Lead-induced fetal nephrotoxicity in iron-deficient rats.

Lead administration (250, 500, 1000, and 2000 ppm, as lead acetate) in drinking water during fetal development (from 15 to 20 days of gestation), in normal and iron-deficient pregnant rats, revealed dose-dependent increases in the lead content of maternal blood that was more marked in iron-deficient animals. The placentae and fetuses did not show a dose-dependent increase in lead content. Lead administration revealed dose-dependent hydropic degeneration of renal proximal cells in the fetuses. The highest dose (2000 ppm lead) and iron deficiency exhibited more lead accumulation in maternal bloo…

Enzyme polymorphism and clinical variability of diseases: a study of diabetes mellitus.

We investigated possible relations among four common neonatal manifestations of diabetic pregnancy (macrosomia, hypoglycemia, hypocalcemia, jaundice) and four enzyme polymorphisms (PGM1, ADA, AK1, ACP1 in a sample of infants born of diabetic mothers. The pattern of associations observed between the two sets of variables is consistent with known differences in enzymatic activity within phenotypes of each system, suggesting that low enzymatic activity may have unfavorable effects on fetal development and on adaptability of the neonate to the extrauterine environment, Some of the polymorphic enzy…

[HIV seropositivity and pregnancy. Apropos of 48 cases (how should they be managed at the present time?)].

We describe the gynaecological and obstetrical management of 48 HIV seropositive, pregnant women and review the literature. We did not find that pregnancy aggravated the development of the condition from the asymptomatic anti-HIV related complex (ARC) and AIDS. It seemed that fetal malformations, especially of the face, occurred most frequently in babies born to HIV seropositive mothers. Materno-fetal transmission of HIV was found in the uterus in 50% of our cases. In conclusion, we recommend safe contraception to prevent pregnancies in anti HIV seropositive women. If that fails, induced first

Fetal hypothalamic transplants: neuronal and neurovascular interrelationships.

This investigation is one of a series aimed at developing an understanding of the neuroanatomical correlates of neuronal and vascular interactions that occur between heterografts of normal fetal hypothalamic tissue transplanted into the third cerebral ventricle of adult Brattleboro rats with homozygous autosomal diabetes insipidus (DI). Rapid vascular invasion of fetal neurografts occurred within 3 days and arose from the mantle plexus of portal capillaries in the underlying host median eminence. Host vessels also invaded from the periventricular stratum and preoptic area. In the ventral regio

[Fetal defects caused by the passive consumption of drugs].

The neuroactive drugs taken by pregnant women have two principals side effects: a teratogenic effects and a postnatal withdrawal effect. Three are the characteristic syndromes: the hydantoin-barbiturate syndrome, the trimethadione-paramethadione syndrome, the fetal alcohol syndrome. The prenatal exposure to heroine and to morphine interfere mainly with the cerebral development. Drug addiction, like postnatale withdrawal syndrome, are mainly due to passive exposure to drug narcotic (heroine, methadone), to sedative-hypnotic agents (barbiturates Diazepam Chlordiazepoxide) and to alcohol. The cig

[Fetal and placental maturation processes towards the end of pregnancy and their diagnosis (author's transl)].

Prenatal fetal maturation is defined as the sum of all growth and differentiation processes necessary for a mature, eutrophic newborn capable of further independent development. These growth and maturation processes, which can be precisely assessed in animal experiments, are subject to genetic and teratogenic influences, but are mainly determined by the duration of pregnancy and uteroplacental function. With the diagnositc procedures available today, such as somography and fetal electrocardiography, the growth and differentiation processes mainly depending on the duration of pregnancy may be a

Structures and development of the venous system in congenital malformations of the brain.

Cerebral venograms of 21 clinical cases of midline anomalies of the brain and of 80 human fetuses between 3 and 8 months old were examined. The diagnostic value of the venogram was fully proven, particularly for the midline cerebral anomalies, where deep cerebral structures and configuration of the tentorium cerebelli were not well visualized by other neuroradiologic maneuvers. Abnormal structures of the deep cerebral veins as well as the dural sinuses in these malformations were proved to retain the venous structures in the early fetal period. The time of formation of these malformations was

IUGR induced by maternal chronic inflammation: long-term effect on offspring's ovaries in rat model-a preliminary report.

PURPOSE: Excess maternal inflammation and oxidative stress while in utero have been known to affect gross fetal development. However, an association between the inflammatory process in utero and the effects on ovarian development and future fertility has not yet been demonstrated. This study focused on LPS-induced chronic inflammation in early pregnancy and its effect on ovarian development and reserves of the offspring, using a rat model. Our aim was to determine whether maternal inflammation in utero disturbs reproductive system development in the offspring, given that maternal inflammation …

Human anterior chamber angle development without cell death or macrophage involvement.

PURPOSE: The iridocorneal angle in the mammalian eye including the trabecular meshwork (TM) develops from undifferentiated mesenchyme/neural crest between the iris root and cornea. The precise mechanisms underlying anterior angle development are unclear, and the contribution of cell death and phagocytic resorption by macrophages in angle development is controversial. In this study, we examined the human anterior chamber angle during various stages of development for evidence of cell death and phagocytic resorption. METHODS: Eyes from the human fetus (F) of 7, 8, 9, 10, 11, 13, 15, 18, 19, 21,

Duplication of mental nerve in a patient with cleft lip-palate and rubella syndrome.

A case of duplication of the mental nerve in a patient with cleft lip, cleft palate, and rubella syndrome is presented. The most vulnerable period of fetal infection by rubella virus corresponds with the critical period of development of the maxilla, mandible, and corresponding orofacial structures. The significance of duplication of the mental nerve is discussed in relation to the influence that this anatomic structure has on the growth and development of the mandible. The concept of activisms to explain such anomalies is reviewed.

Radiological diagnosis of gas gangrene in a fetus at term.

OBJECTIVE: To report a case of gas gangrene in a fetus at term, which was diagnosed by antenatal computed tomography (CT) imaging. CASE REPORT: A 23-year-old primiparous woman, who did not undergo any prenatal health checks until term, developed hypertension, proteinuria, and clouding of consciousness, and experienced intrauterine fetal death. A single, mature fetus with voluminous gas bubbles was observed on CT, which was consistent with a diagnosis of fetal gas gangrene. Following the induction of labor, a dead, malodorous infant was delivered, along with foul-smelling and frothy amniotic f

Diagnosis and management of neonatal alloimmune thrombocytopenia.

Neonatal alloimmune thrombocytopenia (NAT) is a life-threatening bleeding disorder caused by maternal platelet antibodies produced in response to fetal platelet antigens inherited from the father. Antiplatelet antibodies cross the placenta and cause destruction of fetal platelets, leading to severe thrombocytopenia, and potentially bleeding, including fatal intracerebral hemorrhage. Incompatibilities between maternal and fetal platelets for the human platelet antigen 1a (previously called PL(A1)) account for most of the patients with NAT, but other antigens are commonly implicated. Diagnostic

Plasma unconjugated estriol as an indicator of fetal dysmaturity in postterm pregnancy.

The role of twice-weekly plasma unconjugated estriol determinations in the prediction of fetal dysmaturity was studied in 663 patients seen at the Postdates Clinic of the Los Angeles County/University of Southern California Medical Center. The correlation between estriol and gestational age was - 0.055. According to the criteria of the Postdates Clinic protocol, sensitivity, specificity, predictive values of positive results, and those of negative results of estriol determinations in predicting fetal dysmaturity were 44.4, 80.6, 32.9, and 87.1%, respectively. The major deficiency of this test

Toxicokinetic insights into distinct mechanisms of action of two thyroid toxicants: Propylthiouracil and pregnenolone 16α‑carbonitrile.

Thyroid hormones (THs) are critical for metabolic regulation and brain development. Disruptions in TH homeostasis, especially during fetal development, can lead to irreversible neurodevelopmental impairments. Thyroid hormone system-disrupting chemicals (THSDCs), are of growing concern for human health due to their potential to interfere with TH signaling. This study investigates the toxicokinetic properties of two THSDCs: propylthiouracil (PTU), which inhibits TH synthesis, and pregnenolone-16α‑carbonitrile (PCN), which enhances the TH hepatic metabolism. Using in vitro approaches and in vivo …

Placental dysfunction and neonatal mortality induced by Salmonella Abortusequi in a murine model of pregestational infection.

INTRODUCTION: Salmonella Abortusequi is a host-restricted equine pathogen and a major cause of abortion in mares, threatening reproductive health and industry sustainability. This study was aimed to identify factors that adversely affect placental function, contributing to the development of fetal growth restriction (FGR) and neonatal mortality. METHODS: CF1 virgin female mice received a pregestational intragastric dose of Salmonella Abortusequi (108 CFU/mouse). On gestational day 18 (GD18), pregnant females were sacrificed, and maternal tissues and fetoplacental units were collected. In a se

Heterogeneity in the formation of primary and secondary visual fields during human prenatal development.

The human neocortex has a huge surface area with unique cytoarchitectonics, most of which is concealed in sulci. Some cytoarchitectonic fields are associated with macroscopic landmarks. In particular, the primary visual field 17 is associated with the calcarine sulcus. During the prenatal development of the human brain, neocortical gyri and sulci undergo changes and modifications after primary formation. To explore the morphogenetic processes in visual fields during the formation of the primary (provisional) and secondary (permanent) sulci, the occipital lobe of the human fetal brain was studi

Nitric oxide synthase system in the brain development of neonatal hypothyroid rats.

Thyroid hormones play an important morphogenetic role during the fetal and neonatal periods and regulate numerous metabolic processes. In the central nervous system, they control myelination and overall brain development, regional gene expression, and regulation of oxygen consumption. Their deficiency in the fetal and neonatal periods causes severe mental retardation, due to lack of thyroid function, or to iodine deficiency. At the same time, nitric oxide is an atypical neurotransmitter that also has special relevance in neuronal development and plasticity and functions as a vasodilator, regul

Fetal alcohol spectrum disorders and the risk of crime.

Fetal alcohol spectrum disorders (FASD) are an important preventable global health concern. FASD is an umbrella term describing a range of mild to severe cognitive and behavioral problems among individuals prenatally exposed to alcohol. Alcohol causes FASD by interfering with molecular pathways during fetal development involving increased oxidative stress, disturbed organ development, and change of epigenetic gene expression control. Neuroimaging studies into FASD show several neuropathological abnormalities including abnormal brain structure, cortical development, white matter microstructure,…

High-Oxygen Submersion Fetal Thymus Organ Cultures Enable FOXN1-Dependent and -Independent Support of T Lymphopoiesis.

T cell development is effectively supported in fetal thymus organ cultures (FTOCs), which places thymus lobes atop an air-liquid interface (ALI) culture system. The direct exposure to air is critical for its success, as fetal thymus lobes placed in low oxygen submersion (LOS)-FTOCs fail to support thymocyte development. However, submersion cultures performed in the presence of high concentration of ambient oxygen (60~80%) allow for normal thymocyte development, but the underlying mechanism for this rescue has remained elusive. Here, we show that FOXN1 expression in thymic epithelial cells (TEC

Androgens During Infancy, Childhood, and Adolescence: Physiology and Use in Clinical Practice.

We provide an in-depth review of the role of androgens in male maturation and development, from the fetal stage through adolescence into emerging adulthood, and discuss the treatment of disorders of androgen production throughout these time periods. Testosterone, the primary androgen produced by males, has both anabolic and androgenic effects. Androgen exposure induces virilization and anabolic body composition changes during fetal development, influences growth and virilization during infancy, and stimulates development of secondary sexual characteristics, growth acceleration, bone mass accru…
