Ultrasonographic assessment of fetal well-being during late gestation: development of an equine biophysical profile.

Mares with complicated pregnancies (illness, problems at parturition or delivery of an abnormal foal, n = 30) were scanned transabdominally from 298 days gestation to term in order to measure fetal size, evaluate fetal well-being and characterise the intrauterine environment. The results of the last scan obtained prior to parturition were compared to normal data obtained from fetuses of comparable gestational age to develop a biophysical profile specific for the equine fetus. Twelve mares produced a normal foal (positive outcome) and 18 mares delivered 19 abnormal foals (negative outcome). Bot

Estimation of the number of hematopoietic precursor cells during fetal mouse development by covariance analysis.

Differentiation of hematopoietic precursor cells results in the formation of clonally related descendent cells. Using the mosaic expression of beta-galactosidase in female mouse fetuses heterozygous for an X-linked lacZ transgene, we analyzed the clonal relationship of the hematopoietic progeny. The proportion of beta-galactosidase positive cells for different T- and B-lymphoid and myeloid cell populations was determined at different stages of fetal development. We found excellent correlations of the proportion of beta-galactosidase expressing cells for all hematopoietic lineages confirming th…

Endocrine and body growth traits in heifers exposed to testosterone-propionate during early fetal development.

This study was conducted to determine the effects of testosterone-propionate exposure during fetal development on sexual differentiation and growth rates in heifers. Ten pregnant cows were given subcutaneous injections of testosterone-propionate (250 mg/injection) every other day during d 40 to 60 of gestation. Four cows aborted after the end of testosterone treatment, while four heifers (androgenized females) and two bulls (androgenized males) were produced from the six remaining pregnant, testosterone-propionate treated cows. Calves from cows that did not receive exogenous hormone treatment …

Histological observations on thymic development in fetal and newborn mammals subject to intrauterine growth retardation.

Fetal growth retardation resulting from maternal dietary protein deprivation (rats) or Coxsackie virus B3 infection in mid-pregnancy (mice) was associated with delayed thymic development. Histological examination revealed that less darkly staining small lymphocytes were present and that the thymus was not well organised into cortex and medulla as seen in the normal-weight fetuses. In preterm or neonatal piglets, the histological pattern of the thymus did not vary appreciably according to the body weight of the fetus. Hassall's corpuscles were present and the tissues were well organised into co

Maternal microbiota and fetal brain development: a multidisciplinary review of cognitive, neurological, and metabolic outcomes.

OBJECTIVE: The prenatal period represents a critical window for the programming of the central nervous system. This multidisciplinary narrative review aims to synthesize current evidence regarding the maternal microbiota-gut-fetal brain axis, examining how maternal gut composition may influence fetal neurogenesis, synaptogenesis, and long-term neurobehavioural outcomes. MECHANISM: We analyse three primary pathways of influence: the systemic translocation of neuroactive bacterial metabolites (specifically short-chain fatty acids), the modulation of the maternal immune system leading to materna

Stage-, dose- and course-dependent disruption of mice fetal ovarian development following prenatal dexamethasone exposure.

BACKGROUND: Dexamethasone, a synthetic glucocorticoid, is widely used during pregnancy to prevent preterm infant respiratory distress syndrome. While our prior work established that multiple courses of prenatal dexamethasone exposure (PDE) impair offspring ovarian function with transgenerational effects, the specific impacts of exposure stages, doses, and courses remain poorly defined. MATERIALS AND METHODS: Pregnant Kunming mice were treated with dexamethasone during mid-late gestation (GD14-15) or late gestation (GD16-17) with low/moderate/high doses, either as a single course or as two con

Impact of Maternal Smoking During Pregnancy on Fetal and Neonatal Neurobehavioral Development: A Narrative Review.

BACKGROUND: Maternal smoking during pregnancy (MSDP) remains a common source of prenatal drug exposure worldwide. MSDP has been shown to increase fetal health risks and interfere with fetal development. Although newborns prenatally exposed to other substances (e.g., opiates, benzodiazepines) have been shown to have increased stress and withdrawal symptoms, few studies have explored the effect of MSDP on the fetal and neonatal periods. METHODS: We performed a comprehensive search for empirical research articles investigating the effects of MSDP on fetal and neonatal neurobehavior and signs of …

Single-cell resolution uncovers neighboring cell subtypes that share steroidogenic capacity during fetal testis development.

Historically, endocrine cells were perceived to coordinate their output in a uniform manner. Recently however, single-cell technologies have uncovered heterogeneity within these populations, indicating that individual cells may operate as independently regulated units. Using high-resolution tools such as single-molecule fluorescent in situ hybridization (sm-FISH) and single-cell RNA sequencing (scRNA-seq), we investigated the contributions of individual and the collective of fetal Leydig cells to androgen production over time during mouse testis development. Temporal profiles of intratesticula

Thyroid Health and Selenium: The Critical Role of Adequate Intake from Fetal Development to Adolescence.

The thyroid gland plays a crucial role in regulating metabolism and supporting development through the production of the hormones T4 and T3. These hormones are essential during childhood for nervous system myelination, physical growth, puberty, skeletal and dental maturation, and overall metabolic balance. In early infancy, when the hypothalamic-pituitary-thyroid axis is still immature, thyroid dysfunction can result in a range of long-term complications. The metabolism and action of thyroid hormones depend not only on iodine but also on other vital micronutrients, particularly selenium (Se).

The gonadal niche safeguards human fetal germline cell development following maternal SARS-CoV-2 infection.

During pregnancy, germline development is vital for maintaining the continuation of species. Recent studies have shown increased pregnancy risks in COVID-19 patients at the perinatal stage. However, the potential consequence of infection for reproductive quality in developing fetuses remains unclear. Here, we analyze the transcriptome and DNA methylome of the fetal germline following maternal severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. We find that infection at early gestational age, a critical period of human primordial germ cell specification and epigenetic reprog

Impact of luteoma during pregnancy on fetal development and its long-term effects into adulthood.

A woman in her 30s presents to the Differences in Sexual Development Programme at a tertiary care academic medical centre with vaginal stenosis and scarring. Her medical history is significant for virilisation in utero due to a maternal luteoma of pregnancy. Laboratory investigations at the time of birth showed elevated androgens in both mother and daughter. During infancy, she underwent clitoroplasty and vaginoplasty for correction of posterior vaginal fusion. She represented as an adult with vaginal stenosis, with associated physical and psychosocial implications. She was not able to insert

Dose- and stage-dependent toxic effects of prenatal prednisone exposure on fetal articular cartilage development.

Prednisone is frequently used to treat rheumatoid diseases in pregnant women because of its high degree of safety. Whether prenatal prednisone exposure (PPE) negatively impacts fetal articular cartilage development is unclear. In this study, we simulated a clinical prednisone treatment regimen to examine the effects of different timings and doses of PPE on cartilage development in female and male fetal mice. Prednisone doses (0.25, 0.5, and 1 mg/kg/d) was administered to Kunming mice at different gestational stages (0-9 gestational days, GD0-9), mid-late gestation (GD10-18), or during the enti

Default mode network functional connectivity strength in utero and the association with fetal subcortical development.

The default mode network is essential for higher-order cognitive processes and is composed of an extensive network of functional and structural connections. Early in fetal life, the default mode network shows strong connectivity with other functional networks; however, the association with structural development is not well understood. In this study, resting-state functional magnetic resonance imaging and anatomical images were acquired in 30 pregnant women with singleton pregnancies. Participants completed 1 or 2 MR imaging sessions, on average 3 weeks apart (43 data sets), between 28- and 39

Speed and navigation control of thymocyte development by the fetal T-cell gene regulatory network.

T-cell differentiation is a tightly regulated developmental program governed by interactions between transcription factors (TFs) and chromatin landscapes and affected by signals received from the thymic stroma. This process is marked by a series of checkpoints: T-lineage commitment, T-cell receptor (TCR)β selection, and positive and negative selection. Dynamically changing combinations of TFs drive differentiation along the T-lineage trajectory, through mechanisms that have been most extensively dissected in adult mouse T-lineage cells. However, fetal T-cell development differs from adult in w
