IL-7-mediated expansion of autologous lymphocytes increases CD8+ VLA-4 expression and accumulation in glioblastoma models.

The efficacy of T cell-activating therapies against glioma is limited by an immunosuppressive tumor microenvironment and tumor-induced T cell sequestration. We investigated whether peripherally infused non-antigen specific autologous lymphocytes (ALT) could accumulate in intracranial tumors. We observed that non-specific autologous CD8+ ALT cells can indeed accumulate in this context, despite endogenous T cell sequestration in bone marrow. Rates of intratumoral accumulation were markedly increased when expanding lymphocytes with IL-7 compared to IL-2. Pre-treatment with IL-7 ALT also enhanced

ST6GAL1 promotes IBD B cell recruitment to the inflamed colon in a CD22-dependent mechanism.

BACKGROUND AND AIMS: Since the discovery that auto-reactive anti-αvβ6 integrin antibodies are associated with ulcerative colitis, cells from the B cell lineage, especially plasmablasts and plasma cells have increasingly come into the focus of research on inflammatory bowel disease. However, the mechanisms regulating their recruitment from the circulation to the gut remain poorly understood. Here, we explored whether the B cell-specific lectin CD22 interacts with endothelial α2,6-linked sialic acid residues attached by ST6GAL1 to mediate such recruitment in IBD. METHODS: Flow cytometry, transc

Matrix-associated extracellular vesicles modulate smooth muscle cell adhesion and directionality by presenting collagen VI.

The extracellular matrix (ECM) supports blood vessel architecture and functionality and undergoes active remodelling during vascular repair and atherogenesis. Vascular smooth muscle cells (VSMCs) are essential for vessel repair and, via their secretome, can invade from the vessel media into the intima to mediate ECM remodelling. Accumulation of fibronectin (FN) is a hallmark of early vascular repair and atherosclerosis. Here we show that FN stimulates VSMCs to secrete small extracellular vesicles (sEVs) by activating the β1 integrin/FAK/Src pathway as well as Arp2/3-dependent branching of the

pH-responsive neutrophil membrane camouflage Ga-Mn bimetallic nanodecoy triggers apoptosis-immunity-metastasis suppression for tumor therapy.

Mitochondria-mediated apoptosis and immunogenic cell death (ICD) are promising avenues in cancer therapy; however, they encounter challenges such as resistance to apoptosis and inadequate cytoplasmic sensing of mitochondrial DNA (mtDNA). Additionally, metastatic tumors remain challenging due to the lack of efficient systemic therapies that activate the immune system with low side effects. In this study, we introduce a pH-responsive neutrophil membrane camouflage nanoplatform (MGF@laN NPs) designed for the co-delivery of gallium (Ga3+) and manganese (Mn2+) ions, with the objective of orchestrat

Interplay Between Depression and Inflammatory Bowel Disease: Shared Pathogenetic Mechanisms and Reciprocal Therapeutic Impacts-A Comprehensive Review.

Inflammatory bowel disease (IBD) is characterized by chronic inflammation of the gastrointestinal tract. Although the aetiology of IBD remains largely unknown, several studies suggest that an individual's genetic susceptibility, external environmental factors, intestinal microbial flora, and immune responses are all factors involved in and functionally linked to the pathogenesis of IBD. Beyond the gastrointestinal manifestations, IBD patients frequently suffer from psychiatric comorbidities, particularly depression and anxiety. It remains unclear whether these disorders arise solely from reduc

Structural analyses of apolipoprotein A-IV polymorphisms Q360H and T347S elucidate the inhibitory effect against thrombosis.

Apolipoprotein A-IV (apoA-IV) is an abundant lipid-binding protein in blood plasma. We previously reported that apoA-IV, as an endogenous inhibitor, competitively binds platelet αIIbβ3 integrin from its N-terminal residues, reducing the potential risk of thrombosis. This study aims to investigate how the apoA-IVQ360H and apoA-IVT347S mutations affect the structure and function of apoA-IV. These mutations are linked to increased risk of cardiovascular diseases because of multiple single-nucleotide polymorphisms in the C-terminal region of apoA-IV. We postulate that the structural hindrance caus

Bilateral corneal pannus in Crohn's disease and assumed adalimumab-associated ocular surface disease.

PURPOSE: Anti-tumor necrosis factor (TNF)-alpha inhibitors are commonly used in the treatment of inflammatory bowel disease (IBD). Despite a beneficial risk-profile, dermatologic and ocular complications may particularly develop at higher doses. Here, we report a case of bilateral corneal pannus in a patient with Crohn's disease, which occurred following a prolonged adalimumab overdose. OBSERVATIONS: A 19-year-old male with Crohn's disease presented with progressive bilateral blurred vision, conjunctival injection, and light sensitivity. Clinical examination, including best-corrected visual a

The Junctional Epithelium Attachment Is Regulated by Wnt Signaling.

The molecular mechanisms mediating barrier functions of the junctional epithelium (JE) are incompletely understood. The aim of this study was to gain mechanistic insights into how reduced Wnt/β-catenin signaling affects the metabolism, turnover, and attachment of JE cells to the tooth surface. A membrane-permeable selective inhibitor of the Wntless protein, C59, was topically delivered to the JE. Wnt pathway suppression was verified by using Axin2LacZ/+ and Axin2CreERT2/+;R26RmTmG/+ strains of mice. Quantitative analyses were carried out at multiple time points to assess mitotic activity, apop

Immunoinformatics-driven design of a multi-epitope vaccine targeting neonatal rotavirus with focus on outer capsid proteins VP4 and VP7 and non structural proteins NSP2 and NSP5.

Rotaviral gastroenteritis remains a major global health concern, particularly for infants and young children under five years old. Prior to the introduction of the WHO-prequalified rotavirus vaccine, rotavirus (RV) was responsible for approximately 800,000 child deaths annually in developing countries. Although vaccination efforts have reduced this number, RV still causes around 200,000 child deaths each year worldwide. The current WHO-prequalified vaccines are live attenuated and offer limited efficacy of 40-60%, with a slight risk of intussusception in young children. To overcome these limit

Growth factor independent 1B is a metabolic regulator that exerts pleiotropic effects on platelet function.

BACKGROUND: Human variants in the growth factor independent 1B (GFI1B) transcription factor are associated with thrombocytopenia and a variable bleeding phenotype. OBJECTIVES: In this study, we examined the bleeding phenotype associated with the human GFI1BH294fs variant and compared it with a mouse model of abnormal GFI1B function. METHODS: The transcriptomic and proteomic profiles of human GFI1BH294fs platelets were analyzed and compared with those of murine megakaryocytes harboring conditional deletion of Gfi1b. Observed changes were subsequently validated with imaging and western blottin

Enhanced endometrial receptivity via epigallocatechin gallate (EGCG)-loaded menstrual blood-derived exosomes.

Embryo implantation failure is a significant challenge in infertility treatment, accounting for a substantial number of treatment failures. Increasing endometrial receptivity can potentially overcome this issue. This study aims to introduce a novel approach for enhancing endometrial receptivity by preparing epigallocatechin gallate (EGCG)-loaded menstrual blood-derived exosomes. Menstrual blood was used to isolate exosomes, which were then characterized for their size, zeta potential, morphology, and surface markers. EGCG was loaded into the isolated exosomes using sonication. The effects of E

Inhibition of ELOVL6 activity impairs mitochondrial respiratory function and inhibits tumor progression in FGFR3-mutated bladder cancer cells.

OBJECTIVE: Increased de novo fatty acid (FA) synthesis is a hallmark of cancer. ELOVL FA elongase 6 (ELOVL6) catalyze chain elongation of C16 saturated and monounsaturated FAs into C18 species and has been implicated in several cancers. This study investigated the role of ELOVL6 in bladder cancer (BC). METHODS: ELOVL6 expression was compared between BC and nontumor tissues. Human BC cell lines with ELOVL6-knockdown were assessed for proliferation and tumor growth. Metabolic and molecular alterations induced by ELOVL6 inhibition were analyzed using lipidomics and transcriptomics. RESULTS: ELO

KLK7 overexpression promotes an aggressive phenotype and facilitates peritoneal dissemination in colorectal cancer cells.

Colorectal cancer (CRC) incidence and mortality continue to rise globally and new prognostic biomarkers are required for the development of targeted therapies. Several studies have suggested that tissue kallikrein-related peptidases (KLKs), including KLK7, contribute to tumorigenesis. We previously demonstrated KLK7's tumor-promoting role both in vitro and in vivo, but its role in CRC metastasis remains unclear. Here, using the Cancer Genome Atlas (TCGA), we confirmed that KLK7 expression is upregulated in advanced stages of CRC and its association with shorter progression-free survival (PFS)

Leukocyte adhesion deficiency type III in an infant presenting with intestinal perforation and low percentage of natural killer cells: first case report from Iran.

BACKGROUND: Leukocyte adhesion deficiency III (LAD III) is a very rare autosomal recessive primary immunodeficiency characterized by recurrent infections without pus formation and bleeding syndrome of Glanzmann-type and life-threatening infections. The main etiology of this condition is variations in the FERMT3 gene, which encodes kindlin-3, an integrin-binding protein. We present a toddler with unique symptom of intestinal perforation followed by prolonged bleeding due to Glanzmann-like thrombasthenia who was diagnosed as LAD-III. CASE PRESENTATION: This report presents a toddler with leukoc

Macula Densa Alleviates Shiga Toxin-Induced Acute Kidney Injury via CCN1-Mediated Renal Tubular Repair.

Shiga toxins (Stx), produced by Shiga toxin-producing Escherichia coli, preferentially attack renal tissue and frequently induce acute kidney injury (AKI) and renal failure. To prevent irreversible damage, the injured renal tissue, particularly renal tubular epithelium, mounts a remodeling and regeneration response to repair itself. However, how such intrinsic renal repair processes are initiated and coordinated in infected renal tubular regions remains elusive. Herein, we reported that macula densa apparatus, in addition to its conventional role as a salt sensor in nephron, can function as an

ACPA-Induced ATP release and K+ efflux trigger NLRP3 inflammasome activation in rheumatoid arthritis.

OBJECTIVES: Anti-citrullinated protein antibodies (ACPA) is one of the key inducers of the initiation and maintenance of the immune-inflammatory response in Rheumatoid arthritis (RA), yet evidence is lacking on how ACPA drives the inflammatory response. METHODS: Citrullinated collagen-induced arthritis (C-CIA) were performed to evaluate the inflammatory response, especially NLRP3 inflammasome activation, in ACPA + arthritis model. Macrophages differentiated from THP-1 cell line were stimulated by ACPA purified from RA patients' serum, and the localization and interaction of molecules involved

Integrated network toxicology and transcriptomics analysis reveals the pathogenic mechanism linking volatile organic compounds exposure to rheumatoid arthritis.

Volatile organic compounds (VOCs), a significant category of air pollutants, have been linked to an elevated risk of rheumatoid arthritis (RA). However, their precise role in the pathogenesis and pathophysiology of RA remains poorly understood. This study aimed to elucidate the key mechanisms and targets through which VOCs contribute to RA. Potential targets related to both VOCs and RA were identified using STITCH, SwissTargetPrediction, SEA, GeneCards, and OMIM databases. Functional enrichment analysis was conducted with DAVID, while protein-protein interaction networks were constructed using

Mechanical cues enhance chondrocyte function: Insights from mechanoreception, regulation, and biological responses.

Chondrocytes, the sole cell type in articular cartilage, are responsible for synthesizing and maintaining the primary components of the extracellular matrix (ECM). In daily life, chondrocytes are subjected to diverse mechanical stimuli, and the mechanoregulation of their biological responses plays a crucial role in cartilage function. Chondrocytes exhibit remarkable mechanoadaptation, as mechanical stimulation effectively promotes their homeostasis, development, and regeneration-critical factors for regenerative medicine. Thus, a deeper understanding of chondrocyte mechanosensing mechanism is
