Platelets: the first cellular responders in the foreign body response to blood-contacting biomaterials.

Implanting biomaterials gives rise to the foreign body response (FBR), a complex cascade consisting of blood-material interactions, provisional matrix formation, inflammation, wound healing, and remodeling. While tissue engineering seeks to harness this host response to transform implanted materials into living tissue, the FBR can drive various complications that undermine construct function and longevity with significant clinical impact for patients. The past several decades yielded important insights regarding protein adsorption dynamics and the subsequent cellular responders that exert sign…

Revolutionizing care: Recent innovations and the future of treating implant-induced infections and inflammation.

Medical implants are crucial for restoring function and regenerating injured organs and tissues. However, beyond design, material properties and placement, the risk of a foreign body reaction (FBR) upon interaction between the host immune system and the biomaterial, as well as the risk of infection at the implant site significantly influence treatment success. FBR has been confirmed to severely affect the integration of medical implants into the host tissue, often leading to therapy failure. This has led to the engineering of several novel biomaterials to improve the biocompatibility and integ

A Streamlined Workflow for the Isolation of Bovine Satellite Cells From Muscle Tissue.

BACKGROUND: Satellite cells are the primary drivers of skeletal muscle growth and regeneration and are a major cell source in both muscle tissue engineering and cultivated meat. However, current bovine satellite cell isolation can be technically demanding or yield heterogeneous cell populations. METHODS: This study presents a workflow optimized for simplicity and cost-efficiency by integrating rapid muscle processing using electric mincing with magnetic-activated cell sorting targeting Integrin-α7 to reliably enrich satellite cells. The isolated cells were characterized by Pax7 immunostaining…

Sequential addition synthesis of guar gum based ternary hydrogels with kinetic asymmetry in drug release.

Currently, biomedical engineering is placing significant emphasis on hydrogels due to their exceptional capabilities in drug delivery, tissue regeneration, tissue adhesion, blood clot prevention, contrast imaging, safeguarding tissues or organs during radiotherapy, and enhancing the biocompatibility of medical implants. This study explores a temperature-controlled sequential addition method to synthesize pH responsive guar gum-sodium alginate hydrogels with PVA as third component. Obtained hydrogels have been observed with kinetic asymmetry in drug release at specific pH. The sequential additi

Advances in diabetic wound healing: from pathophysiology to emerging therapies.

Diabetic wounds (DWs), particularly those affecting the lower extremities, represent a significant clinical challenge due to their chronic n … utic approaches, including stem cell therapy, gene therapy, nanotechnology-based interventions, and tissue engineering. These strategies improve angiogenesis, alter the polarization of macrophages, and stimulate tissue repair, which can offer new hope for enhancing wound healing in diabetic patients. This review synthesizes current literature on the pathophysiology …

Nature-inspired surface modification strategies for implantable devices.

Medical and implantable devices are essential instruments in contemporary healthcare, improving patient quality of life and meeting diverse clinical requirements. However, ongoing problems such as bacterial colonization, biofilm development, foreign body responses, and insufficient device-tissue adhesion hinder the long-term effectiveness and stability of these devices. Traditional methods to alleviate these issues frequently prove inadequate, necessitating the investigation of nature-inspired alternatives. Biomimetic surfaces, inspired by the chemical and physical principles found in biologic

Cell sheet formation enhances the therapeutic effects of adipose-derived stromal vascular fraction on urethral stricture.

Urethral stricture (US) is a common disease in urology, lacking effective treatment options. Although injecting a stem cells suspension into the affected area has shown therapeutic benefits, challenges such as low retention rate and limited efficacy hinder the clinical application of stem cells. This study evaluates the therapeutic impact and the mechanism of adipose-derived vascular fraction (SVF) combined with cell sheet engineering technique on urethral fibrosis in a rat model of US. The results showed that SVF-cell sheets exhibit positive expression of α-SMA, CD31, CD34, Stro-1, and eNOS.

Optimization of Biologically Inspired Electrospun Scaffold for Effective Use in Bone Regenerative Applications.

Human bone is composed of organic and inorganic composite materials, contributing to its unique strength and flexibility. Hydroxyapatite (HAP) has been extensively studied for bone regeneration, due to its excellent bioactivity and osteoconductivity, which makes it a highly valuable biomaterial for tissue engineering applications. For better therapeutic effects, composite nanofibers containing polyvinyl alcohol (PVA) and polyvinyl Pyrrolidone (PVP) were developed using an electrospinning technique in this study. Herein, hydroxyapatite (a major inorganic constituent of native bone) concentratio…

Enhanced bone regeneration in rat calvarial defects through BMP2 release from engineered poly(ethylene glycol) hydrogels.

The clinical standard therapy for large bone defects, typically addressed through autograft or allograft donor tissue, faces significant limitations. Tissue engineering offers a promising alternative strategy for the regeneration of substantial bone lesions. In this study, we harnessed poly(ethylene glycol) (PEG)-based hydrogels, optimizing critical parameters including stiffness, incorporation of arginine-glycine-aspartic acid (RGD) cell adhesion motifs, degradability, and the release of BMP2 to promote bone formation. In vitro we demonstrated that human bone marrow derived stromal cell (hBMS…

PSpice modeling of cervical and site-focused vagus nerve ultrasonic stimulation for reduced tumor necrosis factor-α production.

Clinical ultrasound is widely used as a diagnostic and therapeutic tool. Recently, it has been used to perform neuromodulation to treat diverse effects, including inflammation reduction through the vagus nerve. Although the mechanism by which ultrasound propagates through tissue for diagnostic purposes has been established, there is not a complete understanding of how it interacts with neurons to elicit excitation and inhibit inflammation. This work presents a novel technique based on a well-established electrical engineering tool, PSpice, to model cervical and site-focused vagus nerve ultraso

Liver Disease: Induction, Progression, Immunological Mechanisms, and Therapeutic Interventions.

The liver is an organ with impressive regenerative potential and has been shown to heal sizable portions after their removal. However, certain diseases can overstimulate its potential to self-heal and cause excessive cellular matrix and collagen buildup. Decompensation of liver fibrosis leads to cirrhosis, a buildup of fibrotic ECM that impedes the liver's ability to efficiently exchange fluid. This review summarizes the complex immunological activities in different liver diseases, and how failure to maintain liver homeostasis leads to progressive fibrotic tissue development. We also discuss a
