FOXC2 regulates macrophage polarization and osteosarcoma progression via the CXCL12/CXCR4 signaling axis.

Forkhead box protein C2 (FOXC2) is frequently overexpressed in osteosarcoma (OS) and has been implicated in tumor proliferation and metastasis; however, its role in regulating the tumor immune microenvironment remains poorly understood. In this study, integrative bioinformatics analyses of the TCGA-SARC (The Cancer Genome Atlas sarcoma cohort) dataset were performed to evaluate the clinical significance of FOXC2 in OS. High FOXC2 expression was significantly associated with poorer overall survival in patients with OS (p < 0.001). Functional experiments demonstrated that FOXC2 knockdown signifi

Dendritic cells development into osteoclast-type APCs by 4T1 breast tumor T cells milieu boost bone consumption.

Bone metastases occur in 70% of patients with advanced breast cancer, causing severe morbidity and increased mortality due to osteolytic lesions driven by osteoclasts (OCs) inside the bone marrow (BM) microenvironment. A reciprocal vicious cycle between bone remodeling system and the tumor itself is established by the release of growth factors stored in the mineralized matrix, which in turn feed the tumor, changing tumor behavior and growth. However, BM is not a passive host microenvironment for circulating tumor cells, but instead can be actively modified by the primary tumor before metastati

Antitumor activity of high-density lipoproteins and their use for targeted drug delivery.

Blood levels of high-density lipoprotein (HDL) are significantly inversely correlated with the risk of numerous malignancies and overall mortality in most tumor types. This review focuses on the antitumor role of HDL and its main protein component, apolipoprotein A-I (apoA-I). The association of HDL/apoA-I with various cancer types and the probable mechanisms of their antitumor action, including cholesterol removal from tumor cells, modulation of the tumor microenvironment, and involvement in the regulation of oncogenic signaling pathways, are discussed. The identified high expression of the S…

A self-directed Trojanbot-enzymatic nanobot in neutrobot for active target therapy of glioblastoma.

Chemotherapy is an important treatment for glioblastoma (GBM) and a key component of comprehensive GBM therapy. However, the blood-brain barrier (BBB) and complex tumor microenvironment (TME) restrict the diffusion of drugs, which greatly reduces the chemotherapeutic effect on GBM. Single strategies, such as cell-based nanobots to cross the BBB or enzymatic nanobots propelled by enriched substrates in the TME for deep tumor penetration, remain inadequate to address multiple barriers and achieve precise targeting. Here, we develop a Trojan horse-inspired enzymatic nanobot-in-neutrobot system (T…

Targeting SRSF1 improves cancer immunotherapy by dually acting on CD8+T and tumor cells.

Serine arginine-rich splicing factor 1 (SRSF1) is a key oncogenic splicing factor in various cancers, promoting abnormal gene expression through post-translational regulation. Although the protumoral function of SRSF1 is well-established, the effects of inhibiting tumor-intrinsic SRSF1 on the tumor microenvironment and its impact on CD8+ T cell-mediated antitumor immunity remain unclear. Our findings indicate that depleting SRSF1 in CD8+ T cells improve antitumor immune function, glycolytic metabolism, and the efficacy of adoptive T cell therapy. The inactivation of SRSF1 in tumor cells reduce…

CAR-T Therapy in GBM: Current Challenges and Avenues for Improvement.

Completed clinical trials of CAR-T cells in glioblastoma (GBM) have revealed key challenges that limit their efficacy. These include incomplete antigen coverage, downregulation of target antigen in response to therapy, exposure to immunosuppressive cells and cytokines in the tumor microenvironment and exhaustion of CAR-T cells. To overcome these challenges, CAR-T cells have been modified to maximize effector function and resist immunosuppression in the tumor while limiting toxicities to the host. Adoption of these novel CAR-T strategies in GBM can overcome the "cold tumor" phenotype of GBM and…

Type I IFN Activating Type I Dendritic Cells for Antitumor Immunity.

Immune checkpoint inhibitors are successful immunotherapy modalities that enhance CD8+ T-cell responses. Although T cells are initially primed in draining lymph nodes, the mechanisms that underlie their reactivation inside the tumor microenvironment are less clear. Recent studies have found that not only is the cross-priming of conventional type 1 dendritic cells (cDC1) required to initiate CD8+ T-cell responses during tumor progression, but it also plays a central role in immunotherapy-mediated reactivation of tumor-specific CD8+ T cells for tumor regression. Moreover, many cancer treatment m…

Polarization of tumor-associated macrophage phenotype via porous hollow iron nanoparticles for tumor immunotherapy in vivo.

Tumor-associated macrophages (TAMs) are the most important components in the tumor immunosuppressive microenvironment, promoting tumor growth and metastasis. Although TAMs have become one of the hot topics of tumor immunotherapy, challenges still remain to achieve TAM-targeted re-polarization therapy. In this work, porous hollow iron oxide nanoparticles (PHNPs) were synthesized for loading a P13K γ small molecule inhibitor (3-methyladenine, 3-MA) and further modified by mannose to target TAMs. The delivery system named PHNPs@DPA-S-S-BSA-MA@3-MA showed good efficiency for targeting TAMs. The in

Innate immune cells in tumor immunity: dual roles, mechanistic insights, and therapeutic opportunities.

The tumor microenvironment (TME) is a complex ecosystem that dynamically regulates tumor progression and therapeutic responses. Within this environment, innate immune cells play prominent dual roles, acting as both critical mediators of anti-tumor surveillance and facilitators of tumor immune escape. This review comprehensively synthesizes the phenotypic plasticity and functional heterogeneity of key innate immune cell subsets in tumor immunity, including tumor-associated macrophages (TAMs), natural killer (NK) cells, tumor-associated neutrophils (TANs), dendritic cells (DCs), as well as emerg…

MSCs delivering LIGHT prime immune response against CAFs to harness antigen loss variants.

The tumor niche promotes immune tolerance, enabling malignant cells to evade surveillance. Mesenchymal stem cells (MSCs) have multipotential differentiation capacity and provide a niche for bone marrow homeostasis. MSCs home to tumor tissues, where they can differentiate into Cancer-associated fibroblasts (CAFs). Within the tumor microenvironment, MSCs drive tumor progression by fostering immune suppression, secreting pro-tumorigenic cytokines, and, in some contexts, maintaining dormancy for later relapse. In this study, we engineered MSCs to deliver the immuno-stimulatory TNF superfamily liga…

Split bullets loaded nanoparticles for amplified immunotherapy.

Dendritic cells (DCs) play central role in adaptive antitumor immunity, while their function is often hampered by low immunogenicity of tumor tissues and surrounding hostile microenvironment. Herein, a "split bullets" loaded nanoplatform that can bidirectionally injure mitochondria (MT) and endoplasmic reticulum (ER) of tumor cells is developed. After cellular uptake, the released "split bullets" separately target to different subcellular destinations and exert distinct effects on DCs: (1) MT-targeted "bullet" recruits peripheral DCs into tumor sites, due to its capability to trigger adenosine
