Genetic variation and fungal infection risk: State of the art.

PURPOSE OF REVIEW: Fungal infections cause significant mortality in patients with acquired immunodeficiencies including AIDS, hematological malignancies, transplantation, and receipt of corticosteroids, biologics or small-molecule kinase inhibitors that impair key immune pathways. The contribution of several such pathways in antifungal immunity has been uncovered by inherited immunodeficiencies featuring profound fungal susceptibility. Furthermore, the risk of fungal infection in patients with acquired immunodeficiencies may be modulated by single nucleotide polymorphisms (SNPs) in immune-rela

The biological and clinical relationship between CD5+23+ monoclonal B-cell lymphocytosis and chronic lymphocytic leukaemia.

A CD5(+)23(+) monoclonal B-cell population is detectable in approximately 3% of the general adult population. The phenotype of the monoclonal CD5(+)23(+) B cells is identical to chronic lymphocytic leukaemia (CLL) with respect to a large number of proteins in addition to the standard diagnostic markers used to identify CLL. Studies in CLL families and direct assessment of genetic features indicate a close biological association between indolent CLL and the CLL-phenotype cells detected in individuals with a normal blood count. Patients with a CLL-phenotype monoclonal B-cell lymphocytosis (MBL)

Detection of the fetus at risk.

Successful reproduction has ensured the survival of the human race despite disease and famine. However, a good standard of living, combined with a well-balanced diet, is closely related to low maternal, perinatal and infant mortality rates. In the third world, anaemia, rheumatic heart disease, tuberculosis and other infections are still common and the poorly nourished are more susceptible to them. In the Western world food is plentiful but often the individual's diet is poorly balanced. Invariably the fetus takes all the nutrients required from the mother and, if a deficiency occurs, this is r

Characterization of a new strain of HHV-6 (HHV-6SF) recovered from the saliva of an HIV-infected individual.

An isolate of the human herpesvirus-6 (HHV-6SF) recovered from the saliva of an HIV-infected individual differs in its cellular host range and certain genomic properties from other HHV-6 strains described. HHV-6SF replicates in adult peripheral blood mononuclear cells (PMC) substantially better than in fetal cord blood PMC and can be grown only in the MT-4 established T cell line. It preferentially infects CD4+ lymphocytes but can replicate in CD8+ cells and peripheral blood macrophages. It also infects neuroblastoma cells and cell lines derived from the gastrointestinal tract. These latter re

Effects of testosterone on gene expression in males and females across 40 human tissues.

Variation in testosterone levels is associated with pronounced health risks, often in a discordant manner between males and females. While studies have demonstrated a sex-specific genetic architecture for testosterone, the biological basis for the differential impact on diseases between the sexes is largely unknown. In this study, we correlated predicted testosterone and within-sex gene expression measures across 40 human tissues to identify genes that show sex-differential control of gene expression and examine how this varies across tissues. Gene expression measures were obtained from the Ge

Prediction of brain age using structural magnetic resonance imaging: a comparison of clinical utility of publicly available software packages.

BACKGROUND: Brain age estimated from structural magnetic resonance images is commonly used as a biomarker of biological ageing and brain health. Ideally, as a clinically useful biomarker, brain age should indicate the current state of health and be predictive of future disease onset and detrimental changes in brain biology. METHODS: In this preregistered study, we evaluated and compared the clinical utility, i.e., diagnostic and prognostic performance, of six publicly available brain age prediction packages using data from the Alzheimer's Disease Neuroimaging Initiative (ADNI). FINDINGS: Bas

Associations Between FTO (Fat Mass- and Obesity-Associated) Gene Allelic Load and Heart Rate Responses to Submaximal Exercise in Humans: A Cross-Sectional Study.

Obesity is a widespread condition strongly linked to increased cardiovascular risk, and heart rate (HR) responses to physical activity are widely used physiological indicators of this relationship. Although the role of fat mass- and obesity-associated (FTO) gene variants in obesity risk is well established, their potential associations with HR changes during and after physical activity (independent of adiposity) have not yet been fully elucidated. This study examined whether common FTO polymorphisms (rs9939609, rs1121980, rs1558902, and rs9941349) and an exploratory FTO multi-variant score (MV

An Integrated Review of Carpal Tunnel Syndrome: New Insights to an Old Problem.

Carpal tunnel syndrome (CTS) is a common entrapment neuropathy characterized by pain, numbness, and impaired function of the hand due to compression of the median nerve at the level of the wrist. Although CTS can develop from repetitive strain, injury, or medical conditions, there are also congenital and genetic risk factors that can predispose individuals to the condition. With respect to anatomical factors, some individuals are born with a smaller carpal tunnel, which increases their susceptibility to median nerve compression. Variations in specific genes, such as those encoding proteins inv

Association of alpha-1-antitrypsin deficiency with vitamin D status: who is most at risk of getting severe COVID-19?

INTRODUCTION: Coronavirus disease 2019 (COVID-19), a new disease that we do not know yet how to treat, is rapidly evolving and has forced us to stay indoors. Surprisingly, a broad range of symptoms has been reported since COVID-19 emergence. Individual variations in susceptibility to SARS-CoV-2 can be due to non-genetic and genetic factors. Alpha-1-antitrypsin deficiency (AATD) is an inherited condition that is associated with an increased risk of liver and lung diseases which may increase susceptibility to COVID-19 infection. At the same time, there could be a possibility of developing non-he

Genetic influences on smoking: a brief review.

Smoking is a complex behavior influenced by environ-mental and genetic factors. A number of twin studies demonstrate genetic influences on multiple aspects of smoking including initiation, dependence, continuation, amount consumed, and cessation. It is likely that this genetic risk involves a complex interaction between multiple genes in different biologic pathways. These may include genes involved in the general aspect of dependence (eg, genes within the dopaminergic system) and those more specific to nicotine dependence (eg, genes encoding nicotinic acetylcholine receptors and nicotine-metab

Distinct bacterial facial cutaneous microbiota composition in adolescents with acne vulgaris: a population-based cross-sectional observational study.

BACKGROUND: Acne vulgaris (acne) is a prevalent dermatological condition associated with distinct facial cutaneous microbiota compositions. However, existing research often has methodological limitations such as insufficient statistical power and the under-representation of diverse adolescent populations, which are highly relevant for understanding acne pathogenesis. OBJECTIVES: To compare bacterial facial cutaneous microbiota compositions between individuals with acne and healthy control participants in a large, multiethnic adolescent population, while adjusting for potential confounders. Venom Variation as a Window into the Ecology and Evolution of Snakes.

Snake venoms are complex biochemical systems that function primarily in prey subjugation and defense, yet their composition varies extensively across individuals, populations, species, and environments. This variation provides a powerful framework for investigating ecological and evolutionary processes. Here, we offer a forward-looking synthesis of snake venom diversity that proposes new research directions and highlights how venom variation can illuminate eco-evolutionary dynamics across biological scales. We review evidence for 10 key contexts in which venom variation arises, including withi

Serum metabolomic analysis reveals key metabolites in drug treatment of central precocious puberty in female children.

Precocious puberty (PP) is a common condition among children. According to the pathogenesis and clinical manifestations, PP can be divided into central precocious puberty (CPP, gonadotropin dependent), peripheral precocious puberty (PPP, gonadotropin independent), and incomplete precocious puberty (IPP). Identification of the variations in key metabolites involved in CPP and their underlying biological mechanisms has increased the understanding of the pathological processes of this condition. However, little is known about the role of metabolite variations in the drug treatment of CPP. Moreove

Effects of genetic ancestry and socioeconomic deprivation on ethnic differences in serum creatinine.

The inclusion of ethnicity in equations for estimating the glomerular filtration rate (eGFR) from serum creatinine levels has been challenged since ethnicity is socially defined and therefore a poor proxy for biological differences. We hypothesized that genetic ancestry (GA) would be more strongly associated with creatinine levels among healthy individuals than self-identified ethnicity. We studied a diverse cohort of 35,590 participants characterized as part of the UK Biobank, grouped by self-reported ethnicity: Black, East Asian, Mixed, Other, South Asian, and White. We used multivariable mo

Analyzing the influence of age and sex in Bothrops pauloensis snake venom.

Considerable heterogeneity and ontogenetic changes in venom composition have already been observed in different species of snakes within the Viperidae family. Since the venom of young and adult can cause distinct pathological effects and because the antivenom may be less effective in neutralizing envenoming by young snakes compared to adults, it is of paramount importance to understand the ontogenetic variation of snake venom. Thus, the present study aimed to analyze and compare the venom of Bothrops pauloensis snakes, searching for possible influences of ontogeny and sex in their biochemical

Analysis of complex patterns of human exposure and immunity to Schistosomiasis mansoni: the influence of age, sex, ethnicity and IgE.

BACKGROUND: Numerous factors may influence Schistosoma infection intensity and prevalence within endemic communities, including exposure-related factors such as local environment and behaviour, and factors relating to susceptibility to infection such as immunology and genetics. While animal studies performed in the laboratory can be tightly controlled, human populations are highly heterogeneous, varying according to demographic characteristics, genetic background and exposure to infection. The heterogeneous nature of human water contact behaviour in particular makes it difficult to distinguish

Assessing PAH exposure during construction work of railroad switches using creosote-impregnated ties.

The toxicological effects of creosote are primarily attributed to exposure to polycyclic aromatic hydrocarbons (PAHs) and related compounds. This study aimed to assess worker exposure by measuring PAHs in the breathing zone using full-shift air samples, urinary concentrations of 1-hydroxypyrene (1-HP), and dermal exposure through a tape stripping technique. Air sampling was conducted during a work shift on Day 4 (Thursday) of the work week, while urine and dermal samples were collected both before and after the shift on the same day. Additional urine samples were collected at the beginning of

Immune-related gene expression links anxiety, depression, and pain outcomes in adolescence.

BACKGROUND: Chronic pain and internalising disorders such as anxiety and depression frequently co-occur during adolescence. Although prospective associations between internalising symptoms and later pain have been reported previously, the biological mechanisms underlying individual differences in this relationship remain poorly understood. Immune- and oxidative stress-related pathways, including the NRF2 gene, may contribute to such variability. This study investigated whether polymorphisms in the NRF2 gene (rs6721961 and rs35652124) are related to the association between anxiety and depressio
