Antagonism of β-klotho signaling by peptide 19 impairs wheel running in male mice and potentiates the cisplatin-induced decrease in wheel running.

BACKGROUND: Cancer-related fatigue results from competition between the metabolic demands of the tumor and those of skeletal muscles. This competition leads to an energy metabolism deficit, which is worsened by the detrimental effects of cancer therapy on mitochondrial function. Mitokines, such as Fibroblast Growth Factor 21 (FGF21), produced under these conditions help tissues adjust their metabolism to cope with cellular stress. However, the metabolic changes they trigger can be harmful if these alterations persist and become uncontrolled. Although FGF21 is involved in the metabolic adaptati

Aqueous Extract of Siraitia grosvenorii Alleviates MAFLD by Modulating Metabolism and Maintaining Gut Homeostasis in High-Fat Diet Fed Mice.

Metabolic dysfunction-associated fatty liver disease (MAFLD) is the most prevalent chronic liver disease worldwide with complex pathogenesis and no approved specific therapy. Siraitia grosvenorii is a widely used medicinal and edible herb, yet its efficacy and underlying mechanisms against MAFLD remain poorly defined. This study explored the protective effects and potential mechanisms of aqueous extract of Siraitia grosvenorii (AESG) on MAFLD. Based on ultra-high-performance liquid chromatography-linear trap quadrupole orbitrap mass spectrometry (UHPLC-LTQ-Orbitrap-MS) analysis, 38 components

Rosmarinic acid alleviates colitis and repairs mucus barrier damage in a gut microbiota-dependent manner.

Rosmarinic acid (RA) significantly alleviates DSS-induced colitis by repairing the damaged colonic mucus barrier and reversing gut microbiota dysbiosis. However, it remains unclear whether the gut microbiota is required for this protective effect. To address this, pseudo-germ-free mouse models and fecal microbiota transplantation (FMT) were used to evaluate whether the gut microbiota is required for RA to restore mucus barrier integrity and alleviate colitis. The study showed that transplanting fecal microbiota from RA-pretreated donor mice into pseudo-germ-free recipients with colitis markedl

Direct oral anticoagulants in cancer-associated venous thromboembolism: trial robustness and clinical trade-offs.

BACKGROUND: Although direct oral anticoagulants (DOACs) are often favored over low-molecular-weight heparin for cancer-associated thrombosis, the robustness of trial evidence supporting their use remains uncertain. OBJECTIVES: We evaluated the efficacy, safety, and statistical robustness of DOACs vs dalteparin in patients with cancer-associated venous thromboembolism (VTE). METHODS: We identified phase 3 to 4 randomized controlled trials comparing DOACs with dalteparin in PubMed, SCOPUS, and EMBASE up to August 2025. Primary efficacy was total recurrent VTE; secondary outcomes included recur

Hibiscus sabdariffa calyx extract protects human keratinocyte cells from fluoranthene-induced ferroptosis via the repression of aryl hydrocarbon receptor.

Polycyclic aromatic hydrocarbons (PAHs) commonly associated with fine particulate matter (PM2.5) is gaining global concern because of their diverse biological toxicity. Fluoranthene (Flu) is a low molecular weight PAH and is reported to induce carcinogenicity, cardiotoxicity and skin damages. Currently, there are no effective treatment strategies to protect the host from PAHs, rather than avoiding the PAH exposure. The understanding of the impact and molecular mechanism of Flu toxicity on keratinocytes as well as effective protective mechanisms against the toxic outcomes remain limited. Herein

Phthalates in face masks and their effects on cellular energetics in human bronchial epithelial cells.

The widespread use of disposable masks has sparked concerns about potential exposure to harmful chemicals, including the already ubiquitous phthalates. It has been demonstrated that there is a causal relationship between the exposure to phthalates and the onset of respiratory diseases. Moreover, there is an increasing body of evidence, which suggests that mitochondria contribute to significant impact to the epithelial function in respiratory diseases. However, the specific effects induced by these compounds at the mitochondrial level in respiratory cells are unknown. The objective of this stud

The Optimal Age of Helicobacter pylori Screen-and-Treat for Gastric Cancer Prevention in the United States.

BACKGROUND: Recent American College of Gastroenterology (ACG) guidelines recommend screening and eradicating Helicobacter pylori (H. pylori) in high-risk racial groups to prevent gastric cancer (GC), but do not provide guidance on the age to screen. We aimed to determine the optimal age for H. pylori screen-and-treat. MATERIALS AND METHODS: We developed a new microsimulation model, MISCAN-gastric, which was calibrated to SEER incidence and clinical studies on the natural history of GC. One-time screen-and-treat at ages 20-65 was compared to a no-screening scenario in terms of cumulative incid

Longitudinal changes in metabolism-related metrics and breast cancer risk: a general population study.

BACKGROUND: Metabolism-related metrics have been widely investigated for their relationship with breast cancer risk but are mostly based on single values. Weight gain during adulthood has been related to an increased risk of breast cancer, while the relationship with changes in glucose and lipids remain largely unknown. METHODS: Women aged 20-80 were included from the general population-based Lifelines cohort when they had two assessments: 2007-2013 and 2014-2017. The following metrics were measured once at each of the two periods: body mass index (BMI), waist to height ratio (WHtR), hemoglob

Antibiotic Prophylaxis for Dental Procedures Following Total Joint Arthroplasty: A Systematic Review and Meta-Analysis.

BACKGROUND: Periprosthetic joint infection (PJI) is a serious complication following total joint arthroplasty (TJA). Hematogenous spread, including from dental procedures, is a proposed mechanism for PJI. While prophylactic antibiotics (PAs) have traditionally been recommended before dental procedures in patients with joint prostheses, recent guidelines question their effectiveness. This study aimed to evaluate the association between dental antibiotic prophylaxis and PJI risk through a systematic review and meta-analysis. METHODS: A systematic literature search was conducted using PubMed, Em
