Nanotechnology - Microbiota synergy in cancer immunotherapy.

The global burden of cancer continues to escalate, yet the efficacy of current targeted therapies and immune checkpoint inhibitors (ICIs) is often hampered by drug resistance and immune escape. The gut microbiota orchestrates tumor progression and ICI efficacy bidirectionally via the "microbiota-immune-tumor" axis. However, traditional interventions, including fecal microbiota transplantation and antibiotics, are limited by inherent drawbacks such as poor targeting. Nanotechnology can overcome this limitation: it not only enables precise targeting of the gut microbiota and reshapes the tumor i

Spatiotemporally Controlled Tumor Photodynamic/Immunotherapy Therapy Based on Upconversion Hybrid Nanosystem.

Immunotherapy holds great promise for cancer treatment, but its clinical application is hindered by the immunosuppressive tumor microenvironment (TME) and the systematic toxicity. Herein, a spatiotemporally controllable smart nanodevice based on upconversion nanoparticles (UCNPs) that can be activated by dual near - infrared (NIR) light for tumor immunotherapy, namely PURH, is developed. PURH is constructed by a five-layer UCNPs surface coated with mesoporous silica, which encapsulates the photosensitizer rose bengal (RB) within the mesoporous channels. Additionally, CpG oligonucleotides (CpG …

Single-cell transcriptomics reveals involution mimicry during the specification of the basal breast cancer subtype.

Basal breast cancer is associated with younger age, early relapse, and a high mortality rate. Here, we use unbiased droplet-based single-cel … n of an aberrant post-lactation developmental program of involution that triggers remodeling of the tumor microenvironment and metastatic dissemination. This involution mimicry is characterized by a highly interactive multicellular network, with involution cancer-associated fibroblasts playing a pivotal role in extracellular matrix remodeling and immunosuppression. On the dual roles and polarized phenotypes of neutrophils in tumor development and progression.

Inconsistencies plague our understanding of the role of neutrophils in cancer and the literature provides evidence for a duality in neutroph … e that cytokines, such as TGF-β and IFN-β, are involved in directing neutrophil polarization by the tumor microenvironment. With the opposite polarity, N2 neutrophils may be detrimental for the host and beneficial for tumor growth, invasion and metastasis, e.g. through proteolysis of extracellullar matrix components, promotion of angiogenesis and mediation of immunosupp …

Dual functional immunostimulatory polymeric prodrug carrier with pendent indoximod for enhanced cancer immunochemotherapy.

Immunotherapy based on checkpoint blockade has been regarded as one of the most promising approaches towards many types of cancers. However, low response rate hinders its application due to insufficient tumor immunogenicity and immunosuppressive tumor microenvironment. To achieve an overall enhanced therapeutic outcome, we developed a dual-functional immuno-stimulatory polymeric prodrug carrier modified with pendent indoximod, an indoleamine 2,3-dioxygenase (IDO) inhibitor that can be used to reverse immune suppression, for co-delivery of Doxorubicin (Dox), a hydrophobic anticancer agent that …

Molecular and microenvironmental networks driving tumor dormancy: pathological mechanisms and emerging clinical strategies.

Although standard therapies including surgery, radiotherapy, and chemotherapy have proven effective in the initial eradication of primary tumors, a substantial proportion of cancer patients remain at persistent threat of recurrence. Accumulating evidence identifies tumor dormancy as a pivotal driver of this clinical challenge. Dormant tumor cells can persist in a quiescent state for extended periods, during which they exhibit marked resistance to conventional therapies and may ultimately trigger disease relapse years after clinical remission. Consequently, deciphering the mechanisms governing

pH-responsive polymeric-peptide complex enhances tumor immunogenic cell death by membrane disruption.

Melittin, the principal bioactive component of bee venom, exhibits potent anticancer activity. However, its clinical application is hindered by nonspecific hemolytic toxicity. In this study, we developed a pH-responsive polymeric-peptide complex (PCM) to improve anti-tumor activity of melittin while minimizing off-target effects. PCM remained structurally stable under physiological conditions (pH 7.4) but underwent rapid disassembly in the mildly acidic tumor microenvironment (pH 6.8) to expose melittin. The exposed melittin bound to tumor cell membranes, compromised membrane integrity through…

Potentiating dendritic cell immunotherapy by interrupting the Semaphorin 4D-induced immune-suppressive barrier.

Immune suppression within the tumor microenvironment (TME) remains a significant barrier to effective cancer immunotherapy, including dendritic cell (DC)-based approaches. We address this by targeting Semaphorin 4D (SEMA4D), a pro-tumorigenic mediator expressed by leading-edge tumor cells and host immune cells, particularly in HER2-positive (HER2pos) breast cancer (BC). Antibody blockade of SEMA4D enhances the efficacy of adoptively transferred tumor-specific conventional type 1 DCs (cDC1s) across multiple HER2pos BC models. Tumor-cell-specific ablation of SEMA4D activity amplifies cDC1-driven…

Modification-Response Integrated Modules Driven Cyclization-Dependent Prodrug Self-Assembly for Reshaping Pro-Apoptotic Tumor Redox Homeostasis.

Affected by the complexity and heterogeneity of the tumor redox microenvironment, chemotherapy often fails to achieve satisfactory clinical outcomes. Modular design of prodrug nanoassemblies presents tremendous potential in upgrading the therapeutic index of chemotherapeutic agents. Given the biochemical vulnerability of tumor redox homeostasis, we fabricated five-membered cyclic chalcogenide-linked paclitaxel prodrug nanoassemblies, which realized tumor site-specific activation and reshaped the pro-apoptotic tumor redox homeostasis. Cyclic diselenide and cyclic disulfide bonds integrated the

4-Arylindolines Bearing a Pyrido[3,2-d]pyrimidine Scaffold as Dual Inhibitors of the PD-1/PD-L1 Interaction and NAMPT for Targeting Tumor Immunoevasion and Metabolism.

Targeting both the PD-1/PD-L1 axis and NAMPT represents a promising therapeutic strategy to overcome resistance to the immune checkpoint blockade. Herein, we developed a novel series of 4-arylindolines bearing a pyrido[3,2-d]pyrimidine scaffold as dual-targeting inhibitors. Among these, compound B14 emerged as the most promising inhibitor, exhibiting IC50 values of 7.5 and 18.8 nM against the PD-1/PD-L1 interaction and NAMPT, respectively, along with favorable metabolic stability. In vitro investigations revealed that B14 significantly enhanced T cell-mediated tumor cell killing, elevated T ce

Reactivating exhausted tumor-infiltrating T cells by a bispecific DC-T cell engager in mice.

Tumor infiltrating T cells (TIL) are key players in the anti-tumor immune response. However, chronic exposure to tumor-derived antigens driv … n to reactivate T cells via IL-2 signalling and co-stimulatory CD80/86-CD28 interactions within the tumor microenvironment. Finally, to mitigate IFNα-induced toxicity, we engineer a Pro-BiDT engager featuring a pro-IFNα and report potent antitumor activity with reduced systemic toxicity. Thus, by bridging DC-T cells together, BiDT treatment enhances the critical communi …

The molecular mechanisms of platelets in tumor invasion and metastasis: a controversial role.

Platelets, traditionally recognized for their role in hemostasis, are now understood to actively contribute to tumor progression, angiogenesis, and metastasis. Through dynamic bidirectional interactions with cancer cells, platelets undergo measurable alterations in count, mean platelet volume, and molecular cargo, including proteins and mRNAs, supporting their potential utility as liquid biopsy biomarkers for early cancer detection. Within the tumor microenvironment, platelets contribute to immune evasion, support tumor cell intravasation and extravasation, enhance the survival of circulating …

MALT1 protease inhibition restrains glioblastoma progression by reversing tumor-associated macrophage-dependent immunosuppression in mice.

MALT1 protease is an intracellular signaling molecule that promotes tumor progression via cancer cell-intrinsic and cancer cell-extrinsic me … GBM tumor growth. The addition of MALT1 inhibitor to temozolomide reduces immunosuppression in the tumor microenvironment, indicating that pharmacological inhibition of MALT1 protease may enhance the efficacy of chemotherapeutic. Thus, our findings suggest that MALT1 protease inhibition represents a promising macrophage-targeted immunotherapeutic strategy for the treat …

YTHDFs as radiotherapy checkpoints in tumor immunity.

Radiotherapy (RT), a cornerstone of cancer treatment, exerts its therapeutic effects primarily by inducing DNA damage in tumor cells and modulating the tumor immune microenvironment (TIME). Despite its efficacy, RT is often counteracted by tumor-intrinsic mechanisms, such as DNA damage repair, as well as immune-suppressive responses. YTHDF proteins, key N6-methyladenosine (m6A) readers, have emerged as pivotal regulators of tumor progression, DNA repair, and immune cell function, making them promising targets for enhancing RT efficacy. In this review, we explore the dual roles of YTHDF protein

Single-cell transcriptomic analysis reveals metastatic and immunosuppressive characteristics in meningioma brain-tumor interface.

BACKGROUND: Meningioma is a common primary intracranial tumor with high recurrence and metastasis rate. Delineating the pathological ecosystem at the brain-tumor interface (BTI) of meningioma is critical for understanding the mechanisms of tumor metastasis and developing effective new therapies. METHODS: To identify biomarkers of early metastasis and discover potential therapeutic targets, we integrated single-cell transcriptome datasets of meningioma, and identified the cell populations and molecular signatures uniquely present at the BTI. RESULTS: A specific BTI-enriched tumor cell populat

Long-term relapse-free survival enabled by integrating targeted antibacteria in antitumor treatment.

The role of tumor-resident intracellular microbiota (TRIM) in carcinogenesis has sparked enormous interest. Nevertheless, the impact of TRIM-targeted antibacteria on tumor inhibition and immune regulation in the tumor microenvironment (TME) remains unexplored. Herein, we report long-term relapse-free survival by coordinating antibacteria with antitumor treatment, addressing the aggravated immunosuppression and tumor overgrowth induced by TRIM using breast and prostate cancer models. Combining Ag+ release with a Fenton-like reaction and photothermal conversion, simultaneous bacteria killing and…

Chemokines driven ovarian cancer progression, metastasis and chemoresistance: Potential pharmacological targets for cancer therapy.

Ovarian cancer is a leading cause of death among women globally often characterized by poor prognosis and aggressive tumor growth. The therapeutic outcomes of ovarian cancer patients are majorly limited by the development of acquired chemo/radioresistance and the lack of targeted therapies. The tumor microenvironment (TME) comprises a diverse population of cells including adipocytes, fibroblasts, tumor cells, and immune cells which play an imperative role in promoting tumor growth, invasion, and malignant phenotypes of cancer cells. The cells present in TME secrete various inflammatory mediato…
