CORONATE: A Novel Questionnaire to Assess the Long-Term Impact of Psoriasis on Patients' Lives.

INTRODUCTION: Psoriasis has a profound impact on patients' quality of life, affecting their emotional well-being, social interactions, and career choices. However, current assessment tools fail to capture the long-term burden of psoriasis. OBJECTIVE: The aim of this study was to develop the CORONATE questionnaire, designed to assess the lifelong impact of moderate-to-severe psoriasis on patients' lives. METHODS: A multicenter cross-sectional study was conducted in 15 Italian dermatology centers. This study included 300 patients (age ≥30 years) diagnosed with moderate-to-severe psoriasis (PAS…

Efficacy and Safety of Adalimumab as Monotherapy Versus Adalimumab With Methotrexate for Psoriasis: A Systematic Review.

Adalimumab is a common biologic used for the management of moderate-to-severe plaque psoriasis. Despite the known importance of adding methotrexate to adalimumab in reducing immunogenicity and improving clinical response, the extent of its benefit remains unclear. This review aimed to systematically compare adalimumab with and without methotrexate, considering their efficacy and safety in the management of psoriasis. This systematic review included randomized controlled trials, long-term follow-up studies, and observational cohorts that investigated adalimumab monotherapy with adalimumab plus …

Development and in vitro/in vivo evaluation of polymeric dissolving microneedle formulation for psoriasis treatment.

Psoriasis is an inflammatory autoimmune skin disease that significantly impacts quality of life. The chronic and recurrent nature of the disease can last a lifetime. Current conventional treatment options have several drawbacks, including inadequate or short-lived efficacy, issues with tolerability and adherence, practical limitations associated with topical applications and phototherapy, and significant safety and cost concerns related to systemic treatments. Generally, symptomatic treatments are applied, but complete recovery is rarely achieved. Researchers are exploring alternative treatmen…

Integrated Assessment of Inflammatory and Lipid-Metabolic Biomarkers in Psoriasis: Implications for Metabolic Syndrome.

(1) Background: Psoriasis is increasingly recognized as a systemic inflammatory disease associated with metabolic comorbidities. However, the hierarchical relationship between inflammatory activation and insulin resistance in driving metabolic syndrome (MetS) remains incompletely defined. This study aimed to characterize the integrated inflammatory-metabolic architecture of psoriasis using multivariate and latent domain modeling. (2) Methods: In this cross-sectional hospital-based study (2020-2022), 235 adult patients with psoriasis were evaluated. Systemic inflammatory markers (NLR, SII, CRP,…

A review of current therapeutics for psoriasis: Systemic and topical use of non-biological drugs.

PURPOSE: Psoriasis is a chronic, immune-mediated inflammatory disease characterized by dysregulated cytokine signaling, keratinocyte hyperproliferation, and recurrent relapses requiring sustained management. Although biologic agents have transformed treatment by selectively targeting IL-17, IL-23, and TNF-α pathways, important limitations persist, including high costs, limited accessibility, treatment failures, and insufficient long-term safety data. Therefore, topical therapies and non-biologic small-molecule systemic agents remain essential, particularly for mild-to-moderate disease and for …

Psoriasis and Risk of Colorectal Cancer: A Nationwide Population-Based Cohort Study in Korea.

BACKGROUND/OBJECTIVES: Psoriasis is a chronic systemic inflammatory disease associated with increased cancer risk; however, its relationship with colorectal cancer (CRC) remains unclear. This question is particularly relevant in Asia, where CRC incidence has been increasing rapidly in recent decades. METHODS: We conducted a nationwide longitudinal cohort study using the Korean National Health Insurance Service-National Sample Cohort (2002-2019). Patients with psoriasis were identified using validated ICD-10 codes and matched 1:4 with controls by age, sex, income, and region. Propensity score …

Changes in Salivary Biomarkers and Oral Immune Parameters in Patients with Psoriasis: A Systematic Review.

Background: Psoriasis is a chronic immune-mediated inflammatory disease characterized by systemic inflammation and complex immune dysregulation that extends beyond the skin and may affect the oral environment. Increasing evidence suggests that saliva may serve as a non-invasive diagnostic medium reflecting both local and systemic pathological processes. This systematic review aimed to critically evaluate current evidence on salivary biomarkers in psoriasis, focusing on inflammatory mediators, oxidative stress parameters, immune-related factors, and oral microbiota alterations, and to assess th…

Prescribing and switching patterns of biologic therapy in patients with psoriasis: real-world experience.

BACKGROUND: The advent of new biologic agents in recent years has increased treatment options for patients with psoriasis. OBJECTIVES: To evaluate the prescribing and switching patterns of biologic agents used for psoriasis in Malaysia. METHODS: Data were obtained from the Malaysian Psoriasis Registry from January 2011 to September 2022. All patients treated with biologic agents were included. Those with at least two entries were selected for subgroup analysis to evaluate switching patterns. 'Continuous therapy' was defined as those who continued with the same biologic in the subsequent foll…

The parkin-γ-tubulin axis regulates epidermal homeostasis and is associated with the susceptibility to psoriasis.

Psoriasis is an inflammatory skin disease marked by excessive proliferation of keratinocytes, and there is accumulating evidence indicating its association with Parkinson's disease (PD). However, the molecular link between these two diseases remains elusive. Herein, we identify a potential role for parkin, a PD-related E3 ubiquitin ligase, in the inhibition of psoriasis pathogenesis. The level of parkin is reduced in psoriatic skin both in clinical samples and in mouse models. Parkin-deficient mice exhibit epidermal hyperplasia, increased keratinocyte proliferation, and enhanced susceptibility…

Dupilumab-Induced Psoriasis in an Adolescent With Severe Atopic Dermatitis: Successful Management With Abrocitinib.

Psoriasiform eruptions have been described in patients treated with dupilumab. This case report describes an adolescent with severe atopic dermatitis who developed a widespread psoriasiform dermatitis 8 months after starting dupilumab. His therapy was switched to abrocitinib resulting in near-complete resolution of the psoriatic lesions with good atopic dermatitis control. This case adds to the limited pediatric literature on dupilumab-induced psoriasis and highlights abrocitinib as an effective therapeutic alternative capable of addressing both conditions concurrently.

Building the foundations for an international patient-centred outcomes set for psoriasis: A scoping study.

BACKGROUND: Value-based healthcare emphasizes outcomes that matter to patients, and patient-centred outcomes sets are vital to its success. For psoriasis, initial work proposed a patient-centred outcomes set in Belgium, but it requires further validation to ensure international applicability. OBJECTIVES: This scoping study aimed to refine the outcomes set in collaboration with patient representatives and dermatologists, in preparation for international validation through a Delphi consensus process. METHODS: An international Working Group of patient representatives and dermatologists was esta…

Development of Arthritis in a Large Real-World Cohort of Patients With Pediatric Onset Psoriasis.

BACKGROUND: Early identification and treatment of psoriatic arthritis (PsA) among patients with psoriasis is important to prevent joint damage. Insights in arthritis onset and the associated clinical factors among pediatric and young adult patients with psoriasis is scarce. OBJECTIVES: To describe pediatric patients with psoriasis who subsequently developed psoriatic arthritis at pediatric age or in young adulthood (JPsA/PsA). We focused on clinical features and timing of onset, and compared clinical features to characteristics of a large cohort of pediatric psoriasis patients. METHODS: Data…
