Ex Vivo High-Dose Ionizing Irradiation-Conditioned Nanovesicles for Enhancing the Immune Effects of Tumor Radiotherapy.

Radiotherapy (RT) exhibits a dose-dependent duality in its impact on antitumor immunity. While high-dose irradiation induces immunogenic cell death (ICD) and enhances immune responses, conventional RT is limited by toxicity and may paradoxically promote immunosuppression via myeloid-derived suppressor cells (MDSCs) infiltration and M2 macrophage polarization. To address this challenge, we leveraged ex vivo high-dose irradiation to generate tumor-derived exosomes (cExo) enriched with immunostimulatory components. Proteomic analysis revealed that cExo were enriched with damage-associated molecul

Regional transient portal ischemia and irradiation as preparative regimen for hepatocyte transplantation.

Hepatocyte transplantation is regarded as a promising option to correct hereditary metabolic liver disease. This study describes a novel method involving regional transient portal ischemia (RTPI) in combination with hepatic irradiation (IR) as a preparative regimen for hepatocyte transplantation. The right lobules of rat livers (45% of liver mass) were subjected to RTPI of 30-120 min. Liver specimens and serum samples were analyzed for transaminase levels, DNA damage, apoptosis, and proliferation. Repopulation experiments involved livers of dipeptidylpeptidase IV (DPPIV)-deficient rats precond

Feasibility of determining external beam radiotherapy dose using LuSy dosimeter.

INTRODUCTION: Radiation dose measurement is an essential part of radiotherapy to verify the correct delivery of doses to patients and ensure patient safety. Recent advancements in radiotherapy technology have highlighted the need for fast and precise dosimeters. Technologies like FLASH radiotherapy and magnetic-resonance linear accelerators (MR-LINAC) demand dosimeters that can meet their unique requirements. One promising solution is the plastic scintillator-based dosimeter with high spatial resolution and real-time dose output. This study explores the feasibility of using the LuSy dosimeter,

Comparison of methods for individualized astronaut organ dosimetry: Morphometry-based phantom library versus body contour autoscaling of a reference phantom.

One of the hazards faced by space crew members in low-Earth orbit or in deep space is exposure to ionizing radiation. It has been shown previously that while differences in organ-specific and whole-body risk estimates due to body size variations are small for highly-penetrating galactic cosmic rays, large differences in these quantities can result from exposure to shorter-range trapped proton or solar particle event radiations. For this reason, it is desirable to use morphometrically accurate computational phantoms representing each astronaut for a risk analysis, especially in the case of a so

Characterization and Etiology of Swollen Muzzles in Irradiated Mice.

Several investigators performing bone marrow transplantation studies have previously reported sporadic increases in mortality that were associated with pronounced swelling in the face, head and neck of mice. Over the past few years, we and others have noted an increasing number of experiments in which mice that have received total-body irradiation (TBI) or partial-body irradiation (PBI) develop swollen muzzles, drastic thickening of the upper lip and redness, bruising and/or swelling around the nose and muzzle and sometimes over the top of the head. We refer to this rapid and extreme swelling

Isolation and characterization of human salivary gland cells for stem cell transplantation to reduce radiation-induced hyposalivation.

BACKGROUND: Recently, we showed that transplantation of 100-300 c-Kit(+) stem cells isolated from cultured salispheres ameliorates radiation-damage in murine salivary glands. The aim of this study is to optimize and translate these findings from mice to man. METHODS: Mouse and human non-malignant parotid and submandibular salivary gland tissue was collected and enzymatically digested. The remaining cell suspension was cultured according to our salisphere culture method optimized for murine salispheres. Salisphere cells were tested using 3D matrix culturing for their in vitro stem cell charact

Development of a mantle cell lymphoma in an ATM heterozygous woman after occupational exposure to ionising radiation and somatic mutation of the second allele.

Predisposition to lymphomagenesis is a well-known phenomenon of ataxia-telangiectasia, a recessive disorder caused by germline inactivation of ATM. ATM encodes a protein implicated in the repair of radiation induced double-strand breaks. Biallelic ATM inactivation was described also in sporadic lymphoid malignancies, supporting a role of ATM as a tumour suppressor gene. It is, however, still unclear whether ATM heterozygotes are at higher risk of tumours. We describe an ATM heterozygous patient, who developed a mantle cell lymphoma (MCL) after occupational exposure to ionising radiation and so

Unraveling the early events of amyloid-β protein (Aβ) aggregation: techniques for the determination of Aβ aggregate size.

The aggregation of proteins into insoluble amyloid fibrils coincides with the onset of numerous diseases. An array of techniques is available to study the different stages of the amyloid aggregation process. Recently, emphasis has been placed upon the analysis of oligomeric amyloid species, which have been hypothesized to play a key role in disease progression. This paper reviews techniques utilized to study aggregation of the amyloid-β protein (Aβ) associated with Alzheimer's disease. In particular, the review focuses on techniques that provide information about the size or quantity of oligom

Using prior information to enhance microwave tomography images in bone health assessment.

BACKGROUND: Osteoporosis is the major cause of bone weakness and fragility in more than 10 million people in the United States. This disease causes bone fractures in the hip or spine, which result in increasing the risk of disabilities or even death. The current gold standard in osteoporosis diagnostics, X-ray, although reliable, it uses ionizing radiations that makes it unfeasible for early and continuous monitoring applications. Recently, microwave tomography (MWT) has been emerging as a biomedical imaging modality that utilizes non-ionizing electromagnetic signals to screen bones' electrica

Two molecularly distinct G(2)/M checkpoints are induced by ionizing irradiation.

Cell cycle checkpoints are among the multiple mechanisms that eukaryotic cells possess to maintain genomic integrity and minimize tumorigenesis. Ionizing irradiation (IR) induces measurable arrests in the G(1), S, and G(2) phases of the mammalian cell cycle, and the ATM (ataxia telangiectasia mutated) protein plays a role in initiating checkpoint pathways in all three of these cell cycle phases. However, cells lacking ATM function exhibit both a defective G(2) checkpoint and a prolonged G(2) arrest after IR, suggesting the existence of different types of G(2) arrest. Two molecularly distinct G

Range of Normalized Glandular Dose for Mammography Using Patient-Specific Glandular Fractions.

Breast cancer is the most common cancer among women, and mammography remains the primary modality for early detection. Because mammography uses ionizing radiation, accurate estimation of normalized glandular dose (DgN) is important for risk assessment. Recent breast dosimetry models, including TG-282, incorporate population-based glandular tissue distributions; however, patient-specific glandular distributions remain unknown from conventional mammographic projections. In previous work (Smith, Dey et al., 2025), glandular fraction (GF) maps were estimated from a single mammographic projection.
