The Na,K-ATPase (NKA) system is the receptor for the cardioactive steroids of plant or animal origin. It is not yet known whether passive ion fluxes traverse the inactivated receptor and thereby contribute to the hormonal, pharmacological or toxic actions of these compounds. To look for putative passive ion-fluxes across the ouabain-NKA complex, we incorporated it into the artificial membrane of liposomes. Under whole-cell recording, bradykinin (BK) produced an initial outward membrane current followed by an inward current in voltage-clamped NG108-15 cells. The initial outward current was associated with a rise in intracellular Ca2+ and was accompanied by the opening of Ca(2+)-dependent K(+)-channels recorded with a cell-attached patch electrode. Using isolated healthy human leucocytes and erythrocytes as model cells, we investigated the inhibitory effect of ethanol, its metabolites and of other toxic alcohols on the active fluxes of rubidium (Rb: equivalent to K) and sodium (Na), and on Na,K-ATPase activity. Proteins obtained from the surface of human teeth in vivo were solubilized in EDTA and subjected to gel permeation, ionic exchange chromatography and amino acid analysis. It was found that the main component was anionic and was eluted between albumin and lysozyme on Sepharose. It contained abundant amounts of serine, glycine and glutamic acid. A salivary phosphoprotein of similar amino acid composition has previously been purified.
