Cytotoxicity of 17 tetrahydroisoquinoline derivatives in SH-SY5Y human neuroblastoma cells is related to mitochondrial NADH-ubiquinone oxidoreductase inhibition.

Since the first report that 1-methyl-4-phenyl-l,2,3,6-tetrahydropyridine induces parkinsonism, various kinds of low-molecular-weight neurotoxins, such as tetrahydroisoquinoline derivatives, have been identified as possible Parkinson's disease-inducing substances. In the present study, we measured four parameters of 17 tetrahydroisoquinoline derivatives, i.e., cytotoxicity in SH-SY5Y human neuroblastoma cells, inhibitory activity towards mitochondrial NADH-ubiquinone oxidoreductase (complex I), affinity for dopamine transporter, and 1-butanol-H2O partition coefficient (as an index of lipophilic…

Inhibition of the cerebellar inositol 1,4,5-trisphosphate-sensitive Ca2+ channel by ethanol and other aliphatic alcohols.

The effects of ethanol and other aliphatic alcohols on the endoplasmic reticulum Ca2+ pump and the inositol 1,4,5-trisphosphate (InsP3)-sensitive Ca2+ channel were studied in pig cerebellar microsomes. Methanol, ethanol and propanol all stimulated ATP-dependent Ca2+ uptake, whereas butanol inhibited this process. Ethanol inhibited InsP3-induced Ca2+ release [half-maximal inhibition at 3.5%, v/v (600 mM)]. However, ethanol affected only the amount of InsP3-releasable Ca2+, without affecting the concentration of InsP3 required to induce half-maximal release. Other alcohols of longer chain length

Growth of methanogenic bacteria in pure culture with 2-propanol and other alcohols as hydrogen donors.

Two types of mesophilic, methanogenic bacteria were isolated in pure culture from anaerobic freshwater and marine mud with 2-propanol as the hydrogen donor. The freshwater strain (SK) was a Methanospirillum species, the marine, salt-requiring strain (CV), which had irregular coccoid cells, resembled Methanogenium sp. Stoichiometric measurements revealed formation of 1 mol of CH(4) by CO(2) reduction, with 4 mol of 2-propanol being converted to acetone. In addition to 2-propanol, the isolates used 2-butanol, H(2), or formate but not methanol or polyols. Acetate did not serve as an energy substr

Wheat-spike-shaped arabinans from Lycium barbarum by alkaline scission-Cascade refinement and its dual-immunomodulatory activity.

To preclude the loss of glycan moieties inherent to the Sevag-mediated deproteination of Lycium barbarum polysaccharides (LBP), this study employed controlled alkaline hydrolysis instead of the classical chloroform-butanol precipitation, subsequently implemented a gradient ethanol-precipitation scheme. This strategy yielded an arabinogalactan fraction and a novel polysaccharide fraction (ALBP-80) from Lycium barbarum. The ALBP-80 was further purified by Bio-gel P100 column chromatography to obtain homogeneous fraction (ALBGP-80). Through chemical methods (monosaccharide composition, methylatio

Cross-Carboxylation of Methanol and Other Alcohols With CO2 Into Asymmetric Alkyl Methyl Carbonates Over a CeO2 Catalyst.

One-pot synthesis of asymmetric alkyl methyl carbonates from CO2, methanol (MeOH), and various alcohols was demonstrated using CeO2 and 2-cyanopyridine as a heterogeneous catalyst and dehydrating agent, respectively. The molar ratio of MeOH to the other alcohol and structure of the latter alcohol governed the distribution of three organic carbonates, that is, asymmetric alkyl methyl carbonate, dimethyl carbonate (DMC), and dialkyl carbonate. For primary alcohols such as ethanol, 1-propanol, and 1-butanol, an equimolar ratio of MeOH to the other alcohol afforded the highest distribution of the …

Chemical Profiling and Immune-Stimulating Activity of Solvent Fractions Derived from Dietary Chlorella.

Chlorella, a genus of single-celled green algae, is widely industrially cultivated as a dietary supplement ingredient due to its high protein content and various nutrients. In this study, the immune-stimulating effect of solvent fractions prepared from Chlorella vulgaris (CV) and Chlorella protothecoides (CP) was evaluated through the expression of immune-related factors, such as TNF-α and IFN-γ in T cells obtained from mice. Both Chlorella species showed immunostimulatory effects in hexane and butanol fractions, but CV increased TNF-α only, while CP elevated both TNF-α and IFN-γ. In addition,

Effects of vitamins and common drugs on reduction of 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone in rat microsomes.

4-(Methylnitrosamino)-1-(3-pyridyl)-butanone (NNK) is a tobacco-specific nitrosamino that requires metabolic activation by cytochrome P450 enzymes. The activation of NNK by cytochrome P450 enzymes leads to the formation of different metabolites. Detoxification of NNK usually occurs via carbonyl reduction to its hydroxyl product, 4-(methylnitrosamino)-1-(3-pyridyl)-butanol (NNAL). In the present study, the influences of common vitamins and P450 modulators on the reduction of NNK by rat microsomes were studied. The formation of NNAL but not other metabolites was detected by the described HPLC me
