Isolation and characterization of a Staphylococcus aureus-lytic jumbo phage LY01 and its local therapeutic efficacy in a porcine dermatitis model.

Porcine dermatitis caused by Staphylococcus aureus (S. aureus) has garnered increasing attention due to the escalating antibiotic resistance, necessitating the development of alternative therapeutic strategies. In this study, a S. aureus-lytic jumbo phage, designated as LY01, was isolated from swine farm effluent and subsequently characterized. Key findings revealed that the bacteriophage exhibited lytic activity against S. aureus with broad host specificity. LY01 possesses a short latent period (30 min), thermal stability (30°C-50°C), and pH stability (pH 5-9). Phylogenetic analysis based on …

RPA-CRISPR/Cas12a-Mediated Isothermal Amplification Technology for Visual Detection of Fusarium proliferatum.

Fusarium proliferatum is a fungal pathogen with an exceptionally broad host range, infecting ornamental plants and forest trees such as Populus and Cedrus deodara, and causing heart rot, root rot, and other diseases that lead to severe yield losses. It can also produce mycotoxins, including fumonisins, which threaten human and animal health. Conventional detection relies on isolation, culture, and morphological identification, which are time-consuming and prone to misidentification. Here, we established a rapid, visual molecular detection method by combining recombinase polymerase amplificatio

Integrated native digestion and molecular weight cutoff enrichment with DIA-MS: a sensitive and selective approach for quantitative analysis of HCP.

Residual host cell proteins (HCPs) in biopharmaceutical production processes not only compromise drug efficacy but also pose risks to patient safety and product stability, particularly when high-risk HCPs are present. Continuous monitoring of HCPs content and species during downstream purification is therefore critical. Although liquid chromatography-mass spectrometry-based proteomics has emerged as a promising approach for HCP identification, its application is hindered by the substantial dynamic range disparity between high-abundance therapeutic proteins and trace-level HCPs (at parts-per-mi

Dual-antigen immunization strategy for nanobody screening against canine parvovirus and canine distemper virus and development of a CPV-targeted competitive ELISA for Giant panda.

Canine parvovirus (CPV) and canine distemper virus (CDV) are major pathogens causing severe diseases in giant pandas and other carnivores. Serological monitoring of host antibody responses is critical for disease surveillance. Here, we constructed a high-quality nanobody library by immunizing Bactrian camels with recombinant CPV VP2 and CDV N proteins, generating a "dual-antigen immunized" library with a capacity of 7 × 109 pfu/mL and > 90 % positivity. Through phage display, we isolated five CPV-specific and three CDV-specific nanobodies. Five CPV nanobodies were expressed as nanobody-HRP fus

The hemagglutinin-like proteins of basal vertebrate influenza-like viruses exhibit sialic-acid receptor binding disparity and their structural bases.

In 2018, two novel influenza-like virus genomes were first identified in basal vertebrates: the Asiatic toads (Bufo gargarizans) and spiny eels (Mastacembelus aculeatus). Their hemagglutinin (HA) proteins exhibit remarkably low amino acid sequences homology (23.0% and 42.8%, respectively) compared to influenza B virus (IBV), their closest canonical influenza virus relative. This study revealed that the Asiatic toad influenza-like virus HA (tHA) demonstrates dual receptor specificity, bound both α2-3 (avian-type) and α2-6 (human-type) sialic acid (SA) receptors, whereas the spiny eel influenza-

Bridging Viral Glycobiology and Lectin Biotechnology for Antiviral and Diagnostic Strategies.

Lectins, proteins that reversibly bind specific glycan motifs, offer dual utility as molecular probes or inhibitors of virus-host interactions. This review explores the molecular interactions between lectins and viral envelope glycoproteins, emphasizing their applications as antiviral agents and diagnostic tools. Enveloped viruses, such as HIV, Influenza, Herpesviruses, and Coronaviruses, exhibit dense glycosylation on their surface proteins, forming a glycan shield rich in high-mannose and complex glycans crucial for viral processes and immune evasion. Lectins exploit these glycan shields by

Saprophyticin S: A Novel Saposin-like, Leaderless Bacteriocin Produced by Staphylococcus saprophyticus P1CEA4 of Porcine Origin.

With antimicrobial resistance (AMR) posing a major global threat, largely driven by antibiotic overuse, the search for alternative antimicrobial strategies has become increasingly urgent. Bacteriocins, due to their specificity and limited impact on host microbiota, are gaining attention as viable substitutes. Here, we report the identification and characterization of a novel bacteriocin produced by Staphylococcus saprophyticus P1CEA4, a strain isolated from meat-producing pigs. This strain exhibited potent direct and extracellular antimicrobial activity. Whole-genome sequencing (WGS) enabled t

Neutrophil Extracellular Traps in the Prognosis of Sepsis: A Current Update.

Sepsis is a dysregulated host response to an infection characterized by the presence of coagulopathy and endothelial dysfunction. Neutrophil extracellular traps (NETs) are networks of extracellular fibers, primarily composed of DNA from neutrophils that bind invasive pathogens. These extracellular traps are involved in the activation and dysfunction of several pathways during the process of sepsis syndrome, including the immune response to injury, inflammation, and coagulation. Those formations consist of many molecules that have been studied as biomarkers for multiple sepsis pathophysiologica

Nanotoxin induces in situ pyroptosis to sensitize αPD1 to ablate metastases in microsattellite stable colorectal cancer.

Nanomedicine has not generated anticancer immunotherapies because of insufficient targeted specificity, lack of immunity, and associated toxicity. We developed a T22-DITOX-H6 (TDX) nanotoxin to selectively release the diphtheria toxin in the cancer cells in immunosuppressed CXCR4-overexpressing (CXCR4 +) colorectal (CRC) models, inducing pyroptosis and anticancer effect. Microsatellite stable (MSS) CRC patients do not respond to current immunotherapy. Since pyroptosis is highly immunogenic, we treated with TDX immunocompetent CXCR4 + CT26 MSS metastatic CRC models to selectively deliver the to

Potential of microRNAs as diagnostic markers for distinguishing malaria severity in samples from an Indian cohort.

BACKGROUND: Cerebral malaria (CM) is a subcategory of severe malaria (SM) and a major cause of death in Plasmodium falciparum infections, driven by the sequestration of infected red blood cells in the microvasculature of host vital organs. Identifying early biomarkers of CM is crucial for timely intervention. This study assessed the potential of microRNAs, produced upon organ injury, as biomarkers of CM. METHODS: Plasma levels of six microRNAs were quantified in patients with CM (n = 43), severe non-CM (SNCM; n = 50), uncomplicated malaria (UM; n = 79), asymptomatic malaria (AM; n = 80), and

A fundamental limit to the effectiveness of traveller screening with molecular tests.

Despite the appeal of screening travellers to prevent case importation during infectious disease outbreaks, evidence shows that symptom screening is largely ineffective in delaying the geographical spread of infection. Molecular tests offer high sensitivity and specificity and can detect infections earlier than symptom screening, suggesting potential for improved outcomes. However, they were used to screen travellers for COVID-19 with mixed success. To investigate molecular screening's role in controlling COVID-19, and to quantify the effectiveness of screening for future pathogens of concern,

Whom tuberculosis tests detect and why it matters: implications for diagnostic algorithms.

Tuberculosis encompasses a spectrum of characteristics-including bacillary burden, clinical severity, and access to care-that are relevant to clinical and epidemiological outcomes and the performance of diagnostic assays. The value of diagnostic assays depends not only on their numerical accuracy, which can vary substantially between populations, but also on which individuals with and without tuberculosis the assays identify. Moreover, detectable features of tuberculosis, such as pathogen burden or host responses, are often correlated, making it difficult to predict the accuracy and impact of

Advancing leprosy risk prediction through identification of a whole blood host transcriptomic biomarker signature including non-coding genes.

Leprosy is a debilitating disease that requires early detection for effective control, yet diagnosis still relies on clinical signs. Previous RNA-Seq analysis of coding genes from leprosy patients' household contacts (HHC) who developed leprosy (progressors) and those who did not, revealed a 4-gene RNA signature, RISK4LEP, that predicted leprosy 4-61 months before clinical onset (AUC: 0.86). To improve this signature, the present study included non-coding genes and applied novel Differential Gene Expression (DGE) analyses and machine learning approaches to the RNA-Seq dataset. This strategy id

Extracellular Vesicle-Inspired Therapeutic Strategies for the COVID-19.

Emerging infectious diseases like coronavirus pneumonia (COVID-19) present significant challenges to global health, extensively affecting both human society and the economy. Extracellular vesicles (EVs) have demonstrated remarkable potential as crucial biomedical tools for COVID-19 diagnosis and treatment. However, due to limitations in the performance and titer of natural vesicles, their clinical use remains limited. Nonetheless, EV-inspired strategies are gaining increasing attention. Notably, biomimetic vesicles, inspired by EVs, possess specific receptors that can act as "Trojan horses," p

What, how, and why? - anti-EHEC phages and their application potential in medicine and food industry.

Enterohemorrhagic Escherichia coli (EHEC) are pathogens that, only in the United States, cause more than 250,000 foodborne infections a year. Since antibiotics or other antidiarrheal agents may increase the hemolytic-uremic syndrome (HUS) development risk, currently only supportive therapy, including hydration, is used. Therefore, many methods to fight EHEC bacteria focus on their use in food processing to prevent human infection. One of the proposed anti-EHEC agents is bacteriophages, known for their bactericidal effect, host specificity, and lack of cross-resistance with antibiotics. In this…
