Mitochondrial redox impairment and enhanced autophagy in peripheral blood mononuclear cells from type 1 diabetic patients.

Type 1 diabetes (T1D) involves critical metabolic disturbances that contribute to an increased cardiovascular risk. Leukocytes are key players in the onset of atherosclerosis due to their interaction with the endothelium. However, whether mitochondrial redox impairment, altered bioenergetics and abnormal autophagy in leukocytes contribute to T1D physiopathology is unclear. In this study we aimed to evaluate the bioenergetic and redox state of peripheral blood mononuclear cells (PBMCs) from T1D patients in comparison to those from healthy subjects, and to assess autophagy induction and leukocyt

Interaction of cyanide and nitric oxide with cytochrome c oxidase: implications for acute cyanide toxicity.

Acute cyanide toxicity is attributed to inhibition of cytochrome c oxidase (CcOX), the oxygen-reducing component of mitochondrial electron transport; however, the mitochondrial action of cyanide is complex and not completely understood. State-3 oxygen consumption and CcOX activity were studied in rat N27 mesencephalic cells to examine the functional interaction of cyanide and nitric oxide (NO). KCN produced a concentration-dependent inhibition of cellular respiration. Cyanide's median inhibitory concentration (IC50) of oxygen consumption (13.2 +/- 1.8microM) was higher than the CcOX IC50 (7.2

Role of adenosine as an endogenous regulator of respiration in hamster brown adipocytes.

The action of endogeneous adenosine on isolated hamster brown adipocytes was examined. Adenosine production from brown adipocytes was measured after labeling of the intracellular nucleotide pool with [3H]adenine. Accumulation of [3H]adenosine in the incubation medium was maximum after 5 min of incubation and was still present after 20 min. When adenosine accumulation was prevented by addition of adenosine deaminase, the stimulatory effects of isoproterenol on oxygen uptake, lipolysis, and adenosine 3',5'-cyclic monophosphate (cAMP) generation were enhanced. However, basal rates of lipolysis an

Regulation and physiological role of cyanide-resistant oxidases in fungi and plants.

Data on the induction and regulation of cyanide-resistant oxidases in eucaryotic microorganisms and higher plants are reviewed. Expression of an alternative oxidase gene can be caused by a decrease in energy charge in cells. Some evidence exists suggesting that cAMP and Ca2+ act as intracellular signals inducing the expression of the alternative oxidase genes. Under certain conditions cells produce alternative oxidases which remain in an inactive state. Activation of the alternative pathway of cell respiration is usually observed when electron transport via cytochromes is inhibited. The physio…

Statins Reduce Intratumor Cholesterol Affecting Adrenocortical Cancer Growth.

Mitotane causes hypercholesterolemia in patients with adrenocortical carcinoma (ACC). We suppose that cholesterol increases within the tumor and can be used to activate proliferative pathways. In this study, we used statins to decrease intratumor cholesterol and investigated the effects on ACC growth related to estrogen receptor α (ERα) action at the nuclear and mitochondrial levels. We first used microarray to investigate mitotane effect on genes involved in cholesterol homeostasis and evaluated their relationship with patients' survival in ACC TCGA. We then blocked cholesterol synthesis with

Oxidative metabolism drives inflammation-induced platinum resistance in human ovarian cancer.

Tumour cells have long been considered defective in mitochondrial respiration and mostly dependent on glycolytic metabolism. However, this assumption is currently challenged by several lines of evidence in a growing number of tumours. Ovarian cancer (OC) is one of the most lethal cancers worldwide, but it continues to be a poorly understood disease and its metabolic features are far to be elucidated. In this context, we investigated the role of tumour necrosis factor receptor-associated protein 1 (TRAP1), which is found upregulated in several cancer types and is a key modulator of tumour cell

Maintenance of cellular acidification in cyanide-treated hepatocytes results from inhibition of Na+/H+ exchange.

Inhibition of respiration by metabolic inhibitors or hypoxia is accompanied by intracellular acidification. Although this acidification is known to promote cell survival during hypoxia, little is known about its mechanism. Given that the Na+/H+ exchanger is known to be a major component of pH regulation in normal hepatocytes, the aim of this study was to determine the effects of inhibition of mitochondrial respiration on intracellular pH (pHi) regulation and Na+/H+ exchange. Cyanide (CN-; 5 mM) plus fructose (20 mM) were used as a model of hypoxic acidosis. pHi was measured with quantitative f

Toxic concentrations of glutamate induce ferroptosis without affecting mitochondrial respiration in rodent brain slice cultures.

Many neuronal disorders are characterized by glutamate toxicity, which is closely associated with mitochondrial and neuronal dysfunction. The aim of this study was to investigate the link between mitochondrial function and mechanisms of cell death induced by glutamate. The study was carried out on male (C57BL/6), male Sprague Dawley acute brain slice (ABSs), male and female organotypic brain slice (BSCs). Mitochondrial respiration, activities of tricarboxylic acid (TCA) cycle enzymes, and the cell death rate (assessed by lactate dehydrogenase (LDH) release and histological analyses) were deter

Nur77 serves as a molecular brake of the metabolic switch during T cell activation to restrict autoimmunity.

T cells critically depend on reprogramming of metabolic signatures to meet the bioenergetic demands during activation and clonal expansion. Here we identify the transcription factor Nur77 as a cell-intrinsic modulator of T cell activation. Nur77-deficient T cells are highly proliferative, and lack of Nur77 is associated with enhanced T cell activation and increased susceptibility for T cell-mediated inflammatory diseases, such as CNS autoimmunity, allergic contact dermatitis and collagen-induced arthritis. Importantly, Nur77 serves as key regulator of energy metabolism in T cells, restricting

Tumor acidosis: Fusing mitochondrial dynamics and lipid metabolism.

Cancer cells experience multiple stresses within tumors, stemming from elevated metabolic activity, including nutrient shortage, waste buildup, hypoxia, and acidosis. According to Groessl et al.,1 acidosis is the dominant environmental factor offering metabolic flexibility to support tumor fitness and resilience to the other stresses by promoting mitochondria fusion and enhancing respiration capacity.

Exposure to maternal obesity programs sex differences in pancreatic islets of the offspring in mice.

AIMS/HYPOTHESIS: Obesity during pregnancy increases offspring type 2 diabetes risk. Given that nearly half of women of child-bearing age in many populations are currently overweight/obese, it is key that we improve our understanding of the impact of the in utero/early life environment on offspring islet function. Whilst a number of experimental studies have examined the effect of maternal obesity on offspring islet architecture and/or function, it has not previously been delineated whether these changes are independent of other confounding risk factors such as obesity, postnatal high-fat-feedi

The potential of Gol-e-Gohar iron ore mine airborne dust to induce toxicity in human lung A549 cells.

Airborne particulates in iron ore mining are a risk factor for adverse human lung effects. In this study, fine particulates deposited on surfaces of about 1.5 m above the ground and 6 meters from a milling unit of the Gol-e-Gohar iron ore mine were collected through wipe sampling. Dust particles less than 5 µm in diameter were separated with an electronic sieve. Aliquots were prepared from the sieved iron ore dust estimated to be equivalent to respiratory exposure in the iron ore mill in the concentrations of 1, 5, 10, 50, 100, and 250 µg/mL, which were intended to represent equivalent inhaled

The accumulation of assembly intermediates of the mitochondrial complex I matrix arm is reduced by limiting glucose uptake in a neuronal-like model of MELAS syndrome.

Ketogenic diet (KD) which combined carbohydrate restriction and the addition of ketone bodies has emerged as an alternative metabolic intervention used as an anticonvulsant therapy or to treat different types of neurological or mitochondrial disorders including MELAS syndrome. MELAS syndrome is a severe mitochondrial disease mainly due to the m.3243A > G mitochondrial DNA mutation. The broad success of KD is due to multiple beneficial mechanisms with distinct effects of very low carbohydrates and ketones. To evaluate the metabolic part of carbohydrate restriction, transmitochondrial neuronal-l

Glutathione-dependent metabolism of trichloroethylene in isolated liver and kidney cells of rats and its role in mitochondrial and cellular toxicity.

Metabolism of trichloroethylene (Tri) by the glutathione (GSH) conjugation pathway was studied in hepatocytes, renal cortical cells, and liver subcellular fractions from male Fischer 344 rats. Toxicity of Tri and some of its GSH- and cytochrome P450-dependent metabolites was then studied in isolated hepatocytes, kidney cells, and liver and kidney mitochondria to assess the relative role of these metabolites in toxicity. Tri metabolism to S-(1,2-dichlorovinyl)glutathione (DCVG), a nephrotoxic metabolite of Tri, was demonstrated in both isolated hepatocytes and kidney cells. This suggests that t

The anticancer properties of the nanocomposite of low-nanoscale Ag nanoparticles obtained in SiO2-grafted polyacrylamide carrier.

BACKGROUND/AIMS: Silver-based nanomedicine is an ongoing trend in anticancer research. Polymers greatly improve the stability of bare metal silver and improve its interaction with cells. We aimed to evaluate the anticancer efficiency of the synthesized Ag/polymer nanocomposite against B95-8 and Wish cell lines. METHODS: A highly stable low-nanoscale Ag/silica-grafted polyacrylamide nanocomposite (Ag/Hyb) with a mean particle size of 6 nm, estimated by TEM, was obtained. Malignant cell lines: B95-8 (Burkitt lymphoma), Wish (human amniotic with marker chromosomes of HeLa), and the control MDCK c

Morphologic effects of a sulfur(IV) aerosol on the nasal cavity of beagle dogs.

Morphologic changes were observed in nasal cavities of beagle dogs after long-term exposure to a respirable sulfur(IV) aerosol at a concentration equivalent to a sulfur dioxide (SO2) concentration of 0.6 mg/m3. The changes were characterized by a thickened epithelial layer resulting from epithelial proliferation, by a loss of secretory material, and by moderate mononuclear cell infiltration.

Anatomy of the pleura.

The pleura is a monolayer of mesothelial cells covering the lung and inner surface of the chest cavity, creating the pleural space. The mesothelial cells rest on a matrix of collagen, elastic fibers, blood vessels, and lymphatics, which allow the lung and chest to expand and contract, protected from friction by the pleural fluid and properties of the mesothelial cells. With a rich blood supply and lymphatic system just deep to the mesothelial layer, the pleura is a dynamic layer protecting the lung and pleural cavity from infection while transmitting the forces of respiration without damage to

[Role of cyclic nucleotides in carotid chemoreception].

The activation of the metabolic systems of rat carotid body under the influence of alkaloids and acids was studied by spectrofluorimetric technique. It has been demonstrated the growth of the cAMR levels being suggested to the role of messenger between chemical stimuli and the flows of glomus cells metabolism. Chemical substances of other types (such as acetylcholine, NaCl saccharose) don't provide any changes of cell respiration and concentration of cAMP in carotid cells. The data obtained provide the evidence of heterogeneity of glomus cella reception mechanisms of the qualitatively differen…

Correction of hypoxemia and hypoglycemia restores muscle mitochondrial respiration and remodels mitochondrial proteome in growth-restricted sheep fetuses.

Placental insufficiency causes fetal hypoxemia and hypoglycemia and is a major driver of fetal growth restriction (FGR). In FGR skeletal muscle, mitochondrial respiration is reduced, partially due to altered mitochondrial protein abundance. We have shown that maternal oxygen and fetal glucose supplementation alleviate fetal hypoxemia and hypoglycemia and improve skeletal muscle satellite cell proliferation. However, its effects on muscle mitochondrial respiratory function and proteomic profiles remain unknown. Here, we tested whether correcting fetal hypoxemia and hypoglycemia restores mitocho

Mitochondrial bioenergetics boost macrophage activation, promoting liver regeneration in metabolically compromised animals.

BACKGROUND AND AIMS: Hepatic ischemia-reperfusion injury (IRI) is the leading cause of early posttransplantation organ failure as mitochondrial respiration and ATP production are affected. A shortage of donors has extended liver donor criteria, including aged or steatotic livers, which are more susceptible to IRI. Given the lack of an effective treatment and the extensive transplantation waitlist, we aimed at characterizing the effects of an accelerated mitochondrial activity by silencing methylation-controlled J protein (MCJ) in three preclinical models of IRI and liver regeneration, focusing
