Distribution of calcium in central nervous system tissues and bones of rats maintained on calcium-deficient diets.

Current changing epidemiological pattern in the Western Pacific area suggests a contribution of the environmental factors to the pathogenetic process of amyotrophic lateral sclerosis (ALS). The condition of unbalanced mineral levels found in the soil and drinking water samples from the ALS foci showing low content of calcium (Ca) and magnesium (Mg) plus high content of aluminum (Al) was experimentally mimicked in this study using rats. In the groups fed low Ca, low Ca-Mg, and low Ca-Mg plus high Al diets, serum Ca levels were lower than that in the group fed the standard diet. Ca content of CN…

Positive inotropic effects of calcium channel antagonists are not necessarily caused by partial calcium channel agonism.

Recently it has been reported that some dihydropyridine calcium channel antagonists (nifedipine, nimodipine, nitrendipine) are able to produce positive inotropic effects in isolated perfused guinea pig hearts. We studied the effects of nifedipine in isolated perfused paced rat hearts under constant pressure and constant flow perfusion conditions. We found that nifedipine is able to produce a positive inotropic effect under constant pressure conditions but not under constant flow conditions. We conclude that nifedipine does not have partial calcium channel agonistic properties and that the posi…

Effects of calcium, calcium entry blockers and calmodulin inhibitors on atrioventricular conduction disturbances induced by hypoxia.

Effects of hypoxia on atrioventricular conduction were investigated in the Langendorff-perfused isolated heart of the rabbit with various extracellular calcium concentrations ([Ca2+]) as well as in the presence of verapamil, nifedipine, N-(6-aminohexyl)-5-chloro-1-naphthalenesulphonamide (W-7) and chlorpromazine. The prolongation of the atrio-His (AH) interval by hypoxia for 7 min was greater with increasing [Ca2+]o ranging from 1.2 to 5.2 mM. At [Ca2+]o of over 3.2 mM under hypoxic conditions, AH block of the Wenckebach type was observed in some cases. Verapamil (5 X 10(-8) M) and nifedipine …

Calcium binding sites in the insect polytrophic egg vesicles as possible markers of the route of inflow of calcium.

Calcium binding sites were revealed in the polytropic egg vesicles of Galleria mellonella (Lepidoptera) by fixation of the ovariole with glutaraldehyde in the presence of excess of calcium. Calcium dependent deposits appeared in the mitochondrial matrix in cells of the epithelial sheath and of the follicular epithelium. The deposits were also seen in the extracellular space around the follicular epithelial cells and around the trophocytes , at the oolemma, and in the coated vesicles of the oocytes at the stage of vitellogenesis. On the other hand, the deposits were not detected in the mitochon…

Failure of total calcium corrected for protein, albumin, and pH to correctly assess free calcium status.

The clinical effectiveness of published algorithms in correcting serum total calcium (CaT) for the effects of protein, albumin, and pH was tested. Corrected calcium (CaC) values obtained by 13 of these methods were compared with values of measured free calcium (CaF) in 55 samples from normal controls and 404 samples from patients with various disorders of calcium metabolism. Three criteria were used to compare either CaC or CaT with measured CaF: 1) the correlation coefficient, 2) the average absolute deviation from measured CaF of the values of CaF predicted by the linear regression of CaF on…

Structure, Stability, and Calcium Transport Mechanism of a Novel Duck Plasma Peptide-Calcium Chelate (LHDVK-Ca).

This study constructed a peptide-calcium chelate (LHDVK-Ca) using the duck plasma-derived pentapeptide LHDVK to investigate its binding mechanism, stability, and intestinal transport behavior. Spectroscopic analysis combined with isothermal titration calorimetry (ITC) revealed that LHDVK binds Ca2+ primarily through the carboxyl group of the aspartic acid residue with a single binding site and moderate affinity. Molecular simulations confirmed that Asp plays a dominant role in coordinating Ca2+. The chelate exhibited enhanced thermal stability and maintained high calcium retention under simula…

Mechanistic insights into oxalate-based desensitizers: Calcium oxalate deposition and reduced calcium release under acid challenge.

OBJECTIVE: Oxalate salts are used clinically as a desensitizing agent; however, the physicochemical mechanisms underlying its interaction with hard dental tissues are unclear. This study aimed at elucidating how oxalic acid treatment affects the demineralization behavior of hydroxyapatite (HAp), focusing on ion redistribution, phase formation, and the reversibility of its protective effect. METHODS: HAp powder was treated with oxalic acid solutions at concentrations ranging from 0.001 to 0.5 M. Calcium and phosphorus released from dentin surfaces during oxalic acid treatment and subsequent la…

Preparation optimization, structural characterization, stability and promoting calcium absorption mechanism of sturgeon cartilage peptide-calcium chelates.

The preparation process of sturgeon cartilage peptide (SCP)‑calcium chelates (SCP-Ca) was optimized in this work, its structure, stability and promoting calcium absorption mechanism were explored. The optimum conditions for preparing SCP-Ca with Ca amount of 44.47 ± 0.87 mg/g were 50 min, 40 °C, pH of 8 and a peptide-to‑calcium mass ratio of 11:1. The C-O and -COO- groups of Asp and Glu mainly attended SCP-Ca formation, and resulted to the change of random coil and β-sheet contents in SCP. SCP-Ca showed good processing and digestive stability. SCP-Ca strengthened calcium transport volume with …

Incremental prognostic value of coronary artery calcium progression within a large community-benefit calcium score registry.

BACKGROUND: Coronary artery calcium (CAC) scoring is a robust cardiovascular risk marker, yet the prognostic value of serial assessment and the relative importance of incident calcium versus progression from elevated baseline CAC remains incompletely defined. METHODS: We analyzed 4166 CLARIFY registry participants (NCT04075162) who underwent ≥2 CAC scans ≥11 months apart (mean interval 4.5 years). Defining CAC progression (ΔCAC>10 Agatston units, AU) identified 1982 progressors and 2184 non-progressors. Annualized absolute, square-root, and log-transformed progression were evaluated for assoc…

Heat stability of high-protein calcium-fortified milk protein concentrate dispersions: Impact of different calcium salts.

Calcium fortification remains a challenge for high-protein beverages. The current study investigated the impact of different calcium fortification, i.e., calcium chloride (CaCl₂), calcium gluconate (CaGlu), and chelated calcium casein pHosphopeptide (CaCPP) on the physicochemical properties and heat stability of high-protein (8.0 % protein) milk protein concentrate (MPC) dispersions. MPC suspensions were fortified with 2.5-15 mM calcium in the form of CaCl2, CaGlu and CaCPP then analyzed for pH, zeta potential, calcium ion activity, heat coagulation time (HCT), sedimentation ratio, ethanol sta…

Mitochondrial Calcium Uniporter (MCU)-Mediated Calcium Overload in Psychoactive Drug Neurotoxicity: From Pathogenesis to Therapeutic Targets.

With rapid societal changes and increasing stress levels, the abuse of psychoactive substances has emerged as a global health crisis. Studies indicate that the mitochondrial calcium uniporter (MCU) plays a pivotal role in neurotoxic damage induced by psychoactive substances. As the primary channel for mitochondrial Ca2+ uptake, MCU dysfunction can lead to Ca2+ overload, oxidative stress, and apoptosis, representing a crucial mechanism underlying neurotoxic damage. Psychoactive substances such as 3,4-Methylenedioxymethamphetamine (MDMA), cocaine, and morphine influence MCU function through mult…

Coronary artery calcium and atherosclerotic cardiovascular disease risk scores in patients with calcium pyrophosphate deposition disease.

OBJECTIVE: Calcium pyrophosphate deposition (CPPD) disease is associated with an increased risk for cardiovascular (CV) events. We examined the atherosclerotic burden by coronary artery calcium (CAC) scores (Agatston score) and compared 10-year atherosclerotic CV disease (ASCVD) risk scores in patients with vs without chondrocalcinosis, a radiographic marker of CPPD. METHODS: We performed a cross-sectional analysis at an academic medical centre, 1991-2022. Among all patients with an Agatston score in routine care, we defined a cohort with chondrocalcinosis detected before the CAC scan. Compar…

EF-hand calcium sensor, EfhP, controls transcriptional regulation of iron uptake by calcium in Pseudomonas aeruginosa.

The human pathogen Pseudomonas aeruginosa (Pa) poses a major risk for a range of severe infections, particularly lung infections in patients suffering from cystic fibrosis (CF). As previously reported, the virulent behavior of this pathogen is enhanced by elevated levels of Ca2+ that are commonly present in CF nasal and lung fluids. In addition, a Ca2+-binding EF-hand protein, EfhP (PA4107), was partially characterized and shown to be critical for the Ca2+-regulated virulence in P. aeruginosa. Here, we describe the rapid (10 min, 60 min), and adaptive (12 h) transcriptional responses of PAO1 t

Mean Versus Peak Coronary Calcium Density on Non-Contrast CT: Calcium Scoring and ASCVD Risk Prediction.

OBJECTIVES: This study sought to assess the relationship between mean vs peak calcified plaque density and their impact on calculating coronary artery calcium (CAC) scores and to compare the corresponding differential prediction of atherosclerotic cardiovascular disease (ASCVD) and coronary heart disease (CHD) mortality. BACKGROUND: The Agatston CAC score is quantified per lesion as the product of plaque area and a 4-level categorical peak calcium density factor. However, mean calcium density may more accurately measure the heterogenous mixture of lipid-rich, fibrous, and calcified plaque ref…

Elucidating the Calcium-Binding Site, Absorption Activities, and Thermal Stability of Egg White Peptide-Calcium Chelate.

With the current study, we aimed to determine the characteristics and calcium absorption capacity of egg white peptide-calcium complex (EWP-Ca) and determine the effect of sterilization on EWP-Ca to study the possibility of EWP-Ca as a new potential calcium supplement. The results of SEM and EDS showed a high calcium chelating ability between EWP and calcium, and the structure of EWP-Ca was clustered spherical particles due its combination with calcium. The FTIR and Raman spectrum results showed that EWP could chelate with calcium by carboxyl, phosphate, and amino groups, and peptide bonds may…

Spontaneous Calcium Oscillations through Differentiation: A Calcium Imaging Analysis of Rat Cochlear Nucleus Neural Stem Cells.

Causal therapies for the auditory-pathway and inner-ear diseases are still not yet available for clinical application. Regenerative medicine approaches are discussed and examined as possible therapy options. Neural stem cells could play a role in the regeneration of the auditory pathway. In recent years, neural stem and progenitor cells have been identified in the cochlear nucleus, the second nucleus of the auditory pathway. The current investigation aimed to analyze cell maturation concerning cellular calcium activity. Cochlear nuclei from PND9 CD rats were microscopically dissected and propa…
