Imaging intercellular biomolecules by using fluorescent protein indicators with lipid-PEG anchors.

Extracellular molecules such as ions, amino acids, nucleotides, and proteins are essential mediators of intercellular communication in multicellular organisms. These diffusible factors contribute to development and homeostasis of organisms by coordinating signaling among distant cells. Despite the widespread use of fluorescent indicators for studying intracellular signaling, introducing genetically encoded indicators often complicates the control of their subcellular localization, making it difficult to distinguish intracellular from extracellular signals. Consequently, monitoring the spatiote

Metabolite-driven remodeling of hepatic lipid metabolism by the plasticizer di-isononyl phthalate.

INTRODUCTION: Phthalates are widely used as plasticizers in consumer products and are suspected to be metabolism-disrupting chemicals. Di-isononyl phthalate (DINP) is commonly recognized as less hazardous substitute for more studied di(2-ethylhexyl) phthalate (DEHP). MATERIALS AND METHODS: The effects of DINP on hepatic lipid metabolism were studied using C57BL/6J mice with diet-induced obesity, and human HepaRG and C3A cell lines. The mice were orally exposed to 0, 1.5, 15 or 150 mg/kg bw/d DINP for 20 weeks, followed by assessment of glucose and insulin tolerance, hepatic histology, transcr

Relationship between lipid profiles and glycemic control in gestational diabetes mellitus women.

BACKGROUND: Gestational diabetes mellitus (GDM), a prevalent pregnancy complication, can adversely impact both maternal and neonatal health post-delivery due to poor glycemic control. Although numerous studies highlight the close association between blood lipids and GDM, the specific correlation between lipid profiles and the prognosis of GDM remains ambiguous. METHODS: This study was conducted at the Women and Children's Hospital of Ningbo University, with data collected between December 2017 and December 2018. A total of 841 GDM participants were categorized into good glycemic control (GGC,…

Peptide Substitutes of PEG: Biological Outcomes on Prospective Lipid Nanoparticle Shielding Materials.

Polyethylene glycol (PEG) is a proven component in lipid nanoparticles (LNPs). PEGylated nanoparticles are known to improve LNP circulation half-life and opsonization profiles. However, the drawbacks of PEG are hypersensitivity reactions due to pre-existing PEG antibodies and poor biodegradability. These factors warrant a search for alternatives. Some large peptides have shown potent stealth properties in protein delivery. Here, we demonstrate that repeating units of serine-threonine-glutamic acid-proline (STEP) peptide conjugated to dimyristoylglycerol (DMG-STEP) and incorporated into lipid n

GABA-mediated regulation of lipid metabolism: a conserved relation with diverse physiological significances.

Gamma-aminobutyric acid (GABA) is a four-carbon non-proteinogenic amino acid that is primarily known for its role as an inhibitory neurotransmitter in mammals. It is synthesized from glutamate by the enzyme L-glutamic acid decarboxylase and metabolized via the GABA shunt pathways. GABA is known to influence various metabolic processes, and recent studies have shown its influence on lipid metabolism. This review explores the distinct relationship between GABA and lipid metabolism by highlighting their role in physiological processes in animals, plants, and microalgae. In animals, GABA acts as a

Curcumin Metabolites and Metabolic Health: Insights into Regulatory Mechanisms of Glucose and Lipid Homeostasis.

Curcumin, a bioactive compound and vibrant yellow pigment extracted from the rhizome of Curcuma longa, has a well-established safety profile and demonstrates defined pharmacological activities. These activities include regulating glucose and lipid metabolism, exerting anti-oxidant effects, and modulating autophagy. However, its pharmacokinetic limitations-notably low oral bioavailability and insufficient blood concentration-present a dichotomy with its significant biological effects. Research indicates that curcumin undergoes rapid and extensive metabolic transformation in vivo, which suggests

Kinetic and Thermodynamic Analysis of Algal Lipid Extraction and Physicochemical Characterization for Biodiesel Production.

Marine macroalgae such as Ulva lactuca are a rich source of lipid, offering a renewable, nonedible biomass for clean energy conversion. These biological macromolecules exhibit structural diversity and functional properties that make them suitable feedstocks for biodiesel production. However, efficient utilization is hindered by a limited understanding of their extraction kinetics and thermodynamic behavior under process conditions. This study investigates the apparent extraction kinetics and thermodynamic behavior of lipids from Ulva lactuca using Soxhlet extraction, while evaluating their pot…

The Emerging Roles of Protein Lipidation in Fertility and Reproductive Disorders: Mechanisms and Therapeutic Implications.

Protein lipidation, a pivotal post-translational modification, dynamically regulates cellular signaling, membrane trafficking, and protein stability, with emerging roles in reproductive physiology. However, the systematic understanding of how distinct lipid modifications orchestrate physiological processes in male and female reproduction remains incomplete. This review systematically elaborates the mechanistic contributions of protein lipidation to gametogenesis, fertilization, and reproductive homeostasis. Finally, we discuss emerging therapeutic strategies targeting lipidation pathways-inclu

Disruption of the angiopoietin-like system connects lipid homeostasis and hypothalamic dysfunction in ALS.

BACKGROUND: Alterations in lipid metabolism are manifestations of amyotrophic lateral sclerosis (ALS) that contribute to the risk and rate of progression. Blood levels of triglycerides and cholesterol are altered in ALS patients and pre-symptomatic gene carriers, but mechanistic insights into these changes are lacking. METHODS: Serum samples from sporadic ALS patients (n = 118), mutated SOD1 and FUS/TARDBP (n = 20, 40, 17, respectively) with age and gender-matched controls (n = 96) were analysed for alterations in the angiopoietin-like protein (ANGPTL) system using enzyme-linked immunosorbent

Lipid and Foam Cell Accumulation as Novel Pathological Features of Benign Prostatic Hyperplasia.

BACKGROUND: Benign prostatic hyperplasia (BPH) encompasses a heterogeneous set of pathological processes that instigate lower urinary tract symptoms (LUTS). Using a mouse model of steroid hormone imbalance-induced lower urinary tract dysfunction, we identified luminal lipid-laden foamy macrophages, although their presence and contribution to human pathology have not yet been established. The objective of this study was to determine whether luminal macrophages are associated with human BPH and to assess how lipid accumulation relates to underlying pathological features and clinical parameters.

Swertiamarin improves glucose and lipid metabolism disorders in T2DM via targeting FXR.

BACKGROUND: Glucose and lipid metabolism disorders are key features of type 2 diabetes mellitus (T2DM). Recent studies indicate that activating the bile acid nuclear receptor FXR can effectively enhance glucose and lipid metabolism in T2DM. Swertiamarin (STM), a compound from Swertia plants affects bile acid metabolism and has been shown to improve glucose and lipid metabolism in T2DM. However, its exact mechanism is unclear. Considering the influence of STM on bile acid metabolism, it is pertinent to investigate whether its therapeutic effects in T2DM are associated with the FXR. PURPOSE: Th

Sophorolipid biosurfactants application as novel approach to lipid extraction from Chlorella cultures.

Algal lipids and high-value biomaterials have diverse applications in industries such as biofuels, nutraceuticals, pharmaceuticals, and cosmetics. They represent a promising alternative to traditional plant oils and animal fats. However, the high costs of extraction and recovery still pose significant challenges. In the present study, a novel method involving treatment of Chlorella sorokiniana cultures with sophorolipid (SL) biosurfactants, obtained from Starmerella bombicola yeast culture, was initially developed to facilitate the release of lipids prior to extraction with ethyl acetate (EA),…

Defining Composition-Cytokine Relationships Enables the Design of Lipid Nanoparticles with Programmed Immunogenicity.

Lipid nanoparticles (LNPs) are central to next-generation vaccines, yet candidate selection remains largely empirical, limiting early identification of formulations associated with rare adverse events such as myocarditis. A better understanding of LNP composition-immunogenicity relationships is therefore critical for rational vaccine design. Here, we profiled a panel of clinically relevant LNP formulations across complementary in vitro and in vivo models to define mechanisms underlying innate immune activation and adaptive responses. We identified three distinct cytokine programs: (i) a monocy

Navigating the Biomolecular Corona: Biological Interactions and Therapeutic Implications of RNA-Lipid Nanoparticles.

Lipid nanoparticles (LNPs) have reshaped RNA therapeutics, underpinning the success of mRNA vaccines and emerging gene therapies. Once introduced into the body, LNPs acquire a dynamic biological identity through the rapid adsorption of biomolecules present in biological fluids, resulting in the formation of a biomolecular corona. Rather than being a passive byproduct, the biomolecular corona substantially alters the physicochemical properties of LNPs and governs key biological processes, including cellular uptake, immune recognition, circulation, and biodistribution, therefore acting as a func
