SGLT2 inhibitors and chronic kidney disease: a narrative review of nephroprotective mechanisms from hemodynamics to metabolism.

Sodium-glucose cotransporter 2 (SGLT2) inhibitors, originally designed for the treatment of type 2 diabetes, have become a critical preventive therapy for chronic kidney disease (CKD). Moreover, they exhibit substantial reductions in the risk of kidney function deterioration, progression to end-stage kidney disease, and cardiovascular mortality in a diverse patient population. This review synthesizes current evidence to clarify the complex mechanisms underlying nephroprotection, which extend beyond modest effects on glycemia and blood pressure control. The review outlines four interrelated pat

X-ray-Responsive Cascade System for Radioimmunotherapy: Tumor Microenvironment Remodeling and Controllable NO Release.

Radioimmunotherapy holds great promise for the treatment of solid tumors. However, radioresistance and immunosuppression mediated by a tumor … underlying the immunosuppressive microenvironment reveal that NO remodels the TME by downregulating transforming growth factor beta (TGF-β), thereby inhibiting epithelial-mesenchymal transition (EMT)-associated proteins to suppress tumor metastasis and abnormal angiogenesis. Furthermore, TGF-β suppression enhances the infiltration of CD8+ T cells and reduces the proportion of FO …

Palmitoylation Induced Activation of SMAD3 Exacerbates Colitis by Promoting Tissue-resident Memory T Cells Differentiation.

Tissue-resident memory T cells (TRM cells) have been shown to play an instrumental role in driving the onset and relapse of inflammatory bowel diseases (IBD). However, the underlying mechanism of TRM cells differentiation and its regulation in intestines remain to be unveiled. Mothers against decapentaplegic homolog 3 (SMAD3) is translocated from nucleus to membrane and activated in response to transforming growth factor beta (TGF-β), which is a key cytokine in the process of TRM cells polarization. Cysteine palmitoylation (S-palmitoylation) is a post-translational modification catalyzed by th…

MGST2hi PMNs mediate NETs and drive pericyte-to-myofibroblast transition in RA-ILD.

Neutrophil extracellular trap (NET) formation has been implicated in the pathogenesis of rheumatoid arthritis-associated interstitial lung disease (RA-ILD), yet the specific neutrophil subsets involved and the mechanisms remain incompletely understood. Here, we identified a significantly expanded population of microsomal glutathione S-transferase 2-high NET-releasing polymorphonuclear neutrophils (Mgst2hi PMNs) in the lungs, bronchoalveolar lavage fluid (BALF), and peripheral blood (PB) of zymosan A-induced interstitial pneumonia-arthritis mouse models. Multimodal Biological Augmentation with Umbilical Cord Mesenchymal Stem Cell Secretome, Recombinant Human Bone Morphogenetic Protein-2, and Allogeneic Umbilical Cord Mesenchymal Stem Cells in Neurofibromatosis Type 1-Confirmed Congenital Pseudarthrosis of the Distal Tibia and Fibula: A Case Report with Mechanistic Scoping Review.

INTRODUCTION: Congenital pseudarthrosis of the tibia (CPT) associated with neurofibromatosis type 1 (NF1) is characterized by convergent dysregulation of three molecular pathways - transforming growth factor-beta/Smad-driven fibrogenesis, RAS-MEK-ERK hyperactivation, and Wnt/β-catenin blockade - none of which is addressed by conventional surgical fixation alone. This case explores the clinical feasibility of a multimodal biological protocol targeting all three pathways simultaneously. CASE REPORT: A 6-year-old male with histopathologically confirmed NF1-associated Crawford type IV CPT of the

Integrative Genome-wide Association Meta-analysis of Aortic Aneurysm and Dissection Identifies Five Novel Genes.

Aortic aneurysm and dissection (AAD) is a multifaceted condition characterized by significant genetic predisposition and a considerable contribution to cardiovascular-related mortality. Previous studies have suggested that AAD subtypes share similar genetic mechanisms; however, these studies investigated the subtypes separately. Here, we performed a large genome-wide association study (GWAS) meta-analysis for AAD by combining its subtypes, including 11,148 cases and 708,468 controls of European ancestry. We identified 24 susceptibility loci, including four novel loci at 1p21.2 (PALMD), 2p22.2

A review of the TGF-β1 pathway in Alzheimer's disease and depression: Possible restoration potential of antidepressants.

Alzheimer's Disease (AD), the most common type of dementia, is increasing rapidly in prevalence, while Major Depressive Disorder (MDD), the most frequent psychiatric condition, continues to pose significant global healthcare challenges. Recent hypotheses suggest that AD and MDD might be interrelated, potentially sharing common pathophysiological mechanisms. One possible central link is the Transforming Growth Factor-beta 1 (TGF-β1) pathway. Reduced TGF-β1 levels, a cytokine involved in inflammatory responses, have been observed in both conditions. In AD, diminished TGF-β1 might contribute to n

Persistent tissue regeneration and transforming growth factor-β induced fibrosis in the masseter muscle of mdx5Cv mice.

Clinical observational studies have shown that patients with Duchenne muscular dystrophy (DMD) often develop orofacial dysfunction. However, most DMD mouse model studies have focused on limb and respiratory muscles, showing an extensive wave of muscle necrosis-regeneration in juveniles followed by a low-intensity chronic disease in adults. In the present study, we investigated the impact of DMD on the masseter muscles in mice and observed persistence and progression of the conditions at least until 12 months of age. Masseter and limb muscles from mdx5Cv mice aged 3, 6, and 12 months were compa

C-C Chemokine Receptor 4 Deficiency Protects Against Abdominal Aortic Aneurysm Formation.

BACKGROUND: The dysregulated immune system, which drives chronic vascular inflammation and remodeling, plays a critical role in the pathogenesis of abdominal aortic aneurysm (AAA). CCR4 (C-C chemokine receptor 4), which is predominantly expressed on T cells and mediates their responses, has been shown to protect against inflammatory diseases including atherosclerosis. However, its role in AAA remains unknown. METHODS: By analyzing hypercholesterolemic CCR4-deficient (Ccr4-/-) mice on an apolipoprotein E-deficient (Apoe-/-) background, we investigated the role of CCR4 in the development of ang

Syndecan-1 regulates lipid metabolism and mitigates fibrosis during the transition from acute kidney injury to chronic kidney disease.

BACKGROUND: The transition from acute kidney injury (AKI) to chronic kidney disease (CKD) is characterized by persistent renal fibrosis, in which abnormal lipid metabolism plays a crucial role. Syndecan-1 (SDC-1) has been implicated in various tissue remodeling processes; however, its role in lipid metabolism and fibrosis during the progression from AKI to CKD is not well understood. METHODS: This study used a murine model of unilateral ischemia-reperfusion-induced AKI-to-CKD progression for in vivo analysis and employed transforming growth factor-beta (TGF-β)-induced fibrosis in Human Kidney
