Type 2 Biomarkers for the Indication and Response to Biologics in CRSwNP.

PURPOSE OF REVIEW: Three biologics targeting type 2 inflammation have been approved for the treatment of severe and uncontrolled chronic rhinosinusitis with nasal polyps (CRSwNP). Nevertheless, around 40-60% of patients do not respond well to these biological treatments. Selecting appropriate patients is crucial to improve treatment outcome of biologics. This review summarizes the literature data on type 2 biomarkers, with a specific focus on the indication to biologics for severe CRSwNP. RECENT FINDINGS: No consensus has been reached on how to define mucosal type 2 inflammation in CRSwNP. Cl

The importance of social zeitgeber in paediatric type 1 narcolepsy: What we can learn from the COVID-19 restrictions adopted in Italy?

The lockdown due to the new coronavirus pandemic (COVID-19) has led to unparalleled changes in several aspects of human behaviour. During the lockdown, the general population delayed sleep timing and spent more time in bed; however, little is known on the effects of COVID-19 restriction on children and adolescents suffering type 1 narcolepsy. In the last months of 2019, we performed follow-up actigraphy in 18 type 1 narcolepsy children and adolescents under stable pharmacological treatment with sodium oxybate. We contacted these patients for a follow-up actigraphy during the first Italian lock

Prevalence, Incidence, and Clearance of Human Papillomavirus Types Covered by Current Vaccines in Men With Human Immunodeficiency Virus in the SUN Study.

BACKGROUND: High-risk anal human papillomavirus (HPV) infection is prevalent among men living with human immunodeficiency virus (HIV); the association between 9-valent (9v) high-risk HPV (HR-HPV) vaccine types and abnormal cytology has not been well characterized. METHODS: We followed a prospective cohort study of persons with HIV at 7 HIV clinics in 4 US cities from March 2004 through June 2012. Annually, providers collected separate anal swabs for HPV detection and cytopathologic examination. Among men, we examined prevalence, incidence, and clearance of 9v HR-HPV vaccine types, compared wi

Human papillomavirus infections among couples in new sexual relationships.

BACKGROUND: No studies have examined human papillomavirus (HPV) infections among couples early in their sexual relationships when transmission is most likely. Our objective was to describe the distribution of HPV infections among recently formed couples, using the partnership as the unit of analysis. METHODS: Women aged 18-24 years attending a university or junior college in Montreal enrolled in a longitudinal study with their new male partners. Self-collected vaginal swabs and clinician-collected swabs from the penis and scrotum were tested for 36 HPV genotypes. Participants self-reported se

[Molecular diagnosis of osteogenesis imperfecta type I].

UNLABELLED: Osteogenesis imperfecta (OI) is caused by mutations in collagen type I genes. In contrast to OI type II, III and IV where there are the structural mutations, in OI type I decreased production of normal collagen is due to the presence of a null allele. Because both pharmacological and gene therapy approaches depend on type of mutation and its consequences, quick and proper diagnosis is required. AIM OF THE STUDY: Application of COL1A1 null allele detection and analysis of collagen synthesized by skin fibroblasts in OI type I diagnosis. MATERIAL AND METHODS: Analysis was carried ou

The central role of the β-cell in diabetes: from monogenic forms to type 1, type 2, and gestational diabetes.

Diabetes encompasses a heterogeneous group of metabolic disorders characterized by hyperglycemia resulting from inadequate insulin secretion and/or impaired insulin action. A central, common feature across all major forms of diabetes is the loss of functional pancreatic β-cell mass. Recent advancements have significantly highlighted the critical role of pancreatic β-cell in diabetes, primarily driven by advances in genome-wide and multi-omics approaches, the availability of isolated human islets, and the data from large clinical trials. This review synthesizes current evidence on the pathogene

Identification of ubiquitination-related biomarkers in osteoarthritis: Combining transcriptome and Mendelian randomization analysis.

BACKGROUND: Osteoarthritis is the predominant joint ailment. Multiple studies demonstrate that the dysregulation of catalytic regulators of ubiquitination and deubiquitination disrupts cartilage imbalance, consequently facilitating the advancement of osteoarthritis. METHODS: Ubiquitination-related biomarkers for osteoarthritis were found by differential expression, weighted gene co-expression network analysis, Mendelian randomization, Receiver Operating Characteristic curves, and expression analyses. Subsequently, an examination of immune infiltration was conducted to evaluate the contrasting

Patients' conceptions of opting out of group-based education for type 2 diabetes in Swedish primary care - a phenomenographic interview study.

PURPOSE: Type 2 diabetes affects everyday life, requiring patients to learn to live with the illness. Although group-based education can improve clinical outcomes and support self-care, it remains underutilised in Sweden. Only about a fifth of healthcare centres offer group-based education, and many patients opt out of participation. Understanding the various reasons patients opt out is crucial for designing group-based education that meets individual needs and promotes long-term disease management. This study aims to describe variations in the conceptions of patients with type 2 diabetes who

Cross-cancer homologous recombination deficiency prediction from whole slide images using transfer learning.

Accurate prediction of Homologous Recombination Deficiency (HRD) is vital for personalized cancer therapy, yet genomic assays are often costly and complex. Predicting HRD from Hematoxylin and Eosin (H&E) stained whole-slide images (WSIs) via deep learning offers an alternative, but model generalization across diverse cancer types remains challenging. We developed a three-stage transfer learning framework to improve HRD prediction across cancer types. After establishing baseline models on data from eight cancer types, the TCGA-BRCA model was selected as a pretrained model for transfer learning

Epicardial adipose tissue volume and diabetic retinopathy.

INTRODUCTION: We explored the association between epicardial adipose tissue (EAT) volume and diabetic retinopathy. METHODS: We used clinical data from a monocentric mixed retrospective and prospective observational study of 1093 individuals living with diabetes who had a computed tomography (CT) scan in order to calculate their coronary artery calcium (CAC) score. This scan was also used to compute EAT volume. For the present study, only persons whose diabetic retinopathy status was known (i.e., yes/no) were included. RESULTS: We included 1037 individuals living with diabetes (type 2 79.1 %,

Persistence of Vaccine Serotype Carriage and Differences in Pneumococcal Carriage by Laboratory Method and Sample Type in Indigenous Individuals in the Southwest United States.

BACKGROUND: In the United States, the invasive pneumococcal disease incidence in Indigenous populations is higher than in the general population. Molecular detection and/or specimen sampling from multiple body sites could enhance our understanding of pneumococcal carriage, a prerequisite for disease. METHODS: Persons aged <5 and ≥18 years from the Navajo Nation and White Mountain Apache Tribal lands were enrolled in an observational carriage study from October 2015 through September 2017. Swabs from the nasopharynx (all participants) and oropharynx (adults only) were collected and tested by e

Current prospects of hereditary adrenal tumors: towards better clinical management.

Adrenocortical carcinoma (ACC) and pheochromocytoma/paraganglioma (PPGL) are two rare types of adrenal gland malignancies. Regarding hereditary tumors, some patients with ACC are associated with with Li-Fraumeni syndrome (LFS), and those with PPGL with multiple endocrine neoplasia type 2. Recent studies have expanded this spectrum to include other types of hereditary tumors, such as Lynch syndrome or familial adenomatous polyposis. Individuals harboring germline TP53 pathogenic variants that cause LFS have heterogeneous phenotypes depending on the respective variant type. As an example, R337H
