The pneumococcus obtains its energy from the metabolism of host glycosides. Therefore, efficient degradation of host glycoproteins is integral to pneumococcal virulence. In search of novel pneumococcal glycosidases, we characterized the Streptococcus pneumoniae strain D39 protein encoded by SPD_0065 and found that this gene encodes a beta-galactosidase. The SPD_0065 recombinant protein released galactose from desialylated fetuin, which was used here as a model of glycoproteins found in vivo. A pneumococcal mutant with a mutation in SPD_0065 showed diminished beta-galactosidase activity, exhibi

The cholinergic nervous system can inhibit the systemic inflammation accompanying sepsis by virtue of a specific action of acetylcholine on alpha7 cholinergic receptors. The current authors sought to determine the effect of nicotine, an alpha7 cholinergic receptor agonist, on the host response to pneumonia caused by Streptococcus pneumoniae. Mice were intranasally infected with S. pneumoniae and treated with nicotine or saline intraperitoneally using a treatment schedule shown to improve host defence against abdominal sepsis. Nicotine treatment was associated with a transiently enhanced growth

The immune response induced by Toxoplasma gondii is characterized by Th1 immune mechanisms. We previously demonstrated that C57BL/6 mice infested with Myocoptes musculinus and infected with T. gondii by intraperitoneal route undergo accelerated mortality according to Th2 immune mechanisms induced by the acarian. To evaluate whether infection with M. musculinus influences T. gondii-induced Th1 response in a resistant mouse lineage, BALB/c, which develops latent chronic toxoplasmosis in a way similar to that observed in immunocompetent humans, this study was done. The animals were infected with

Francisella tularensis is a pathogenic bacterium whose virulence is linked to its ability to replicate within the host cell cytosol. Entry into the macrophage cytosol activates a host-protective multimolecular complex called the inflammasome to release the proinflammatory cytokines interleukin (IL)-1beta and -18 and trigger caspase-1-dependent cell death. In this study, we show that cytosolic F. tularensis subspecies novicida (F. novicida) induces a type I interferon (IFN) response that is essential for caspase-1 activation, inflammasome-mediated cell death, and release of IL-1beta and -18. Balamuthia mandrillaris, a soil amoeba, is the causative agent of Balamuthia granulomatous amoebic encephalitis, a life-threatening brain infection. This amoeba is acquired from contaminated soil and may enter the host through cutaneous lesions or through nasal passages, migrating to the lungs or brain. During invasion, B. mandrillaris has access to components of the extracellular matrix (ECM) of the host. Therefore, we investigated the interaction of B. mandrillaris with 3 ECM glycoproteins (collagen-I, fibronectin and laminin-1) that are encountered in host connective tissues and at the basa

The development of resistance to sulfadoxine-pyrimethamine by Plasmodium parasites is a major problem for the effective treatment of malaria, especially P. falciparum malaria. Although the molecular basis for parasite resistance is known, the factors promoting the development and transmission of these resistant parasites are less clear. This paper reports the results of a quantitative comparison of factors previously hypothesized as important for the development of drug resistance, drug dosage, time of treatment, and drug elimination half-life, with an in-host dynamics model of P. falciparum m

In the host, the antigen excreted by schistosomes in the circulating blood is concentrated in the urine. A mouse monoclonal antibody of the IgM class type lambda, directed against an epitope of the intestinal epithelium of the adult worm, is obtained. The antigen found in the urine of the host as well as the monoclonal antibody has been previously characterized. It is of a polysaccharidic nature, is thermostable and specific for the genus Schistosoma. The antigen is found at all stages of the life cycle and, particularly, in the egg where it is found in large amounts. Detection of the antigen

In recent years, numerous studies have revealed that dysregulation of mitochondria-organelle interactions is a common feature underlying various pathological processes and pathogen infections. For instance, in Alzheimer's disease (AD), dysfunction of mitochondrial-associated ER membranes (MAMs) leads to calcium overload and oxidative stress, while cancer cells enhance glycolysis by remodeling mitochondria-Golgi interactions. Targeting these key interacting nodes has shown significant therapeutic potential. Although technological advances have uncovered some underlying mechanisms, the spatiotem

Nsp15 is an endoribonuclease, highly conserved among coronaviruses, that helps the virus evade detection in host cells by cleaving uridine-rich viral RNA sequences. Those sequences would otherwise trigger immune response pathways. Its essential role in pathogenesis and highly conserved nature make Nsp15 an attractive target for therapeutic intervention. While its crystal structure and uridine specificity are well-established, the influence of RNA structural dynamics and divalent cations on Nsp15 activity remains less understood. Leveraging single-molecule Pulsed Interleaved Excitation (PIE)-FR

The growing threat of viral pandemics, along with the emergence and re-emergence of various viruses, highlights an urgent need for innovative antiviral strategies. Traditional antivirals typically target viral proteins, but this approach often faces challenges like drug resistance and specificity. In contrast, host-based antivirals present a promising alternative by focusing on cellular factors essential for viral replication, which can potentially limit viral adaptation and resistance. Kinase inhibitors are a notable class within these therapies, as viruses frequently exploit kinases to aid i

Recurrent urinary tract infections (rUTIs), driven by multidrug-resistant (MDR) uropathogens, intracellular reservoirs, and biofilms, progressively circumvent conventional antibiotic therapy. Bacteriophage (phage) therapy has re-emerged as a precision medicine strategy offering narrow host specificity, self-amplification at the infection site, and intrinsic antibiofilm activity via depolymerases. This minireview evaluates the biological rationale and translational evidence for phage therapy in MDR-rUTIs, emphasizing the clinical distinction between standardized trials for acute uncomplicated i…

Extracellular vesicles (EVs) produced by probiotic bacteria are increasingly recognized as crucial mediators of host-microbe communication. However, the molecular composition and biological interpretation of bacterial EV proteomes are heavily influenced by the isolation methods. In this study, we systematically compared ultracentrifugation (UC) and size exclusion chromatography (SEC) for isolating EVs from Lactobacillus reuteri, assessing their impact on EV yield, purity, and proteomic profiles. Although UC yielded significantly more EVs than SEC, it also resulted in lower purity, as evidenced

INTRODUCTION: The zoonotic bacterium Yersinia pseudotuberculosis (YPS) primarily causes gastrointestinal and extraintestinal diseases in animals, but can also infect humans. Due to the concerted action of a broad spectrum of virulence determinants associated with an infection by YPS, the host specificity of certain phylotypes and determinants affecting the epidemiology of the disease are still poorly understood. In 2025, a case of mastitis of a cow in Germany was attributed to YPS, with the bacterium being isolated at various time points.
METHODS: The isolates were analysed in-depth using cla…

Viral infectious diseases pose a persistent challenge to global livestock production, animal welfare, and food security, emphasizing the critical role of early host defense mechanisms in limiting viral replication and transmission. As frontline sensors of infection, pattern recognition receptors (PRRs) detect viral nucleic acids and trigger antiviral innate immune responses via coordinated downstream signaling. This review summarizes the functions of three major PRR families, Toll-like receptors (TLRs), RIG-I-like receptors (RLRs), and the cyclic GMP-AMP synthase-stimulator of interferon genes

Regulatory T cells (Tregs) are key mediators of immune tolerance and play a critical role in limiting excessive immune activation in conditions such as autoimmunity, transplantation, and graft-versus-host disease. Tregs are broadly classified into thymic-derived Tregs (tTregs) and peripherally induced Tregs (pTregs), which differ in lineage stability, epigenetic regulation, and functional plasticity. The suppressive function of tTregs is supported by stable expression of the transcription factor FOXP3, reinforced by demethylation of the Treg-specific demethylated region (TSDR). In contrast, pT

Unambiguous identification of HIV reservoirs is essential for their characterization and for developing curative strategies. A major obstacle to curing HIV is the persistence of silent viral genomes in host cells that evade immune detection and resist therapy. Here, we present a fluorescence microscopy-based method for direct in vivo visualization of full-length double-stranded HIV-1 DNA, an achievement not previously realized. To achieve this, we developed an advanced imaging platform adapted for replication-competent HIV-1. This system leverages a bacterial-derived two-component tagging stra

Histopathologic melanoma diagnosis extends beyond melanocytic cytology to encompass non-melanocytic features: solar elastosis patterns, stromal regression, adnexal relationships, epidermal reaction patterns, and the host inflammatory response. These "old school" low-power clues are particularly valuable on sun-damaged skin, where benign nevi, reactive melanocytic hyperplasia, and melanoma in situ share overlapping features. Quantitative data support two elastosis-based signs: the "umbrella sign" (reduced elastosis beneath the lesion's central third; PPV (Positive Predictive Value) for nevus, 9
