Risk and protective factors associated with brain grey matter patterns in a population-based cohort of cognitively unimpaired 70 years old.

BACKGROUND: Ageing involves heterogeneous brain grey matter (GM) patterns that may overlap with dementia-related changes. We evaluated cognitively unimpaired older adults to identify specific GM patterns, their clinical and cognitive profiles, and longitudinal trajectories. METHODS: We analysed 746 participants from the Gothenburg H70 study using random forest cross-sectional clustering based on MRI measures of cortical thickness and subcortical volume across 41 regions. Using regression-based models, we examined associations with clinical, MRI variables, biochemical, and CSF Alzheimer biomar

Assessment of visually guided reaching in prodromal Alzheimer's disease: a cross-sectional study protocol.

INTRODUCTION: Recent evidence has implicated the precuneus of the medial parietal lobe as one of the first brain areas to show pathological changes in Alzheimer's disease (AD). Damage to the precuneus through focal brain injury is associated with impaired visually guided reaching, particularly for objects in peripheral vision. This raises the hypothesis that peripheral misreaching may be detectable in patients with prodromal AD. The aim of this study is to assess the frequency and severity of peripheral misreaching in patients with mild cognitive impairment (MCI) and AD. METHODS AND ANALYSIS:

Impaired set shifting is associated with previous falls in individuals with and without Parkinson's disease.

BACKGROUND: Individuals with Parkinson's disease (PD) are at increased risk for falls, which lead to substantial morbidity and mortality. Understanding the motor and non-motor impairments associated with falls in PD is critical to informing prevention strategies. In addition to motor symptoms, individuals with PD exhibit non-motor deficits, including impaired set shifting, an aspect of executive function related to cognitive flexibility that can be measured quickly with the Trailmaking Test. RESEARCH QUESTION: To determine whether impaired set shifting is associated with fall history in peopl

Differential Effect of APOE ɛ4 Status and Elevated Pulse Pressure on Functional Decline in Cognitively Normal Older Adults.

BACKGROUND/OBJECTIVE: The APOE ɛ4 allele and increased vascular risk have both been independently linked to cognitive impairment and dementia. Since few studies have characterized how these risk factors affect everyday functioning, we investigated the relationship between APOE ɛ4 genotype and elevated pulse pressure (PP) on functional change in cognitively normal participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI). METHODS: 738 normally aging participants underwent APOE genotyping, and baseline PP was calculated from blood pressure indices. The Functional Activities Quest

Impact of Cervical Sagittal Alignment Parameters on Neck Disability.

STUDY DESIGN: Retrospective cross-sectional study. OBJECTIVE: Determine if pre-operative cervical alignment serves as an independent predictor of pre-operative disability as measured by the neck disability index (NDI). SUMMARY OF BACKGROUND DATA: There is growing interest in the relationship between cervical sagittal alignment and clinical outcomes. While prior studies have shown that C2-C7 sagittal vertical axis (SVA) correlates with worse NDI scores in post-operative patients, no studies to date have examined the impact of cervical sagittal parameters on pre-operative disability in patient

Assessment and characterization of back pain and biopsychosocial influences in lower limb amputation: scoping review.

Back pain affects over half of individuals with lower limb amputation, yet research findings on associated factors remain fragmented. In view of the evolution of pain science in the general population, this fragmentation likely stems from significant variability in the identification, characterization, and classification of pain, as well as inconsistent assessment of associated biopsychosocial factors. This scoping review aims to examine how back pain is currently identified, assessed, classified, and characterized in people with lower-limb amputation, to determine the gaps and inconsistencies

Subjective and objective cognitive differences in adults in mid-life and older age with autism or high autistic traits: a systematic review and meta-analysis.

BACKGROUND: Research examining ageing in autistic populations, once largely neglected, has expanded in recent years. Cognitive decline and dementia are key public health concerns, and it remains unclear whether autism is related to atypical cognitive ageing. A synthesis of quantitative evidence on cognition in autistic adults in mid- to later-life is needed. This systematic review and meta-analysis is the first to do this. METHODS: Studies comparing objective and subjective cognition in autistic and neurotypical adults aged 50+ were included. Given the underdiagnosis of autism in this age gro

Micro- and Nanoplastics in the Human Brain: Mechanistic Plausibility, Translational Challenges, and Links to Neurological Disease Trends.

The exponential growth in plastic production since the mid-twentieth century has led to the pervasive presence of micro- and nanoplastics (MNPs) across ecosystems and human exposure pathways, coinciding with a rising global burden of neurological disorders. Increasing evidence demonstrates that MNPs are not confined to peripheral tissues but can accumulate even in the human brain, raising concerns about their potential contribution to neurological disease. This structured review synthesizes global trends in plastic production, environmental MNP burden, and human exposure, together with emergin

Do carbon dioxide, volatile organic compounds and atmospheric pressure affect the cognitive performance of occupants in indoor environments? Results of a large-scale experiment with simulated flights.

BACKGROUND: Indoor air quality, especially carbon dioxide (CO2) and volatile organic compounds (VOCs), has been linked to cognitive impairment in previous studies, even below guideline levels. Most previous studies employed small cross-over designs and varied only one indoor air factor. OBJECTIVE: This study aimed to investigate the influence of various combinations of indoor air parameters at typical and elevated levels on cognitive performance. METHODS: We conducted a randomized controlled trial with 398 healthy adults aged 18-69 years. Participants were randomly assigned to 11 groups and
