Mouse forkhead L2 maintains repression of FSH-dependent genes in the granulosa cell.

The forkhead transcription factor forkhead box L2 (FOXL2) is expressed in granulosa cells of small and medium follicles in the mouse ovary. … We found that mouse FOXL2 represses the activities of the mouse Star, Cyp19a1, Cyp11a1 promoters in CHO cells, but may not repress the Ccnd2 promoter, and identified the minimal mouse Star, Cyp19a1, and Cyp11a1 promoter regions responsive to FOXL2 regulation. We then knocked down Foxl2 in mouse granulosa cells using siRNA, which resulted in significantly in …

Assessing the Adjuvant Potential of Chinese Hamster Ovary Host Cell Proteins Using an In Vitro Dendritic Cell Assay.

Host cell proteins (HCPs) are process-related impurities of therapeutic protein production and may affect product quality or patient safety. In clinical trials, certain HCPs (e.g., PLBL2 or CCL2) that co-purify with the therapeutic protein have been associated with immune reactions in patients. In this study, we examined the adjuvant potential of six commonly detected HCPs from CHO cells (PRDX1, S100A4, PLBL2, CCL2, CLU, and YWHAE) using an in vitro dendritic cell (DC) maturation assay. Recombinant HCPs were expressed in CHO cells to mimic manufacturing conditions. PRDX1, S100A4, and PLBL2 cau…

Identification of Orthosteric GABAB Receptor Ligands by Virtual Screening and In Vitro Validation.

The GABAB receptor (GABAB-R) is a heterodimeric class C G-protein coupled receptor (GPCR) associated with numerous neurological and neuropsychiatric disorders and is an interesting target for drug development. Each subunit has an extracellular part called the Venus flytrap domain (VFT), and the VFT of the GABAB1a/b subunit contains the orthosteric γ-aminobutyric acid (GABA) binding site. In the present study, we have used a combined ligand- and structure-based virtual screening (VS) campaign to identify putative compounds binding to the orthosteric binding site. Based on the VS, 34 ligands wer

Ferritin Nanoparticle Delivery of the E2 Protein of Classical Swine Fever Virus Completely Protects Pigs from Lethal Challenge.

Classical swine fever (CSF), caused by the classical swine fever virus (CSFV), results in significant economic losses to the swine industry … aptive immune responses induced by the E2-ferritin NPs and the monomeric E2 protein produced by the CHO cells or a baculovirus expression system. The results revealed that the NP-delivered E2 protein elicited higher titers of neutralizing antibodies than did the monomeric E2 protein in pigs. Importantly, only the NP-delivered E2 protein significantly induce …

Identification, expression, function and localization of a DUF985 domain-containing hypothetical gene from amphioxus Branchiostoma belcheri.

The progress in genome sequencing has led to an increasing submission of uncharacterized hypothetical genes with the domain of unknown funct … xpressed in a tissue-specific manner, with most abundant levels in the hepatic caecum and ovary. In CHO cells transfected with the expression plasmid pEGFP-N1/BbDUF985, the fusion protein was targeted in the cytoplasm of CHO cells, suggesting that BbDUF985 is a cytosolic protein. The microtubule-associated protein tau is phosphorylated by Syk.

Aberrant phosphorylation of tau protein on serine and threonine residues has been shown to be critical in neurodegenerative disorders called … ucleinopathies. We report herein that Syk is also a tau kinase, phosphorylating tau in vitro and in CHO cells when both proteins are expressed exogenously. In CHO cells, we have also demonstrated by co-immunoprecipitation that Syk binds to tau. Finally, by site-directed mutagenesis substituting the tyrosine residues of tau with phenylalanine, we established …

Translational diffusion of individual class II MHC membrane proteins in cells.

Single-molecule epifluorescence microscopy was used to observe the translational motion of GPI-linked and native I-E(k) class II MHC membrane proteins in the plasma membrane of CHO cells. The purpose of the study was to look for deviations from Brownian diffusion that might arise from barriers to this motion. Detergent extraction had suggested that these proteins may be confined to lipid microdomains in the plasma membrane. The individual I-E(k) proteins were visualized with a Cy5-labeled peptide that binds to a specific extracytoplasmic site common to both proteins. Single-molecule trajectori…

Newly identified Chinese hamster ovary cell mutants defective in peroxisome biogenesis represent two novel complementation groups in mammals.

We isolated peroxisome biogenesis mutants from Chinese hamster ovary (CHO) cells, using the 9-(1'-pyrene)nonanol/ultraviolet (P9OH/ UV) method and wild-type CHO-K1 cells that had been stably transfected with cDNA encoding Pex2p (formerly peroxisome assembly factor-1, PAF-1). Three mutant cell clones, ZP110, ZP111, and ZP114, showed cytosolic localization of catalase, thereby indicating a defect in peroxisome biogenesis, whereas ZP112 and ZP113 contained fewer but larger catalase-positive particles. Mutant ZP115 displayed an aberrant, tubular structure immunoreactive to anti-catalase antibody.

Establishment and characterization of chromosomal aberrations in human cholangiocarcinoma cell lines by cross-species color banding.

Cholangiocarcinoma (CC), a malignant neoplasm of the biliary epithelium, is usually fatal because of difficulty in early diagnosis and lack of availability of effective therapy. Furthermore, little is known about the genetics and biology of CC. Only a few reports concerning cytogenetic studies of CC have been published, and few cell lines have been established. We recently established four CC cell lines, designated as SCK, JCK, Cho-CK, and Choi-CK, and report the first application of cross-species color banding (RxFISH) and multiple chromosome painting for the characterization of the chromosom

Two-dimensional liquid chromatography (2D-LC) isolation and purification of immunoglobulin G (IgG) and extracellular vesicles (EVs) from cell-culture supernatant using capillary-channeled polymer (C-CP) fiber columns.

There is a diversity of therapeutic agents which are manufactured through cellular processes. Prominent among these are the monoclonal antibodies (mAbs) with immunoglobulin G (IgG) being the most common variant. Newer entries to the biotherapeutic palette are the extracellular vesicles (EVs), nanosized membrane-bound particles that are released by cells into the extracellular environment, emerging as viable drug delivery vectors possessing low immunogenicity. IgG is commonly manufactured in bioreactors using Chinese hamster ovary (CHO) cells cultured in suspension, with its primary purificatio

Hydrophobicity analysis of Chinese Hamster Ovary host cell proteins using analytical hydrophobic interaction chromatography and LC-MS.

The surface hydrophobicity of the host cell proteins (HCP) present in Chinese Hamster Ovary (CHO) cell cultures can have a significant effect on the design and performance of the downstream process. This study examined the use of analytical hydrophobic interaction chromatography (HIC) in combination with mass spectrometry (LC-MS) to probe the surface hydrophobicity of the broad array of HCP present in CHO cell cultures. The HCP covered a range of hydrophobicity, with good reproducibility between cell culture samples. The GRAVY number was ineffective at predicting the hydrophobicity of the indi

Commentary on "Predicted plasma 25-hydroxyvitamin D and risk of renal cell cancer." Joh HK, Giovannucci EL, Bertrand KA, Lim S, Cho E, Department of Medicine, Seoul National University College of Medicine, Seoul, South Korea. J Natl Cancer Inst 2013; 105(10):726-32. [Epub 2013 Apr 8]. doi: 10.1093/jnci/djt082.

BACKGROUND: Although the kidney is a primary organ for vitamin D metabolism, the association between vitamin D and renal cell cancer (RCC) remains unclear. METHODS: We prospectively evaluated the association between predicted plasma 25-hydroxyvitamin D [25(OH)D] and RCC risk among 72,051 women and 46,380 men in the period from 1986 to 2008. Predicted plasma 25(OH)D scores were computed using validated regression models that included major determinants of vitamin D status (race, ultraviolet B flux, physical activity, body mass index, estimated vitamin D intake, alcohol consumption, and postmen

Active oxygen species are involved in the induction of micronuclei by arsenite in XRS-5 cells.

The human carcinogen, arsenic, is genotoxic to mammalian cells in vitro. The mechanism is largely unknown, although the involvement of free radicals has been suggested. Since the X-ray sensitive Chinese hamster ovary cell line, XRS-5, is also sensitive to several free-radical generating agents, including H2O2, we have used this cell line to test whether the genotoxic effect of arsenite is mediated via the generation of active-oxygen species. The results indicate that the XRS-5 cells are more sensitive to arsenite in terms of cell-killing and micronucleus induction compared to the parental CHO-
