THE MORPHOLOGY OF THE PARASITE (HISTOPLASMA CAPSULATUM) AND THE LESIONS OF HISTOPLASMOSIS, A FATAL DISEASE OF TROPICAL AMERICA.

Histoplasmosis is a fatal infectious disease of tropical America resembling kala-azar of India. It is characterized clinically by splenomegaly, emaciation, irregular pyrexia, leucopenia and anemia. The pathological features are the invasion of endothelial cells in the smaller lymph and blood vessels and capillaries by enormous numbers of a small encapsulated microorganism (Histoplasma capsulatum) causing necroses of the liver with cirrhosis, splenomegaly, psudo-granulomata of the lungs, small and large intestines, with ulceration of the latter, and necrosis of lymph nodes draining the injected

Impacts of artificial light at night on interactions between four trematode parasites and their marine invertebrate hosts: species-specific effects?

Parasitism is influenced by anthropogenic stressors, among which artificial light at night (ALAN) remains understudied. With the expansion of urban areas and the development of lighting technology, the range and intensity of ALAN's effects on host-parasite interactions are increasing. Trematodes, with their complex life cycles, are particularly sensitive to anthropogenic stressors, especially during the transmission of motile cercariae (infective stages) from the first to the second intermediate host while exposed to external conditions. We examined how three types of ALAN (warm white LED, col

Mutations in apicoplast rRNA genes are associated with clindamycin resistance and impair the ability of malaria parasites to infect mosquitoes.

Drug resistance hampers malaria treatment and control. Resistance to nearly all clinically used antimalarials has emerged and spread globally. With multi-drug-resistant parasites now on the rise, understanding resistance mechanisms and their ability to spread is crucial for optimal treatment and control strategies. Clindamycin is an apicoplast-targeting antimalarial used as a partner compound in second-line treatment combinations, but mechanisms of clindamycin resistance remain largely unexplored, and it is unclear whether resistant parasites could spread readily. We selected in vitro for clin…

Gastrointestinal Parasites of Wild Birds in Mexico: A Synthetic Review of the Current Knowledge and Approaches for Bird Parasitological Studies.

Parasites are part of ecosystems and can regulate populations. The diseases they cause may negatively impact wildlife, sometimes linked to the imbalance created by anthropogenic environmental changes. Mexico harbors more than 10% of the world's avifauna, but there have been few studies on gastrointestinal (GIT) parasites in wild birds of Mexico. The aim of this review was to provide a comprehensive summary of knowledge on GIT parasites of Mexican wild birds and offer suggestions for approaches for future parasitological research on birds in Mexico. We analyzed 85 documents reporting gastrointe…

Zinc pyrithione inhibits blood-stage parasites of Plasmodium falciparum and its combinatorial effect with dihydro-artemisinin and chloroquine in culture.

The malarial parasite Plasmodium falciparum has evolved resistance to several antimalarial drugs, posing a significant challenge to the effective management and treatment of malaria in endemic areas. Drug repurposing has emerged as a potential alternative strategy for addressing this issue. This study aimed to identify an FDA-approved microbicidal agent, zinc pyrithione (ZPT), against mixed blood-stage parasites of P. falciparum chloroquine-sensitive (Pf3D7) and resistant strains (PfINDO). Based on the time-inhibition kinetics assay, the parasite viability was significantly inhibited by ZPT tr…

Co-infection of intestinal protozoa and intestinal parasites with Helicobacter pylori among HIV AIDS patients at Jimma Medical Center, Ethiopia.

BACKGROUND: Human Immunodeficiency Viruses (HIV) infected people are at increased chance of immunocompromission. In combination with this immunocompromission, the incidence of co-infection with intestinal parasites, intestinal protozoal and Helicobacter pylori can be escalated by different factors including similar mode of transmission, poor economic status, synergistic pathogenesis, and geographic topography of more than two organisms. A large number of world population is either affected or at risk of infection from diversified intestinal parasites, intestinal protozoal and H. pylori. OBJEC…

Identification of genomic regions associated with resistance to gastrointestinal parasites in an indigenous sheep by single- and multiple-locus methods.

We investigated the association between 157 SNPs located in 75 candidate genes involved in the immune system and proxy traits for resistance to gastrointestinal nematodes in sheep. A total of 211 lambs from eight flocks were sampled. Nematode eggs per gram were counted and classified as: (i) Strongyles, (ii) Nematodirus spp., (iii) Trichuris spp. and (iv) Marshallagia marshalli. Single- and multiple-locus models were used to test the marker-trait associations. Seven significant SNPs were identified on chromosomes OAR6, 15, 16, and 19. These findings provide insights for breeding nemarode-resis

Assessing morphological preservation of gastrointestinal parasites from fecal samples of wild capuchin monkeys (Cebus imitator) stored in ethanol versus formalin.

The copromicroscopic identification of gastrointestinal parasites is a common, cost-effective method vital to understanding host-parasite interactions. However, its efficacy depends on effective preservation of the samples. In this study, we compare the preservation of ethanol and formalin preserved gastrointestinal parasites collected from a wild population of Costa Rican capuchin monkeys (Cebus imitator). Fecal samples were collected, halved, and stored in either 10% formalin or 96% ethanol at ambient temperature, then microscopically screened for the presence of parasites. Parasites were mo…

Parallel detection of multiple zoonotic parasites using a real-time fluorogenic loop-mediated isothermal amplification-based quadruple-sample microfluidic chip.

Zoonotic parasites pose significant health risks globally. In the present study, we combined a microfluidic chip with loop-mediated isothermal amplification (on-chip LAMP) to detect five zoonotic parasites: Toxoplasma gondii, Cryptosporidium parvum, Cryptosporidium hominis, Clonorchis sinensis, and Taenia solium. This method enabled the simultaneous parallel analysis of five genetic markers from a maximum of four samples per chip. The on-chip LAMP assay was conducted in a highly automated format via the addition (by pipetting) of each sample in a single operation. The reaction was performed in…

Strasseriolides display in vitro and in vivo activity against trypanosomal parasites and cause morphological and size defects in Trypanosoma cruzi.

Neglected diseases caused by kinetoplastid parasites are a health burden in tropical and subtropical countries. The need to create safe and effective medicines to improve treatment remains a priority. Microbial natural products are a source of chemical diversity that provides a valuable approach for identifying new drug candidates. We recently reported the discovery and bioassay-guided isolation of a novel family of macrolides with antiplasmodial activity. The novel family of four potent antimalarial macrolides, strasseriolides A-D, was isolated from cultures of Strasseria geniculata CF-247251…
