Lineage and organ signals sequentially build organ intrinsic nervous systems.

Organ intrinsic nervous systems (OINSs) are critical components of the body-brain axis and coordinate visceral organ function with systemic physiological control1-7. Despite their importance, how these distinct neural architectures arise from a common neural crest cell origin has remained unclear. Here we present a systems-level, cross-organ analysis of OINS development, integrating lineage tracing, 3D imaging, single-cell transcriptomics and genetic perturbations across the heart, pancreas, intestine and lungs. We show that differences in neural crest cell migratory trajectories prefigure the

Repair of Corneal Epithelial Defects.

The corneal epithelium, a stratified squamous non-keratinized layer of 50-60 μm thickness, forms the outermost barrier of the cornea and provides both optical clarity and protection against trauma, infection, and fluid imbalance. It plays a vital role in protecting the eye and maintaining visual clarity. A range of conditions, including trauma, metabolic disorders, microbial infection, and limbal stem cell deficiency, can lead to chronic corneal epithelial defects and subsequent visual impairment. Epithelial renewal is a continuous process, primarily sustained by stem cells located at the limb

Nephronectin Promotes Migration of the Periocular Mesenchyme via EGFR Signaling.

PURPOSE: Nephronectin (Npnt) is an extracellular protein with various functions during development and homeostasis of various tissues, but its role during early mouse cornea development remains unexplored. We characterized the expression of Npnt and investigated its function during cornea development. METHODS: We performed in situ hybridization and immunohistochemistry on ocular sections obtained from embryonic day (E)11.5-E16.5 mouse embryos. Wild-type and Npnt mutant embryos were analyzed for corneal thickness, cell number, and cell proliferation. We assessed the expression of Npnt receptor

Changes in the gut intra-epithelial lymphocyte landscape induced by erythropoietin treatment in a non-diseased mouse model.

Erythropoietin (EPO) is a glycoprotein hormone primarily known for its central role in red blood cell production, but it is now also recognized for its immunomodulatory effects on a range of immune effectors. Several studies indicate that EPO helps regulate intestinal barrier integrity, although its effect on intraepithelial lymphocytes (IELs) is still unknown. Using multiparameter flow cytometry and both conventional and unsupervised analyses, we conducted an in-depth phenotypic assessment of IELs from the murine duodenum, jejunum, ileum and colon. Our results revealed that in EPO-treated C57

Exploring the impact of syndecans in prostate cancer: Stage-specific roles and therapeutic implications.

Syndecans (SDCs) 1-4 are a family of transmembrane heparan sulfate proteoglycans (HSPGs) that regulate cell-cell communication, adhesion, extracellular matrix organization, and signaling pathways involved in tumor biology. In prostate cancer (PCa), accumulating evidence suggests that SDCs contribute to tumor progression, therapeutic resistance, and interactions within the tumor microenvironment. However, their specific, stage-dependent roles remain incompletely understood. This review provides an integrated synthesis of current experimental and clinical evidence on SDC1-SDC4 in PCa, complement

Chaperone-mediated autophagy supports organ regeneration and fibroblast quiescence in mouse models of fibrosis.

The core of organ fibrosis formation lies in the regenerative defects of parenchymal cells and in the excessive activation of fibroblasts, yet methods to simultaneously address these pathological cell types remain lacking. Here, we found that the expression of the chaperone-mediated autophagy (CMA) limiting factor lysosome-associated membrane protein type 2A (LAMP2A) was consistently down-regulated in mouse models of bleomycin-induced pulmonary fibrosis, carbon tetrachloride (CCl4)-induced liver fibrosis, and folic acid-induced renal fibrosis. We also confirmed a low CMA score in patients with

Integrative Proteomic Analysis Implicates Inhibition of Intracellular Protein Trafficking in Therapy-Induced Migrastasis in Prostate Cancer.

Background: Dysregulated cell migration leading to metastasis remains the primary cause of cancer-related mortality. It has been challenging to understand how cells regulate migration. We have previously created the first selective inhibitor of cell migration, KBU2046. Here, we use it as a probe to identify regulatory processes. Methods: Metastatic and primary human prostate cancer cells were treated for different times and at different concentrations with KBU2046. Immunofluorescent microscopy examined protein localization in cells. Label-free mass spectrometry (MS) was performed on total cell

Inhibiting thromboinflammation via interleukin-4 binding to platelet glycoprotein VI and suppression of thrombosis.

Inflammation and coagulation are intricately linked. Although some studies have documented proinflammatory drivers of coagulation, the role of anti-inflammatory cytokines and the underlying mechanisms in the modulation of coagulation, platelet function, and thrombosis remains poorly characterized. In this study, we demonstrate that interleukin-4 (IL-4), an anti-inflammatory cytokine, plays a crucial role in regulating platelet function and thrombus formation. IL-4 inhibited collagen-induced calcium mobilization, P-selectin expression, ATP release, integrin αIIbβ3 activation, platelet spreading

Murine Models of Hemostasis: How to Assess Bleeding in Mice and Clinical Relevance of These Models for Testing New Therapeutics.

CLINICAL PROBLEM: Congenital bleeding disorders stem from genetic defects affecting procoagulant proteins (such as von Willebrand Factor, factor VIII, or factor IX) or platelet proteins (such as integrin αIIbβ3 or glycoprotein Ibα). Despite advances in our knowledge of the hemostatic system, there are still gaps in our understanding of the interplay between the various hemostatic actors, limiting the development of efficient therapeutic agents for each of these disorders. RECOMMENDATIONS: Mouse models have a proven record in their use as preclinical tools for the development and testing of th

Rescue of stereocilia architecture and hearing function by AAV-CIB2 and AAV-CIB3 in a mouse model of recessive deafness DFNB48.

Pathogenic variants in the calcium and integrin-binding protein 2 (CIB2) are among the leading causes of nonsyndromic prelingual hearing loss (DFNB48). Loss of Cib2 in mice causes severe stereocilia bundle defects, impaired mechanoelectrical transduction (MET), and profound hearing loss. To determine whether a critical window of opportunity exists to restore the stereocilia architecture and to rescue hearing function, we developed and investigated the effect of adeno-associated virus (AAV)-mediated gene delivery in Cib2 mutant mice. A single administration of the AAV-Cib2 vector was performed

Single-cell analysis identifies ATC-like cells driving progression in relapsed follicular thyroid carcinoma.

Follicular thyroid carcinoma (FTC) is prone to early distant metastasis and has a poor prognosis compared with papillary thyroid carcinoma (PTC). This study aimed to unravel the cellular and molecular mechanisms underlying FTC progression and its transformation into the aggressive anaplastic thyroid carcinoma (ATC). Through single-cell RNA sequencing (scRNA-seq) profiling of 46 739 cells from PTC, follicular variant PTC (FVPTC), relapsed FTC (RFTC), and ATC, we reconstructed a comprehensive molecular trajectory of thyroid carcinoma progression. Our analysis revealed that PTC, FVPTC, and FTC po

Carrier-free targeted co-assembly of glycyrrhizin and puerarin overcomes ocular barriers for corneal alkali burn therapy via macrophage reprogramming.

Corneal alkali burns (CAB) represent a devastating form of ocular trauma characterized by the rapid penetration of alkaline agents, triggering a destructive cascade of inflammatory infiltration, neovascularization, and fibrotic remodeling that compromises ocular transparency. Current pharmacological interventions are largely palliative and severely hampered by rapid precorneal clearance and poor epithelial penetrability. To overcome these formidable dynamic and static ocular barriers, we engineered a carrier-free, targeted nanoplatform via the spontaneous co-assembly of natural bioactives glyc

Recombinant humanized type III collagen improves ovarian function via ITGA2-mediated mitochondrial function restoration in granulosa cells and extracellular matrix remodeling.

Ovarian aging is characterized by depletion of the follicular reserve. The extracellular matrix (ECM) provides the essential microenvironmental niche for follicles, and aged-related alterations in ECM composition may adversely affect follicular dynamics. However, the precise relationship between matrix components and ovarian aging remains unclear. In this study, we first observed a marked reduction in type III collagen (Col III) as a key alteration associated with ovarian aging. Furthermore, we discovered that as a biomaterial, recombinant humanized type III collagen (rhCol III) ameliorated ov

Brain targeting and trafficking of extracellular vesicles in central nervous system diseases: a therapeutic roadmap.

Extracellular vesicles (EVs) mediate intercellular signaling in the central nervous system (CNS) by transferring lipids, proteins, and nucleic acids among neurons, glia, endothelium, and immune cells. Brain targeting depends on a linked sequence: EV ligands and adsorbed protein coronas engage receptor modules, select endocytic routes, determine intracellular fate, and define the therapeutic readouts. These fates include lysosomal degradation, recycling, rare cytosolic delivery, or transport across the blood-brain barrier (BBB). In disease, the same pathways can disseminate proteopathic seeds a

Collagen-heparan sulfate microcarriers support mesenchymal stem cell culture in microgravity and streamline post-flight recovery.

Mesenchymal stem cells (MSCs) hold broad therapeutic potential, yet the absence of validated, scalable adherent culture methods remains a barrier to orbital biomanufacturing. Here, we report the first integrated workflow for MSC culture on pre-seeded microcarriers (µCs) encompassing cryopreservation, ambient thaw, and expansion aboard the International Space Station. We engineered heparan sulfate-collagen (HS/COL) µCs with defined sulfation levels to modulate integrin engagement and factor-release kinetics and benchmarked them against commercial µCs and suspension controls. Our results demonst

Cancer-associated fibroblast-derived POSTN as a metabolic "switch" shapes lipid-stressed macrophage polarization in gastric cancer.

BACKGROUND: Cancer-associated fibroblasts (CAFs) foster an immunosuppressive tumor microenvironment (TME) and confer resistance to immune checkpoint blockade (ICB) in gastric cancer (GC). However, the mechanisms by which specific CAF subsets regulate metabolic crosstalk and immune evasion remain poorly defined. METHODS: We integrated bulk and single-cell RNA-sequencing data, spatial transcriptomics, and clinical cohorts of GC patients treated with immunotherapy. Functional validation was performed using in vitro co-culture systems, multiple murine models, and lipid nanoparticle (LNP)-encapsul

Liver-inspired decellularized hydrogel platform to eradicate osteosarcoma and reconstruct bone defects.

Osteosarcoma (OS), the most prevalent primary malignant bone tumor, presents significant clinical challenges following surgical resection, including irregularly-shaped bone defects, residual microtumors, and inadequate bone regeneration. Inspired by the liver's remarkable regenerative capacity, an injectable hydrogel was fabricated from liver-decellularized extracellular matrix (dECM) by incorporating liposomes co-loaded with gold nanoparticles (GNPs) and doxorubicin (DOX), which was designated as GL/DOX@dECM. Functionally, this hydrogel exhibited prominent thermosensitivity, enabling facile i

Development of Experimental Platforms to Assess Helicobacter pylori HopQ Interaction with Host CEACAM Molecules.

BACKGROUND: Helicobacter pylori is an extracellular bacterium responsible for various gastrointestinal diseases, such as peptic ulcers and gastric cancer. It uses multiple mechanisms to colonize the harsh, acidic environment of the stomach and establish its pathogenic processes, mostly through CagA translocation. While cell surface integrin molecules were previously believed to be the main mediators anchoring H. pylori and facilitating this process, recent studies highlight the critical role of the interaction between the bacterial adhesin Helicobacter pylori outer membrane protein Q (HopQ) an

Polymer-engineered liposome-delivered Ebselen against tumor through GSH/H2O2-responsive disruption of redox homeostasis and direct p53 activation.

Ebselen (EbSe), an organic selenium-based compound with potential antitumor activity, faces great challenges of poor aqueous solubility, which impairs its bioavailability and exacerbates potential toxicity. Liposomes, as established clinical tools for delivering small molecule drugs, could help improve the therapeutic effectiveness by overcoming their aqueous solubility limitations to reduce side effects and increase accumulation in tumor tissue. In this study, we engineered a novel nano-delivery system (EbSe@LNP) using clinically validated cationic liposomes, ionizable lipids, and amphiphilic
