Basement membrane invasion by first trimester human trophoblast: requirement for branched complex-type Asn-linked oligosaccharides.

Trophoblast cells of normal first trimester human placenta share with malignant tumor cells the ability for significant cellular proliferation and invasion of basement membranes. Because tumor cell metastasis in vivo and invasion of basement membranes in vitro have recently been shown to require the expression of -GlcNAc beta 1-6 Man alpha 1-6 Man beta 1-branched complex type Asn-linked oligosaccharides in tumor cell surface glycoproteins, we decided to determine if such structures were also necessary for invasion by trophoblast cells. We report here that invasive first trimester trophoblasts …

Use of sodium-chloride separated human skin in detection of circulating anti-basement membrane zone antibodies.

The sensitivity of the indirect immunofluorescence (IIF) technique for detection of circulating basement membrane zone (BMZ) antibodies was evaluated, employing NaCl-separated human skin and intact skin as substrate. Consecutive serum samples from 12 patients with clinically, histologically and immunohistologically verified bullous pemphigoid (BP) were investigated in parallel on both substrates, in dilutions ranging from 1:10 to 1:1,280. All BP sera showed linear deposits of IgG at the BMZ on intact skin, with titres ranging from 10 to 160. On NaCl-separated skin, all BP sera produced a linea…

IgG subclass distribution of autoantibodies to glomerular basement membrane in Goodpasture's syndrome compared to other autoantibodies.

The IgG subclass distribution of autoantibodies to glomerular basement membrane (anti-GBM antibodies) was investigated and compared to the distribution of liver-kidney microsomal (LKM) autoantibodies in chronic active hepatitis, to antimitochondrial autoantibodies (AMA) in primary biliary cirrhosis, and to the subclass distribution of total serum IgG within a healthy population. Solid phase assays for the demonstration of these autoantibodies were performed with four mouse monoclonal antibodies specific for each human subclass to provide quantitative data for the autoantibodies. In addition, t…

Generation of monoclonal antibodies against human carcinosarcoma cell line for identification of myoepithelium and basement membrane.

An immunohistochemical method is described for identification of myoepithelial cells and basement membrane for cryostat tissue sections of normal, benign, and in situ carcinomas of the breast using two monoclonal antibodies 155C1 and 155D10 generated against human breast carcinosarcoma cell line HS578T. In the majority of infiltrating ductal carcinomas of the breast, there was a discontinuity in the myoepithelial cell layer, as a result an intact basement membrane could not be visualized. The reactivity of these two monoclonal antibodies might prove useful in the study of myoepithelial differe…

Immunohistochemical localization of myoepithelial cells and basement membrane in normal, benign and malignant human breast lesions.

Distributions of actin and type IV collagen were investigated immunohistochemically as markers for myoepithelial cells and basement membranes. Carnoy's and Methacarn-fixed, paraffin-embedded tissues from 103 human breast lesions from 103 patients were examined; 65 with carcinomas, 27 with mastopathies, 9 with fibroadenomas and 2 with phyllodes tumours. Fifty-five samples of the normal mammary gland tissue adjacent to tumours were also included for comparison. In normal breast and benign breast diseases, type IV collagen was identified around the mammary glandular cells and actin-positive cells…

Sequence of reconstitution of seven basement-membrane components following split-thickness wound induction in primate skin.

Recent studies have shown an orderly sequence of expression of structural antigens within the skin basement-membrane zone of the human fetus … nofluorescence technique for the expression of seven antigens normally found in intact primate skin basement membrane. By the fourth day following wounding, laminin, type-IV collagen, and fibronectin were all detectable along the dermoepidermal junction. In contrast, bullous pemphigoid antigen, epidermolysis bullosa acquisita antigen, cicatricial pemphigoid antigen …

Regeneration of mouse peripheral nerves in degenerating skeletal muscle: guidance by residual muscle fibre basement membrane.

The part played by basement membrane in the guidance of peripheral nerve growth in vivo has been assessed by examining the capacity of degenerating mouse muscle to support the regeneration of the cut sciatic and saphenous nerves. Ethanol and formaldehyde-fixed gluteus maximus muscles were implanted around the contralateral cut nerves. The subsequent nerve growth into the degenerating muscle was assessed by silver staining after 3, 4 and 10 days. By 4 days, linear axonal growth was seen, parallel to the length of the muscle fibres, and coinciding with the onset of degeneration of the sarcoplasm…

Albumin deposition in dermal capillary basement membrane in parents of type 1 (insulin-dependent) diabetic patients.

Skin biopsies were performed to examine dermal capillary basement membranes for albumin by immunofluorescence microscopy in children and young adults with Type 1 (insulin-dependent) diabetes from simplex and multiplex families, their non-diabetic parents and control subjects. Circumferential vessel staining for albumin was 12.0 +/- 14.4% (mean +/- SD) in diabetic patients from multiplex families, 18.8 +/- 20.0% in diabetic patients from simplex families, and 18.5 +/- 24.2% in unaffected parents from simplex families. These values were significantly higher (p less than 0.005) than those obtaine…

Basement membrane nephropathy: a new classification for Alport's syndrome and asymptomatic hematuria based on ultrastructural findings.

Renal biopsy specimens from 19 patients with asymptomatic hematuria, normal glomeruli on light microscopic examination, and negative immunofluorescence were studied to characterize the ultrastructural changes of the glomerular basement membrane. Three groups of patients were identified. Four patients (group 1) had type I basement membrane nephropathy, characterized by marked thickening and lamellation of the basement membrane in a pattern resembling that of Alport's syndrome. Seven patients (group 2) had type II basement membrane nephropathy, which was characterized by extensive attenuation of…

Schönlein-Henoch glomerulonephritis. Characteristic ultrastructural changes in the glomerular basement membrane and localisation of osmiophilic deposits.

In glomerulonephritis accompanying the Schönlein-Henoch syndrome (SHS) a characteristic subepithelial basement membrane change is present in 85% of cases. The subepithelial change is a reaction to subepithelial deposits and consists of a garland or dome-like new formation of thin densa lamellae. This change is much more frequent in SHS than in IgA-nephritis or idiopathic glomerulonephritis or any other systemic disease. Furthermore, subepithelial deposits (50% of cases) are nearly as frequent as subendothelial deposits (65%) and more often present than formerly assumed.

Antigenic and morphologic regeneration of epidermal basement membrane in short term organ culture of human skin.

In fetal life the development of bullous pemphigoid antigen is preceded by a morphologically detectable basement membrane zone (BMZ). The present study investigated the possibility that similar morphologic and antigenic changes of the BMZ occur in short term organ culture of human skin. Twenty small pieces from each of four neonatal foreskin specimens were cultured under standard conditions. At regular intervals two pieces from each specimen were removed. One piece was frozen and cryostat sections were processed for indirect immunofluorescence using a patient's serum that was proven to have ci…

C4d deposition and multilayering of peritubular capillary basement membrane in posttransplantation membranous nephropathy indicate its association with antibody-mediated injury.

Membranous nephropathy (MN) may develop as recurrence or de novo after transplantation. Recently, autoimmune or alloimmune responses to unspecified glomerular antigens have been considered as a pathogenetic mechanism. To explore the relationship between antibody-mediated injury and posttransplantation MN, we tested C4d positivity using polyclonal antibody in renal allograft biopsy samples diagnosed as posttransplantation MN. A total of 19 cases (16 males and 3 females), including 2 recurrent and 7 de novo forms, were the subject of the study. On light microscopy, stage II was the most common (

Basement Membrane-Related Genes C1QTNF3, PTGER2, CAMK2N1, PRSS36, and B3GNT7: Novel Biomarkers for Coronary Artery Disease.

BACKGROUND: Basement membrane-related genes (BMRGs) play a pivotal role in the pathogenesis of several diseases. However, their association with coronary artery disease (CAD) remains unexplored. Therefore, this investigation is designed to elucidate the involvement of BMRGs in CAD mechanisms. METHODS: This study incorporated CAD-related datasets (GSE113079 and GSE125856) and BMRGs. Initially, differentially expressed genes (DEGs) were identified between CAD and control (CTL) groups in GSE113079 and GSE125856. Subsequently, weighted gene coexpression network analysis (WGCNA) was carried out to…

Kidney Biopsy of a Patient With Double Positive Antiglomerular Basement Membrane and Myeloperoxidase-antineutrophil Cytoplasmic Antibodies.

We report on an 82-year-old woman who developed rapidly progressive renal failure and whose serum creatinine level increased from 1.46-7.18 mg/dL over a 3-week period. Tests for antiglomerular basement membrane (anti-GBM) antibodies (136 U/mL) and myeloperoxidase-antineutrophil cytoplasmic antibodies (MPO-ANCA; 161 IU/mL) were both positive. Kidney biopsy revealed acute-phase lesions of necrotizing crescentic glomerulonephritis with fibrin deposition in 28 of 33 glomeruli. This finding was consistent with anti-GBM glomerulonephritis. Concentric necrotic lesions with fibrin deposition were seen…

Abnormal basement membrane results in increased keratinocyte-derived periostin expression in psoriasis similar to wound healing.

The psoriatic skin resembles wound healing, and it shows abnormalities at the basement membrane (BM), also in the non-lesional skin. Fibroblast-derived dermal periostin has well-known functions in wound healing and Th2-mediated diseases, such as atopic dermatitis. Here we show that serum periostin level was elevated in psoriatic patients, remarkably in the systemically treated ones. Obvious periostin positivity was detected in basal keratinocytes of the non-lesional, lesional, and previously-lesional psoriatic vs. healthy skin. Ex vivo skin models were generated to examine how different skin i…

Complement-Mediated Thrombotic Microangiopathy in a Patient With Antiphospholipid Syndrome and Anti-glomerular Basement Membrane Antibodies.

Thrombotic microangiopathy (TMA) is a range of diseases characterized by thrombocytopenia, microangiopathic hemolytic anemia, and organ injury. Complement-mediated TMA is a rare, life-threatening subtype of TMA that occurs due to the uncontrolled activation of the alternative complement pathway in the absence of normal regulation, often resulting from deficiencies of various regulatory proteins. Anti-glomerular basement membrane (anti-GBM) disease, previously known as Goodpasture syndrome, is a life-threatening form of vasculitis in which immunoglobulin G autoantibodies bind to the alpha-3 cha…
