes. One of the major lipid aldehydes formed during lipid peroxidation is 4-hydroxy-2-nonenal (4-HNE). 4-HNE is well known to coval

The fruitfly Drosophila melanogaster as an in vivo model of ferroptosis.

Ferroptosis is an iron-dependent form of cell death involved in cancer, ischemia-reperfusion injury, inflammation, and neurodegeneration. It can be triggered in two ways: class 1 inhibitors such as erastin block system xc-, the cystine/glutamate antiporter composed of xCT and 4F2hc/CD98, causing glutathione depletion, whereas class 2 inhibitors such as RSL3 directly inhibit glutathione peroxidase 4, causing lipid peroxidation. Ferroptotic cell death is suppressed by radical-trapping antioxidants like ferrostatin-1. We investigated the effects of erastin and RSL3 on Drosophila melanogaster life

Optimization of anchovy oil microencapsulation using wall materials: effects on oxidative stability and encapsulation efficiency.

Microencapsulation is widely applied to improve the stability of highly unsaturated oils; however, the effectiveness of encapsulation largely depends on the composition of wall materials. This study systematically evaluated the influence of different wall material systems on the encapsulation efficiency and oxidative stability of spray-dried anchovy oil powders. Maltodextrin, sodium caseinate, gum arabic, and whey protein were used individually and in binary combinations as encapsulating matrices. The resulting microcapsules were characterized in terms of encapsulation efficiency, surface oil

Interdroplet Electrostatic Interactions Contribute to Dark Oxidation in Nighttime Sea Sprays.

Nighttime oxidative processing in the marine boundary layer remains poorly understood because conventional photochemical oxidant sources are strongly diminished after sunset. Here we show that freshly emitted sea spray exhibits enhanced oxidative activity under dark conditions, with oxidative signals consistently exceeding those of the corresponding source seawater at three coastal sites in China. Controlled laboratory experiments further demonstrate that salinity strongly suppresses oxidation in isolated droplets, whereas interacting droplet populations retain pronounced hydrogen peroxide and

Effect of acute intraperitoneal injection of methylglyoxal on organ injury and oxidative stress in chicks (Gallus gallus domestics).

Methylglyoxal (MGO) is an α-dicarbonyl compound and produced from several metabolic pathways, including glucose metabolism. This compound induces oxidative stress and organ damage in mammals and is associated with various diseases, including diabetes. Birds typically have higher blood-glucose levels than mammals, yet they rarely develop diabetic complications. This observation suggests that birds possess resistance to MGO, preventing the onset of diabetes-related issues. Nevertheless, the effects of MGO in birds remain poorly understood. In this study, the impact of high-dose MGO affect on org

Di(2-ethylhexyl) phthalate induces male reproductive toxicity through mitophagy-dependent ferroptosis of spermatocytes in mice.

Di(2-ethylhexyl) phthalate (DEHP), a widely utilized plasticizer, impairs male reproductive function; however, the precise mechanisms underlying this effect have yet to be fully elucidated. This study investigates DEHP-induced spermatocyte toxicity and identifies therapeutic strategies. Integrated network toxicology and proteomics delineated testicular toxicity mechanisms through multi-dimensional analyses. We demonstrate that DEHP exposure induces spermatocyte ferroptosis via PINK1/Parkin-mediated mitophagy. Mechanistically, the bioactive metabolite MEHP promotes NRF2 degradation through the

Harnessing autophagy-dependent ferroptosis: Mechanisms and therapeutic opportunities in human diseases.

Ferroptosis represents an iron-dependent form of cell death driven by lipid peroxidation. Autophagy is an evolutionarily conserved lysosomal degradation pathway, and its dysregulation may trigger or facilitate cell death. Autophagy-dependent ferroptosis (ADF) denotes a specific subtype of ferroptosis executed via selective autophagy, which proceeds by boosting labile iron liberation, accelerating lipid peroxidation, and disrupting cellular antioxidant defense systems. Multiple selective autophagic cascades-including ferritinophagy, lipophagy, mitophagy, clockophagy, as well as autophagic turno

Phytoconstituents-mediated Targeting of Ferroptosis for the Treatment of Cardiovascular Disease.

Ferroptosis, an instance of iron-dependent programmable cell death that results from oxidative stress & lipid peroxidation, has garnered interest due to its associations with cardiovascular diseases, such as atherosclerosis, myocardial infarction, as well as heart failure. Unlike necrosis or apoptosis, ferroptosis involves unique metabolic pathways that disrupt cellular redox balance and lipid homeostasis, leading to substantial cell damage in cardiovascular tissues. It is becoming recognized that phytoconstituents-bioactive compounds derived from plants-can modify ferroptosis pathways and pro

MAFF regulates ferroptotic sensitivity through iron homeostasis and fatty acid synthesis.

Ferroptosis is an iron-dependent form of regulated cell death driven by lipid peroxidation. In this study, we identify the transcription factor v-maf musculoaponeurotic fibrosarcoma oncogene homolog F (MAFF) as a key regulator that promotes breast cancer progression while simultaneously sensitizing tumor cells to ferroptosis. Integrated RNA-sequencing and chromatin immunoprecipitation (ChIP)-sequencing revealed that MAFF directly regulates genes involved in iron and fatty acid metabolism. MAFF enhances intracellular iron availability by transactivating SLC11A2 and NCOA4, expanding the labile i

GPX4 in the Tumor Microenvironment: Not Just Inhibiting Ferroptosis, but Immuno-Metabolic Regulation.

Glutathione peroxidase 4 (GPX4) is canonically viewed as the primary suppressor of ferroptosis, yet its role in the tumor microenvironment (TME) extends far beyond antioxidant catalysis to encompass immuno-metabolic regulation. In this review, we synthesize recent advances in enzymology, immunology, and cancer metabolism to propose a "lipid peroxidation threshold" framework, wherein GPX4 sets cell-type-specific thresholds that determine susceptibility to ferroptosis across tumor cells, CD8+ T cells, dendritic cells (DCs), and myeloid populations. We discuss how these thresholds are dynamically

Sympathetic signaling activation alleviated acute respiratory distress syndrome via the HDC/SLC7A11 axis in lipopolysaccharide-induced macrophages.

Acute respiratory distress syndrome (ARDS) represents a severe pulmonary condition characterized by excessive inflammation, wherein alveolar macrophages (AMs), pivotal components of the innate immune system, play a critical role in the pathogenesis of the disease. Despite its high morbidity and mortality, effective targeted therapies for ARDS remain unavailable. Norepinephrine (NE) is an endogenous neurotransmitter with immunomodulatory and anti-inflammatory properties, and has been reported to mitigate ARDS symptoms in sepsis models. While sympathetic signaling exerts protective effects, the

Mercuric Chloride-Mediated Neurotoxicity in Chicken Embryos: Disruption of the Blood-Brain Barrier Integrity and Iron Overload-Induced Ferroptosis.

Mercuric chloride (HgCl2) is an environmental pollutant with neurotoxicity. The blood-brain barrier (BBB) serves as a critical protective interface, shielding the brain from exposure to heavy metals and maintaining neurological homeostasis. Ferroptosis, a programmed cell death initiated by iron-dependent lipid peroxidation, was recently reported to be associated with various neurological disorders. Nevertheless, the relationship between HgCl2-induced neurotoxicity, BBB dysfunction, and ferroptosis remains unclear. This study investigated the effects of embryonic exposure to HgCl2 (35, 50, and
