Endometriosis is a chronic systemic disease affecting ~10% of women, yet its genetic basis and molecular mechanisms remain poorly understood. Hence, here we conducted a genome-wide association study of endometriosis and adenomyosis in ~1.4 million women, including 105,869 cases, aiming to expand loci discovery across ancestries, dissect symptom-specific effects and integrate multi-omic data. We identified 80 genomic regions associated with endometriosis risk, including 37 new loci, of which 5 are also associated with adenomyosis. We identified putative causal variants underlying over 50 of the

Stripe rust of wheat caused by Puccinia striiformis f. sp. tritici (Pst) remains a major constraint to wheat production in India due to the rapid evolution and frequent emergence of virulent pathotypes. Rapid and reliable discrimination of Pst pathotypes is essential for effective resistance deployment and surveillance. In the present study, transcriptome-wide simple sequence repeats (SSRs) and single nucleotide polymorphisms (SNPs) were exploited to develop and validate molecular markers for pathotype-specific detection of Pst pathotypes prevalent in North India (110S119, 238S119, 46S119, 110

Cold tolerance in sorghum is largely stage- and environment-specific yet partially shared genetic control allows improvement across developmental stages with mostly modest, trait-specific trade-offs. Cold tolerance is a major constraint to sorghum production in temperate environments, and its improvement is therefore a key breeding objective. However, cold tolerance varies across developmental stages and environments, and selection based on stage-specific phenotypes may overlook shared genetic components. We analysed a diversity panel of 394 sorghum genotypes evaluated across 10 environments r

Rituximab is an anti-CD20 monoclonal antibody used in autoimmune diseases, including glomerular diseases. The FCGR2A (rs1801274) and FCGR3A (rs396991) variants have been suggested to affect rituximab efficacy; this study evaluates their impact on rituximab efficacy in glomerular diseases. A clinical trial was performed including adults with glomerular diseases who required rituximab (NEFRTX; EudraCT: 2020-000484-23). Patients received 1.0 g or 0.5 g of rituximab on day 1 (± day 14); biochemical parameters and rituximab concentration were measured, and FCGR2A and FCGR3A variants were characteri

BACKGROUND: Magnusiomyces clavatus is an emerging fungal pathogen primarily infecting immunocompromised patients hospitalized in hematological wards. Its remarkable genetic homogeneity makes high-resolution phylogenetic analysis challenging, hindering efforts to accurately discriminate strains and complicating epidemiological surveillance, particularly in the context of hospital outbreaks.
METHODS: In this study, we provide the most comprehensive phylogenomic framework to date for this species by analyzing a dataset of 62 whole-genome sequences, including four novel genomes from clinical and

To clarify whether specific immune cell phenotypes exert a causal influence on diabetic nephropathy (DN), we conducted a Mendelian randomization (MR) investigation using large-scale genetic datasets. Summary-level data for immune cell traits and DN were obtained from recent genome-wide association studies and the FinnGen consortium. Genetic instruments were selected under the core MR assumptions, ensuring strong relevance and minimal confounding. The inverse-variance weighted approach served as the primary analytical framework. Complementary analyses, including MR-Egger regression, Cochran Q t

BACKGROUND: In India, indigenous camel populations have evolved under diverse ecological conditions, leading to the emergence of several distinct breeds with specialised adaptive traits. Understanding the genomic architecture and population structure of such camel populations is essential for their conservation and sustainable utilisation. In the present study, whole-genome sequencing data were generated and analysed for Indian dromedary camels representing nine recognised breeds, along with Indian Bactrian (double-humped) and Arabian Peninsula dromedaries with the primary objective to investi

Prenatal paternity testing in fetuses with chromosomal aneuploidy poses interpretative challenges for traditional short tandem repeat (STR) analysis, whereas non-invasive SNP-based approaches may offer advantages. This study compared STR and non-invasive SNP testing for paternity determination in two fetuses with Down syndrome (trisomy 21). Amniotic fluid and parental peripheral blood samples were collected. STR typing was performed using the STRtyper-21G system. Non-invasive prenatal paternity testing (NIPPT) was conducted on maternal plasma cell-free fetal DNA using a custom capture panel of

BACKGROUND: Modern commensal rodent pest control often relies on anticoagulant rodenticides (ARs), yet the development of resistance driven by mutations in the Vkorc1 gene has become a growing challenge in rodent management. Surveying the mutations in the Vkorc1 gene in rodents is essential to understanding the rodenticide resistance profile and its impacts on the environment. This study aimed to assess the distribution and frequency of Vkorc1 polymorphisms in house mice and Norway rats in New York, New Jersey, Pennsylvania, and Washington, DC, USA.
RESULTS: A total of 147 house mice (Mus mus

Catecholamine-O-methyl transferase (COMT) catalyzes the O-methylation of catechol-structured compounds such as catecholamines and catecholestrogens. A genetic variation in the human COMT gene (rs4680 or Val158Met) significantly reduces three-to-four-fold catecholamine metabolism and influences neurotransmitter levels, thereby modulating preeclampsia (PE) susceptibility. The present meta-analysis was conducted to evaluate the link between the COMT Val158Met variant and PE risk. Case-control studies published until November 20, 2025 were systematically searched from PubMed, Google Scholar, and W

Osteoporosis in women is strongly influenced by menopause, a major physiological transition that reshapes bone metabolism. Although low bone mineral density (BMD) in premenopausal women and osteoporosis in postmenopausal women share the clinical outcome of skeletal fragility, it remains unclear whether they reflect a shared molecular program or distinct regulatory mechanisms. Here, we compared genetic signals associated with premenopausal and postmenopausal low BMD in Korean women using two independent genotyping platforms with distinct variant coverage. After allele harmonization and heteroge

The melatonin receptor 1 A (MTNR1A), a highly conserved G protein-coupled receptor (GPCR), mediates crucial physiological functions. Its structure features an N-terminus responsible for melatonin binding and a C-terminus that initiates downstream signaling pathways to modulate target gene expression. Given MTNR1A's ability to form homodimers or heterodimers with other GPCRs, its expression levels are critical for the precise control of cellular signaling. This review article provides a comprehensive update on MTNR1A, highlighting recent developments concerning its expression distribution, gene

INTRODUCTION: Growth Hormone (GH), secreted by the anterior pituitary gland, is a key regulator of postnatal growth. Mutations in the GH1 gene can lead to isolated GH deficiency (IGHD), a rare disorder characterized by growth failure and severe short stature. The aim of this study was to identify the genetic basis of IGHD in three siblings born to consanguineous parents.
METHODS: Three siblings were diagnosed with short stature due to GH deficiency (stimulated GH peak levels between 0.07 and 0.77 µg/L). To identify their genetic cause, whole-exome sequencing (WES), multiplex ligation-dependen

Major depressive disorder (MDD) is a multifactorial psychiatric disorder increasingly associated with immune-inflammatory mechanisms. Pro-inflammatory cytokines, particularly tumor necrosis factor-alpha (TNF-α) and interleukin-1 beta (IL-1β), have been implicated in the pathophysiology of MDD through their influence on neuroinflammation, neurotransmitter regulation, and hypothalamic-pituitary-adrenal (HPA) axis dysfunction. This systematic review aimed to evaluate the association between TNF-α and IL-1β gene polymorphisms and susceptibility to MDD, treatment response, and related clinical outc
