Hierarchical Design Synergizing Local Dynamics Optimization and Global Rigidification Unlocks the Catalytic Potential of d-Erythrose-4-phosphate Dehydrogenase.

A key challenge in enzyme engineering is coordinating local catalytic dynamics with global structural integrity. Here, we present a hierarchical strategy integrating local dynamics optimization with global scaffold rigidification, using d-erythrose-4-phosphate dehydrogenase (Epd) as a model. Guided by evolutionary and conformational network analyses, we first engineered local active-site dynamics, yielding a double mutant (G14A/A234S) with 3-fold higher kcat and improved thermostability (ΔTm = 2 °C). Global rigidification further empowered the preoptimized active site, producing a quadruple mu

Novel Multienzymatic Cascade for the Conversion of Salicin From Plant Extract Into Salicylic Acid.

This study presents a novel multienzymatic strategy for the efficient and green production of valuable salicylic acid from plant-derived salicin. The biocatalytic route couples β-glucosidase for deglycosylation with laccase and xanthine oxidase (XO) for subsequent oxidation. The primary objective of this approach was to assess the feasibility of enzyme coupling within a single one-pot reaction. The results demonstrate that this strategy successfully enabled the transformation of salicylic alcohol into salicylic acid, mediated by a compatible laccase/TEMPO and XO system in a one-step process at

Bovine xanthine oxidase electrocatalysis: substrate oxidation and its role in nitrite reduction.

Xanthine oxidase plays a central role in purine catabolism in animals, plants and bacteria where it oxidises hypoxanthine to xanthine and xanthine to uric acid. In this work we have immobilised bovine xanthine oxidase on a glassy carbon electrode within a glutaraldehyde film and demonstrated hypoxanthine and xanthine oxidase electrocatalysis using mediator methylene blue as a mediator. The role of xanthine oxidase in the reduction of nitrite to nitric oxide has been probed in two ways. Using the electrochemically reduced mediator methyl viologen, nitrite reduction by xanthine oxidase is compli

Development of Novel Catalytic Deep Eutectic Solvent for Esterification and Elucidation of Dynamic Solvent-Enzyme-Substrate Adaptability.

Biocatalytic ester synthesis often encounters bottlenecks like vast substrate polarity differences and reliance on toxic solvents or activated acyl donors, severely limiting industrial expansion. To address this, a novel catalytic deep eutectic solvent (CDES) system composed of cyclohexanone, stearic acid, and N-methylimidazole (CS-NMI) was developed. This study utilized an extreme esterification, sucrose stearate synthesis, as a model reaction due to the immense substrate sizes and polarity differences. After optimization, the conversion for sucrose stearate synthesis reached 98.2± 0.7%, rema

Hierarchically engineered enzyme-packed bed reactor via 3D scaffold design and zwitterionic microenvironment for sustainable continuous catalysis.

Immobilized enzyme packed bed reactors have contributed significantly to green and sustainable chemistry, yet design strategies at both molecular and system levels are needed. In this study, polylactic acid scaffolds were printed using 3D printing. The microporous scaffold with a high specific surface area was obtained through an etching-activation process. The enzyme sources and polymer microenvironments of site-specific immobilized His-tagged penicillin G acylase were screened. The polysulfobetaine methacrylate microenvironment displayed better relative activities, affinity, storage stabilit

Glycolysis-compatible urethanases for polyurethane recycling.

Recycling thermoset polyurethanes is hindered by their cross-linked structures and chemically stable urethane bonds. Although chemo-enzymatic approaches offer promise, known urethanases remain inefficient under industrial glycolysis conditions. Here, we present GRASE [graph neural network (GNN)-based recommendation of active and stable enzymes], a GNN-based framework that integrates self-supervised and supervised learning to identify efficient, glycolysis-compatible urethanases. Among these, AbPURase exhibited two orders of magnitude greater activity than previously known enzymes in 6 molar di

Structure-Guided Engineering of Carbonyl Reductase LbCR to Simultaneously Enhance Catalytic Activity and Thermostability toward Bulky Ketones.

(S)-2-Chloro-1-(2,4-dichlorophenyl)ethanol ((S)-TCPE) is an important building block for the synthesis of antifungal drug luliconazole. Herein, a carbonyl reductase (CR) from Levilactobacillus brevis (LbCR) was identified for synthesis of (S)-TCPE. Through comprehensive Ala scanning and site-saturated mutagenesis (SSM) targeting the residues surrounding the substrate-binding pocket, the "best" variant LbCRM4 (N96V/E145A/A202L/M206A) was developed, which displays a 26.0-fold increase in catalytic activity, 83.5-fold enhancement in half-life (t1/2) at 40 °C (101.4 h), excellent enantioselectivit

Biomimetic transmembrane transport of multi-enzymes to fabricate heterogeneous bioreactors with photoenzymatic reduction of CO2.

The cascade of bio-/chemo-catalysis with quantitative integration and optimal activity is vital for catalytic conversion, but highly challenging. Here, we innovate a co-immobilization strategy by mimicking cellular transmembrane transport using ionic covalent organic framework capsules (iCOFcap) to co-encapsulate multi-enzymes and chemocatalysts. Thanks to the swellable polyethylene glycol and the adjustable charge of iCOFcap shells, multi-enzymes can be adsorbed onto the iCOFcap shells and then transported stepwise into the inner cavity of iCOFcap by adjusting electrostatic interaction betwee

From toxin to chiral building block: Engineered aldolase-catalyzed regioselective conversion of formaldehyde into L-glyceraldehyde.

Formaldehyde (FALD) is a volatile and highly toxic compound widely used in industry and a major environmental pollutant due to its genotoxic and carcinogenic effects. Developing efficient methods to convert formaldehyde into value-added, non-toxic products is essential for both environmental protection and chemical sustainability. In this study, we present a biocatalytic cascade for the selective enzymatic conversion of formaldehyde into enantiopure L-glyceraldehyde, a high-value chiral C3 compound. The system employs a structurally engineered fructose-6-phosphate aldolase (GaFSA) from Gilliam

Regiocontrol of the Bulk Polymerization of Lysine Ethyl Ester by the Selection of Suitable Immobilized Enzyme Catalysts.

The development of a green and facile method for the controlled synthesis of functional polypeptides is desired for sustainable material applications. In this study, the regioselective synthesis of poly(l-lysine) (polyLys) via enzyme-catalyzed aminolysis was achieved by bulk polymerization of l-lysine ethyl ester (Lys-OEt) using immobilized Candida antarctica lipase Novozym 435 (IM-lipase) or trypsin (IM-trypsin). Structural characterization of the obtained polyLys revealed that IM-lipase resulted solely in ε-linked amide bond formation, whereas IM-trypsin predominantly provided α-linked polyL

Boosting catalytic efficiency of lipase by regulating amphiphilic microenvironment through reversible addition-fragmentation chain transfer polymerized modifications on polyacrylonitrile fiber.

Lipases are increasingly attracting attention in green and sustainable biodiesel production. Currently, the research emphasis lies in immobilizing unstable lipase onto carriers to enhance its performance. Polyacrylonitrile fiber (PANF) is considered to be a promising material for lipase immobilization due to its excellent properties. In this study, functional carriers with regulated surface hydrophobicity were obtained by loading functional groups on PANF via reversible addition-fragmentation chain transfer (RAFT) polymerized modification, and Candida rugosa lipase (CRL) was covalently immobil

Rational design of short-chain dehydrogenase/reductase for enantio-complementary synthesis of chiral 1,2-diols by successive hydroxymethylation and reduction of aldehydes.

Enantiopure 1,2-diols are widely used in the production of pharmaceuticals, cosmetics, and functional materials as essential building blocks or bioactive compounds. Nevertheless, developing a mild, efficient and environmentally friendly biocatalytic route for manufacturing enantiopure 1,2-diols from simple substrate remains a challenge. Here, we designed and realized a step-wise biocatalytic cascade to access chiral 1,2-diols starting from aromatic aldehyde and formaldehyde enabled by a newly mined benzaldehyde lyase from Sphingobium sp. combined with a pair of tailored-made short-chain dehydr

Internal Illumination to Overcome the Cell Density Limitation in the Scale-up of Whole-Cell Photobiocatalysis.

Cyanobacteria have the capacity to use photosynthesis to fuel their metabolism, which makes them highly promising production systems for the sustainable production of chemicals. Yet, their dependency on visible light limits the cell-density, which is a challenge for the scale-up. Here, it was shown with the example of a light-dependent biotransformation that internal illumination in a bubble column reactor equipped with wireless light emitters (WLEs) could overcome this limitation. Cells of the cyanobacterium Synechocystis sp. PCC 6803 expressing the gene of the ene-reductase YqjM were used fo
