[Study on the relationship between microsatellite alterations of RASSF1A gene and the development of cervical carcinoma].

OBJECTIVE: To explore the relationship between microsatellite alterations of RASSF1A gene and the development of cervical carcinoma, and HPV16 infection. METHODS: Two sites of microsatellite polymorphism of RASSF1A gene were selected, we used polymerase chain reaction (PCR) technique to detect the LOH and MSI of cervical tissues, and to detect the infection state of HPV16. RESULTS: There were significant differences of LOH rates at the two sites between clinical stage and pathological grade (P < 0.05). Significant differences were noted between the cervical carcinomas with lymph node metasta

Minichromosome maintenance protein expression in benign nevi, dysplastic nevi, melanoma, and cutaneous melanoma metastases.

BACKGROUND: Minichromosome maintenance (MCM) proteins are a recently elucidated group of polypeptides intimately involved in DNA replication and appreciable only in cycling cells. In other organ systems their expression has proven more prognostically useful than cell proliferation markers such as Ki-67 and proliferating cell nuclear antigen. To date, the evaluation of MCM proteins in melanocytic neoplasms has not been undertaken. OBJECTIVE: We sought to determine whether MCM protein 2 (the most extensively evaluated of the MCM protein family) is present in melanocytes from benign nevi, dyspla

Increased expression of podoplanin in malignant astrocytic tumors as a novel molecular marker of malignant progression.

Podoplanin (aggrus) is a mucin-like transmembrane sialoglycoprotein that is expressed on lymphatic endothelial cells. Podoplanin is putatively involved in cancer cell migration, invasion, metastasis, and malignant progression and may be involved in platelet aggregation. Previously, we showed upregulated expression of podoplanin in central nervous system (CNS) germinomas, but not in non-germinomatous germ cell tumors, except for parts of immature teratomas in limited numbers. However, little information exists about its role in CNS astrocytic tumors. In this study, 188 astrocytic tumors (30 dif

Deregulated expression of the PCPH proto-oncogene in human breast cancers.

We performed a study on the expression of the PCPH protein in samples corresponding to normal, pre-malignant and malignant stages of the human mammary gland by using protocols of immunohistochemistry and Western blot analysis with anti-PCPH specific antibodies. Results obtained from the immunohistochemical study showed that PCPH was undetectable in samples of normal breast and of benign diseases, with the exception of glands presenting apocrine metaplasia, in which an intense PCPH stain was observed both in the basal cytoplasm of the secretory cells and in the apocrine secretion. On the contra

Opposite regulation of tenascin-C and tenascin-X in MeLiM swine heritable cutaneous malignant melanoma.

Interactions between tumour cells and surrounding extracellular matrix (ECM) influence the growth of tumour cells and their ability to metastasise. It is thus interesting to compare ECM composition in tumours and healthy tissues. Using the recently described MeLiM miniature pig model of heritable cutaneous malignant melanoma, we studied the expression of two ECM glycoproteins, the tenascin-C (TN-C) and tenascin-X (TN-X), in normal skin and melanoma. Using semiquantitative RT-PCR, we observed a 3.6-fold mean increase of TN-C RNAs in melanoma compared to normal skin. Both stromal and tumour cell

c-myc is not useful as prognostic immunohistochemical marker in cutaneous melanoma.

BACKGROUND: Several studies indicate that immunohistochemical detection of the c-myc oncogene might serve as an additional prognostic marker in malignant melanoma. OBJECTIVE: To study c-myc expression in paraffin-embedded cutaneous melanoma and to correlate to metastatic potential and onset of metastases. METHODS: Cytoplasmic c-myc protein expression was visualized using the APAPP method, and reactivity (percent total tumor cells stained) was assessed in 62 formalin-fixed paraffin-embedded primary cutaneous melanomas (21 not metastasizing, mean Breslow 3.0 +/- 2.9 mm, 41 metastasizing, mean

Expression of the c-kit (CD117) molecule in normal and malignant hematopoiesis.

The c-kit proto-oncogen (CD 117) has been shown to be present in several cell types including normal and neoplastic hemopoietic cells. Among normal BM cells, CD117 expression has been found in about half of the CD34+ precursors including progenitors committed to the erythroid, granulo-monocytic, and megakaryocytic cell lineages. In addition, strong CD117 expression is detected in bone marrow mast cells as well as in a small subset of NK cells displaying strong reactivity for CD56, and in a relatively important proportion of CD3 /CD4 /CD8 prothymocytes. These results suggest that CD117 expressi

Widespread transcription of the growth hormone-releasing peptide receptor gene in neuroendocrine human tumors.

GH-releasing peptides are a new class of potent GH secretagogs (GHS) in vivo and in vitro. In normal man GHS also elicit low but definite ACTH and prolactin secretion. Recently it was shown that patients with pituitary corticotrope adenomas respond to GHS with a dramatic rise in ACTH secretion, and it has been suggested that GHS may provide a diagnostic tool to differentiate Cushing's disease from the ectopic ACTH syndrome. GHS actions are mediated by a G protein-coupled receptor recently characterized and cloned in man and rat. In this study we analyzed GHS receptor (GHS-R) expression in vari

Hepatocarcinogenic effect of vinyl carbamate in the C57Bl/10J strain mouse.

The genotoxic carcinogen vinyl carbamate was dosed to C57Bl/10J strain mice for 35 weeks, and the study terminated after week 59. A main study group of 55 males and 50 females was dosed 6 mg/kg vinyl carbamate once weekly by intraperitoneal injection, whilst a reference group of 10 animals/sex were kept undosed. From week 39 onwards there was a high incidence of mortality, which was often associated with acute internal abdominal hemorrhage. Mice of both sexes killed from 34 weeks onwards frequently showed macroscopic evidence of blood-filled, cyst-like structures in the liver. Upon histopathol

Autocrine activity of interleukin 6 secreted by hepatocarcinoma cell lines.

Among several rat hepatoma cell lines known to secrete interleukin 6 (IL6), the HTC.JZ1 line stands out as a high-level producer. HTC.JZ1 cells were stimulated to secrete up to fourfold increased amounts of IL6 over 24 hours by treatment with lipopolysaccharides (LPS). Both functional IL6 levels, measured as hepatocyte stimulating factor (HSF) activity, and IL6 mRNA concentrations were increased proportionally by exposure to LPS. Similarly, IL6 mRNA was induced by LPS treatment in cultured primary rat hepatocytes. The induction of Il6 mRNA by LPS was inhibited both in primary hepatocyte and he

MiR-130a Acts as a Tumor Suppressor MicroRNA in Cutaneous Squamous Cell Carcinoma and Regulates the Activity of the BMP/SMAD Pathway by Suppressing ACVR1.

Cutaneous squamous cell carcinoma (cSCC) is a malignant neoplasm of the skin resulting from the accumulation of somatic mutations due to solar radiation. cSCC is one of the fastest increasing malignancies, and it represents a particular problem among immunosuppressed individuals. MicroRNAs are short noncoding RNAs that regulate the expression of protein-coding genes at the post-transcriptional level. In this study, we identify miR-130a to be downregulated in cSCC compared to healthy skin and precancerous lesions (actinic keratosis). Moreoever, we show that its expression is regulated at the tr

A palmitoylation-associated three-gene signature predicts prognosis, immune features, and drug sensitivity in gastric cancer: functional validation of CTHRC1.

BACKGROUND: Gastric cancer (GC) is a heterogeneous malignancy that requires improved molecular tools for prognostic stratification. Protein palmitoylation regulates membrane localization and signaling, but the clinical relevance of palmitoylation-related genes (PRGs) in GC remains unclear. METHODS: Gene Expression Omnibus, The Cancer Genome Atlas, and GeneCards data were integrated to identify PRGs. Differential expression analysis, weighted gene co-expression network analysis, and three machine-learning algorithms were used to select key genes. Diagnostic and prognostic performance was evalu

DNA Hypomethylation of MIR21 Drives Hsa-miR-21-5p Expression in High-Grade Meningiomas and Reshapes Transcriptomic Signatures of Oncogenic Pathways and Intercellular Communication.

Meningiomas are the most common intracranial tumors. DNA methylation analysis in benign and aggressive meningiomas showed decreased MIR21 methylation and overexpression of hsa-miR-21-5p in atypical and anaplastic tumors. Transcriptomic analysis of distinct WHO grade meningiomas showed multiple predicted hsa-miR-21-5p target genes as differentially expressed. They were mainly related to processes of intercellular and intracellular signaling. Intercellular communication in meningioma was investigated using the deposited scRNA-seq dataset and deconvolution of our RNA-seq data. We found WHO grade-

Integrative transcriptomic and experimental analyses identifies SDF4 as an astrocyte-expressed regulator of glioma progression through PI3K-AKT signaling.

BACKGROUND: Glioma is an aggressive brain tumor with poor prognosis and high recurrence, which significantly affects patients' quality of life. Identifying new biomarkers and therapeutic targets is crucial for improving diagnosis and treatment. This study focuses on the SDF4 gene, which has been implicated in various cancers; however, its role in glioma remains underexplored due to limited research. METHODS: To comprehensively investigate the role of SDF4 in glioma, we integrated bulk, single-cell, and spatial transcriptomic analyses with experimental validation. This approach includes bioinf

Lactylation-mediated remodelling of the breast cancer microenvironment: single-cell multidimensional analysis and prognostic model construction.

BACKGROUND: The breast cancer tumour microenvironment (TME) exhibits marked cellular and metabolic heterogeneity that contributes to disease progression and therapeutic resistance. Lactylation, a lactate-derived post-translational modification, has emerged as a potential link between metabolic reprogramming and tumour-associated transcriptional and immune changes. However, its cell-type distribution and clinical relevance in breast cancer remain incompletely defined. METHODS: We integrated 26 scRNA-seq samples spanning ER+, HER2+, and triple-negative breast cancer (TNBC), comprising 98,572 ce

Single-cell transcriptomics reveals FXR1 as an actionable target for siRNA therapy in ovarian cancer.

Ovarian cancer is one of the leading causes of cancer-related mortality among women and remains exceptionally difficult to manage and treat effectively in the clinic. Fragile X-related protein 1 (FXR1) is highly amplified and overexpressed in ovarian and several other cancers. FXR1 is a key regulator of the translation of multiple oncogenes and therefore represents a vulnerable target for cancer therapy. RNA interference (RNAi) of FXR1 using a locked nucleic acid (LNA) form of siRNA (siFXR1-LNA) inhibits tumor growth, ascites formation, and metastasis of ovarian cancer more efficiently than th

Metabolic-immune crosstalk in head and neck squamous cell carcinoma: CD44 and APP identified as causal therapeutic targets via integrated lactylation-Mendelian randomization analysis.

BACKGROUND: Patients with head and neck squamous cell carcinoma (HNSCC) continue to face poor prognosis, highlighting an urgent need for new diagnostic markers and therapeutic targets. While metabolic reprogramming and immune microenvironment dysregulation are crucial drivers of HNSCC progression, the key causal molecular mechanisms linking these processes remain elusive. Post-translational modifications, especially protein lactylation, may serve as a vital interface for this metabolic-immune "crosstalk". METHODS: We developed an integrative analytical framework merging lactylation proteomics

E2F1-mediated transcriptional regulation of TMEM33 promotes colorectal cancer proliferation, metastasis, and EMT.

Colorectal cancer (CRC) remains one of the most lethal malignancies worldwide, with tumor metastasis representing the primary driver of treatment failure and poor prognosis. Transmembrane Protein 33 (TMEM33) has been implicated in tumor progression across several cancer types, yet its expression pattern, biological function, and upstream transcriptional regulation in CRC remain undefined. In this study, TMEM33 expression in CRC and matched adjacent normal tissues was assessed by quantitative reverse transcription polymerase chain reaction (qRT-PCR), Western blot, and immunohistochemistry (IHC)
