A structurally altered D,L-amino acid TCRalpha transmembrane peptide interacts with the TCRalpha and inhibits T-cell activation in vitro and in an animal model.

Protein-protein interactions in the membrane are pivotal for the cellular response to receptor-sensed stimuli. Recently, it has been demonstrated that an all-d-amino acids analogue of the TCRalpha transmembrane peptide (CP) is recruited to the TCR complex and inhibits T-cell activation in vitro and in vivo, similarly to the wild-type CP peptide. Here we investigated the relative contributions of the secondary structure of CP compared to its side chains in the association of CP with the TCR. We disrupted the secondary structure of CP by replacing two positive residues, needed for the interactio

Transcriptomic analysis of cecal mucosal immunity in SPF White Leghorn chicks infected with precocious and parent strains of Eimeria tenella.

INTRODUCTION: Eimeria tenella (E. tenella) preferentially invades the cecum of young chickens and causes enormous economic losses to the global poultry industry. METHODS: In this study, we established chicken infection models for the virulent parent strain and its precocious attenuated line, and collected samples at 2 dpi and 6 dpi as two critical time points. By integrating pathogenicity evaluation, transcriptome sequencing, detection of mucosal immune-related indicators, and homologous challenge protection assays, we systematically revealed the distinct molecular mechanisms underlying strai

Interleukin 6 Drives Durable T Cell-Mediated Immunity to Pancreatic Cancer.

BACKGROUND & AIMS: Tumor immune resistance is recognized as a contributor to low survivorship in pancreatic ductal adenocarcinoma (PDAC). The inflammatory cytokine interleukin-6 (IL-6) promotes polarization of CD4 T cell populations away from immune tolerance, and induces differentiation of cytotoxic CD8 T cells. This work aims to test whether IL-6 could stimulate an anti-tumor response in PDAC METHODS: We overexpressed IL-6 in multiple KrasG12D/+, Tp53R172H/+, Pdx1-Cre (KPC) cell lines, which were orthotopically implanted in mice (OT-PDACIL6). We followed mouse survival and measured tumor gro

Insights into the mechanism of intestinal flora imbalance and immune disorder in co-morbidity of pneumonia and diarrhea in children.

Pneumonia and diarrhea are the two leading causes of death in children under five years of age, and these two conditions often present as a comorbidity, where the same child experiences respiratory and digestive system infection symptoms simultaneously or sequentially. Clinical data indicate that the incidence of secondary diarrhea in children hospitalized with pneumonia is high, significantly prolonging hospital stays and affecting prognosis. In recent years, the proposal of the gut-lung axis theory has provided a novel perspective for understanding this comorbidity phenomenon. The gut-lung a

Single-Cell Multiomics Decoding of TCIRG1-Mediated Cuproptosis Circuitry Rewiring Immune-Metabolic Landscape in Ischemic Stroke.

Ischemic stroke is one of the leading causes of disability and mortality worldwide. Its pathogenesis extends beyond focal cerebral ischemia-induced neuronal injury, encompassing profound immune dysregulation and secondary inflammatory responses. Recent studies have highlighted T cell dysfunction, particularly T cell exhaustion, as a critical driver of post-stroke immunosuppression and increased susceptibility to infection. However, the molecular mechanisms underlying this process remain unclear. This study focuses on a novel immunoregulatory axis involving cuproptosis, metabolic reprogramming,

Uncovering potential molecular biomarkers for cancer-associated secondary lymphedema through integrated analyses of RNA-sequencing, machine learning, and clinical data.

BACKGROUND: Cancer-associated secondary lymphedema (CASL) commonly occurs after tumor-related lymph node dissection and radiotherapy. Nevertheless, the mechanisms of CASL remain unclear, and there are no specific molecular markers for its diagnosis and treatment. METHODS: In this study, RNA sequencing was performed on adipose tissues from 10 normal controls and 40 patients with CASL. Differentially expressed genes were screened using two machine learning algorithms to identify potential molecular markers for CASL. Subsequently, seven machine learning algorithms were employed to develop predic

Lung cancer after allogeneic hematopoietic stem cell transplantation.

INTRODUCTION: Long-term outcomes of allogeneic hematopoietic cell transplantation (HCT) are improving, increasing the relevance of secondary malignancies, including lung cancer. The clinical utility of immune checkpoint inhibitors (ICIs) in this setting remains unclear and concerns regarding graft-versus-host disease (GvHD) persist. METHODS: In this international cohort study, we evaluated clinical characteristics, survival outcomes, and toxicities in patients diagnosed with non-small cell lung cancer (NSCLC) or small-cell lung cancer (SCLC) and a history of HCT. RESULTS: A total of 77 patie

The Relationship between Traumatic Brain Injury and Systemic Health: Dissecting the Somatic and Neurological Effects.

INTRODUCTION: Traumatic brain injury (TBI) is a neurological disorder caused by external biomechanical forces such as sports impacts and vehicular accidents. It is a leading cause of morbidity and long-term disability across all age groups. Beyond acute damage, TBI contributes to chronic neurodegenerative processes and persistent neurological sequelae. Growing clinical and experimental evidence suggests that TBI is also associated with systemic alterations affecting the cardiovascular, respiratory, renal, gastrointestinal, and immune systems. METHODS: This narrative integrative review synthes

Combinatorial Delivery of Low-Dose Radiotherapy and Immunotherapy to Patients with Immune-Excluded Tumors Enhances CD8+ T-cell Functionality.

PURPOSE: Immune-checkpoint blockade (ICB) has demonstrated efficacy across tumor types. However, "cold" tumors characterized by low intraepithelial T-cell infiltration exhibit poor responsiveness. We investigated whether low-dose radiotherapy (LDRT) could enhance ICB efficacy in patients with multimetastatic immune-excluded solid tumors. PATIENTS AND METHODS: We conducted a multicohort phase I clinical trial (RACIN) involving 25 patients treated with escalating doses of LDRT in combination with a backbone regimen of nivolumab, ipilimumab, aspirin, or celecoxib and low-dose cyclophosphamide. Tonsil-derived mesenchymal stem cells alleviate skin inflammation by modulating neutrophil extracellular trap formation and T cell migration.

Skin inflammation arises from complex interactions among immune cells, particularly T cells and neutrophils. Mesenchymal stem cells (MSCs) exhibit potent immunomodulatory properties, but the specific roles of tonsil-derived MSCs (T-MSCs) in regulating neutrophil extracellular trap (NET) formation and cell death, as well as T cell migration in inflammatory skin conditions, remain poorly defined. In this study, the therapeutic effects and mechanisms of T-MSCs were investigated in a 2,4-dinitrochlorobenzene (DNCB)-induced skin inflammation model, with a focus on NET formation and T cell migration

Memory CD4+ T cells in chronic uveitis: pathogenicity, mechanisms, and therapeutic implications.

Chronic non-infectious uveitis (NIU) is a vision-threatening inflammatory disease in which relapses often occur despite corticosteroids, conventional immunosuppression, and biologic therapy. A growing body of experimental and human evidence indicates that disease persistence is not explained solely by transient effector inflammation. Instead, autoreactive memory CD4+ T cells can survive after the initiating trigger and retain retinal antigen responsiveness, sustaining persistent inflammation and reinitiating acute flares and exacerbations. In experimental chronic autoimmune uveitis, a specific

Chemokines and Motor Symptoms in First-Episode Psychosis: Are We Overlooking One of the Puzzle Pieces in Understanding the Neurological Immune Mechanisms behind Them?

INTRODUCTION: The role of chemokines in motor abnormalities (MAs) in first-episode psychosis (FEP) is underexplored. Investigating immune biomarker levels in FEP, their association with MAs, and their differences with individuals without FEP may reveal therapeutic targets. METHODS: Thirty-eight patients and thirty-four controls were included. Primary outcomes assessed group differences in chemokines related immune whole blood biomarkers, including innate (CCL2, CCL3, and CCL11), compensatory (PPARα, CXCL1, and CB2), natural immune chemotaxis biomarkers (CXCL2 and CXCR4), and growth factors (L

Advancing Biomarker Discovery and Therapeutic Targets in Duchenne Muscular Dystrophy: A Comprehensive Review.

Mounting evidence underscores the intricate interplay between the immune system and skeletal muscles in Duchenne muscular dystrophy (DMD), as well as during regular muscle regeneration. While immune cell infiltration into skeletal muscles stands out as a prominent feature in the disease pathophysiology, a myriad of secondary defects involving metabolic and inflammatory pathways persist, with the key players yet to be fully elucidated. Steroids, currently the sole effective therapy for delaying onset and symptom control, come with adverse side effects, limiting their widespread use. Preliminary
