Bacterial susceptibilities to piperacillin/tazobactam in a tertiary care hospital: 5-year review.

Several factors influence the speed of development of antibacterial resistance, among which is the amount of antibiotic consumption. During the 3-year period 1998-2000, the consumption of piperacillin/tazobactam (pip/tazo) increased by 85% in our hospital. Five years ago we conducted a comparative in vitro study to evaluate susceptibilities of microorganisms to pip/tazo. The objective of the present study was to re-evaluate in vitro susceptibilities to pip/tazo, compared to other beta-lactams, and the potential impact its increased consumption might have on its susceptibility patterns. The stu

Combined use of type A and F botulinum toxins for blepharospasm: a double-blind controlled trial.

Type A botulinum toxin has widened its clinical range of applications, but the risk of developing antibodies limits the repeated use of high-dose injection. To minimize the risk, mixing different types of toxin might reduce the antigenic presentation of a specific toxin and associated proteins. At the same time, inhibition of the neuromuscular release process at the multiple sites might potentiate the clinical response or the duration of action. We compared the effectiveness of a mixture of type A and type F botulinum toxins with that of type A or type F toxin alone for treating patients with

Combination antimicrobial therapy of acute Burkholderia pseudomallei infection in a mouse model.

Burkholderia pseudomallei is the causative agent of melioidosis, a disease endemic in tropical and subtropical regions of South-East Asia and Northern Australia. Antimicrobial therapy regimens for treatment of acute septicemic melioidosis are of variable efficacy. Ceftazidime is the current antibiotic of choice and is commonly administered with other agents such as cotrimoxazole or doxycycline. The emergence of resistant strains of B. pseudomallei and the persistence of high mortality rates prompted the present study. Using an established mouse model of acute disseminated B. pseudomallei infec

A comparison of the effectiveness of three regimens in the prevention of Pneumocystis carinii pneumonia in human immunodeficiency virus-infected patients.

BACKGROUND: Pneumocystis carinii pneumonia (PCP) is a major cause of morbidity and the leading cause of death in patients with the acquired immunodeficiency syndrome. The prevention of the occurrence and recurrence of PCP is a cornerstone in the treatment of patients infected with the human immunodeficiency virus. There are few studies comparing PCP prophylactic regimens. METHODS: The efficacy of three regimens for prophylaxis against PCP was assessed in a retrospective chart review of 211 human immunodeficiency virus-infected patients at risk for the disease. Over the course of the 2-year st

Initiation Patterns and Transitions Among Adults Using Stimulant Drugs: Latent Transition Analysis.

BACKGROUND: The fourth wave of the drug overdose epidemic in the United States includes increasing rates of stimulant-involved overdose. Recent studies of transitions leading to stimulant misuse have shown complex patterns that are not universally applicable because they have isolated individual populations or individual behaviors. A comprehensive analysis of transitions between behaviors and the associations with present-day problematic drug use has not been conducted. OBJECTIVE: This study aims to determine whether adults from the general population who use stimulants initiate use through a

Curcumin impacts on cisplatin sensitivity determining apoptotic boost in mesothelioma.

Cancer is one of the main diseases causing one-third of the worldwide death every year. The recent progresses achieved in chemotherapy have increased patients' survival, however, drug efficacy is limited by the occurrence of serious side effects and the development of drug resistance. To overcome these problems, research interests, aim to develop alternative therapies based on the use of polyphenols, such as curcumin. Polyphenols are effective against different cancers and in combination with standard drugs, display synergism enhancing anticancer effects. Malignant mesothelioma (MM) is an aggr

Sub-additive (antagonistic) interaction of lacosamide with lamotrigine and valproate in the maximal electroshock-induced seizure model in mice: an isobolographic analysis.

BACKGROUND: Launching polytherapy with two or three antiseizure drugs (ASDs) in patients with epilepsy is still problematic. The choice of ASDs to combine them together is usually based on clinicians' experience and it requires knowledge about mechanisms of action of the studied ASDs and their drug-drug interactions, whose nature may be favorable, neutral or unfavorable. To characterize three-drug interaction among lacosamide (LCM), lamotrigine (LTG) and valproate (VPA), the type I isobolographic analysis was used. The antiseizure effects of three-drug combination were analyzed in a model of m

The molecular targets of approved treatments for pulmonary arterial hypertension.

Until recently, three classes of medical therapy were available for the treatment of pulmonary arterial hypertension (PAH)--prostanoids, endothelin receptor antagonists and phosphodiesterase type 5 (PDE5) inhibitors. With the approval of the soluble guanylate cyclase stimulator riociguat, an additional drug class has become available targeting a distinct molecular target in the same pathway as PDE5 inhibitors. Treatment recommendations currently include the use of all four drug classes to treat PAH, but there is a lack of comparative data for these therapies. Therefore, an understanding of the

Thioridazine in PLGA nanoparticles reduces toxicity and improves rifampicin therapy against mycobacterial infection in zebrafish.

Encapsulating antibiotics such as rifampicin in biodegradable nanoparticles provides several advantages compared to free drug administration, including reduced dosing due to localized targeting and sustained release. Consequently, these characteristics reduce systemic drug toxicity. However, new nanoformulations need to be tested in complex biological systems to fully characterize their potential for improved drug therapy. Tuberculosis, caused by infection with the bacterium Mycobacterium tuberculosis, requires lengthy and expensive treatment, and incomplete therapy contributes to an increasin

Evidence for a synergistic interaction between phenytoin and phenobarbital in experimental animals.

Combined anticonvulsant effects of phenytoin and phenobarbital in mice and rabbits were studied using the isobolographic method which was based on the plasma concentrations of these drugs. Anticonvulsant activity of test drugs alone or in combination was determined by the method of maximal electroshock seizure test. Response of each animal to drug action was recorded as abolition of the tonic hindlimb extensor component of maximal electroshock seizure. It was demonstrated that the minimal effective plasma concentrations of phenytoin and phenobarbital alone for protection against maximal electr

["Granuloma pouch" reduction after treatment with indomethacin and diflunisal and collateral effects of these drugs on gastric mucosa].

In the present work it was determined the ability of non-steroidal anti-inflammatory drugs (Diflunisal and Indomethacin) to inhibit formation of "granuloma pouch" exudate induced in rats. This study shows that the combined administration of Diflunisal and Indomethacin exerts similar anti-inflammatory activity as the single drugs. There does not seem to be additive anti-inflammatory effects of the drugs in combination but, if anything, a weak antagonism. The association of the two compounds produces a reduced harmful effect on the gastric mucus.

TREATMENT OF PSORIATIC ARTHRITIS. Psoriatic arthritis is a chronic inflammatory rheumatic disease with a heterogeneous clinical presentation, combining peripheral arthritis and skin psoriasis, and frequently enthesitis and dactylitis. The treatment of peripheral arthritis is based on anti-inflammatory drugs, conventional disease-modifying drugs such as methotrexate, and if the disease remains active, targeted disease-modifying drugs (biotherapies, or targeted synthetic drugs). The management of psoriatic arthritis combines treatment of the joints, treatment of the skin psoriasis, and global me

Insights into How Spinosad Seed Treatment Protects Onion From Onion Maggot (Diptera: Anthomyiidae).

Onion maggot, Delia antiqua (Meigen), is a serious pest of onion Allium cepa L. in northern temperate regions. Over the last decade, D. antiqua has been managed principally using a pesticide seed treatment package containing the reduced-risk insecticide spinosad. While spinosad protects onion seedlings from D. antiqua, very little is known regarding how protection occurs. The main objectives of this study were to assess susceptibility of 1- and 2-wk-old larvae to spinosad through two different modes of exposure: ingestion and contact, and to evaluate larval feeding behavior in choice and no-ch
