Primary central nervous system lymphoma following transfer of human peripheral blood lymphocytes into SCID mice.

Severe combined immunodeficiency (SCID) mice are genetically deficient in both B and T cells. To study immune-mediated phenomena in the CNS, myelin basic protein-reactive T cell clones, admixed with peripheral blood lymphocytes as a source of antigen-presenting cells, derived from a healthy human donor, were injected intracerebrally (IC) into 10 SCID mice. One mouse developed quadriplegia 2 months after the last injection. Autopsy revealed marked meningeal and parenchymal infiltration by large cell lymphoma. There was no evidence of lymphoma outside of the CNS. The majority of the tumor cells

Human peripheral blood lymphocytes transplanted into SCID mice constitute an in vivo culture system exhibiting several parameters found in a normal humoral immune response and are a source of immunocytes for the production of human monoclonal antibodies.

Human PBL from vaccinated healthy blood donors, which was transplanted i.p. into mice with severe combined immunodeficiency (SCID), exhibited an Ag-dependent humoral Ir against tetanus toxoid. This Ir was dose dependent and was completely abrogated by immunizing with large amounts of Ag, suggesting a high dose tolerization of the B cells. A dose-dependent selection of specific, high affinity B clonotypes was also suggested, since immunization with low concentrations of tetanus toxoid produced antisera with higher avidity than immunizations using a high dose of Ag. The production of human Ig an

Induction of colony-stimulating factor(s) after administration of a traditional Chinese medicine, xiao-chai-hu-tang (Japanese name: shosaiko-to).

Colony stimulating factor (CSF)-rich serum was obtained from mice injected intraperitoneally (ip) with shosaiko-to, a traditional Chinese herbal medicine. Transfer of the CSF-rich serum into naive mice augmented the resistance against Listeria monocytogenes. A dose-dependent induction of CSF was observed in mice given shosaiko-to via intravenous route as well as via intraperitoneal route. Since the serum CSF induction was observed in both lipopolysaccharide (LPS)-responder C3H/He mice and LPS-non-responder C3H/HeJ mice, the effect of shosaiko-to seemed to be independent of possibly contaminati

Genome editing of the rutin-degrading enzyme gene FtRDE2 reduces seed bitterness in Tartary buckwheat.

Tartary buckwheat (F. tataricum) is a climate-resilient pseudocereal, yet its global adoption is constrained by its bitter seeds. Here, we employed the CRISPR/Cas9 system to disrupt the rutin-degrading enzyme 2 gene (FtRDE2), which catalyses the hydrolysis of rutin to quercetin, a major contributor to bitterness. Two independent ftrde2 mutant lines showed no detectable rutinosidase activity: mutant seed extracts maintained stable rutin levels and lacked the rapid accumulation of quercetin observed in wild-type controls. Consistent with these biochemical changes, sensory evaluation showed that …

Improving oral cancer surgery by objective intraoperative assessment of resection margins: a new era.

PURPOSE OF REVIEW: Achieving adequate resection margins (i.e., ≥5 mm of healthy tissue surrounding the tumor) in oral cavity squamous cell carcinoma (OCSCC) is difficult. This review discusses recent developments to guide surgical resection. It highlights the transition from the subjective conventional approaches to emerging, objective photonics-based methods. RECENT FINDINGS: Specimen-driven intraoperative assessment of resection margins (IOARM) has significantly improved surgical outcomes. However, IOARM is subjective; moreover, it lacks widespread adoption due to reliance on a dedicated te…

Engineering Complex Molecular Intercalation for Efficient, Sustainable Exfoliation of Van Der Waals Layered Materials.

Intercalation-based exfoliation offers a cost-effective approach to producing 2D materials. While molecular intercalation surpasses ionic methods in preserving the crystallinity and intrinsic properties of 2D materials, its broader adoption is limited by weak driving forces and slow kinetics. To address these challenges, we propose an interlayer modulation strategy using H3PO4-amine complexes as mediators. Structural and molecular dynamics analyses demonstrate that these complexes regulate interlayer proton activity through strong hydrogen bonding, thereby optimizing interlayer dynamics to ena…

[The rise of genetic therapies: urgent moral and societal questions].

Genetic therapies offer long-awaited hope for patients with rare genetic diseases and are emerging as cancer treatment as well. While these innovations appear promising, they are also extremely expensive and raise concerns about how they fit into an already overburdened healthcare system. What are the societal implications when these treatments become available? And how can we guide their adoption in the Dutch healthcare system in an ethically responsible manner? We describe the emerging landscape of genetic therapies, and specifically examine the ethical concerns surrounding their implementat…

Establishing a robotic kidney transplant program for obese recipients.

The integration of minimally invasive and robotic techniques into transplant surgery has led to reduced perioperative morbidity and shortened hospital stays. Robotic-assisted kidney transplantation (RAKT) is now established across multiple transplant centers, offering expanded access to care—particularly for patients with elevated body mass index (BMI), a population traditionally underserved by conventional surgical approaches. Despite its growing clinical promise, widespread adoption of RAKT remains limited, primarily due to the absence of a structured implementation framework. In this review…

Responsibility cannot be abdicated: a mandate for Patient-Centred Precision Care in perioperative brain health.

The perioperative period represents both a major driver of healthcare costs and a substantial source of largely preventable neurological harm. Postoperative delirium affects up to 50-70% of specifically vulnerable surgical cohorts, but adoption of physiologically aligned, low-cost preventive strategies is often delayed until supported by large definitive trials. In contrast, reactive and costly treatments are frequently implemented despite weaker evidentiary standards. This inverted evidence hierarchy creates perverse economic incentives that perpetuate patient harm and inefficient resource ut…

Harnessing cover crop-rock weathering synergies for climate-smart agriculture.

The use of cover crops (CCs) is increasingly promoted to diversify cropping systems and advance agricultural sustainability. Yet, CC adoption can involve context-dependent trade-offs, including resource competition and elevated greenhouse gas (GHG) emissions. In this opinion article, we propose enhanced rock weathering (ERW) as a complementary strategy to improve biogeochemical synchrony within CC systems. By synthesizing emerging evidence, we show how CC-ERW interactions can synergistically enhance carbon sequestration, nutrient cycling, GHG mitigation, and soil food-web functioning, mainly v…
