Transcriptome Analysis Reveals the Gene Expression Changes in the Silkworm (Bombyx mori) in Response to Hydrogen Sulfide Exposure.

Hydrogen sulfide (H2S) has been recognized for its beneficial influence on physiological alterations. The development (body weight) and economic characteristics (cocoon weight, cocoon shell ratio, and cocoon shell weight) of silkworms were increased after continuous 7.5 µM H2S treatment. In the present study, gene expression changes in the fat body of silkworms at the 5th instar larvae in response to the H2S were investigated through comparative transcriptome analysis. Moreover, the expression pattern of significant differentially expressed genes (DEGs) at the 5th instar larvae was confirmed b

Bio-Scaffolds as Cell or Exosome Carriers for Nerve Injury Repair.

Central and peripheral nerve injuries can lead to permanent paralysis and organ dysfunction. In recent years, many cell and exosome implantation techniques have been developed in an attempt to restore function after nerve injury with promising but generally unsatisfactory clinical results. Clinical outcome may be enhanced by bio-scaffolds specifically fabricated to provide the appropriate three-dimensional (3D) conduit, growth-permissive substrate, and trophic factor support required for cell survival and regeneration. In rodents, these scaffolds have been shown to promote axonal regrowth and

Marginal sealing around integral bilayer scaffolds for repairing osteochondral defects based on photocurable silk hydrogels.

Osteochondral repair remains a major challenge in current clinical practice despite significant advances in tissue engineering. In particular, the lateral integration of neocartilage into surrounding native cartilage is a difficult and inadequately addressed problem that determines the success of tissue repair. Here, a novel design of an integral bilayer scaffold combined with a photocurable silk sealant for osteochondral repair is reported. First, we fabricated a bilayer silk scaffold with a cartilage layer resembling native cartilage in surface morphology and mechanical strength and a BMP-2-

Non-invasive monitoring of in vivo hydrogel degradation and cartilage regeneration by multiparametric MR imaging.

Numerous biodegradable hydrogels for cartilage regeneration have been widely used in the field of tissue engineering. However, to non-invasively monitor hydrogel degradation and efficiently evaluate cartilage restoration in situ is still challenging. Methods: A ultrasmall superparamagnetic iron oxide (USPIO)-labeled cellulose nanocrystal (CNC)/silk fibroin (SF)-blended hydrogel system was developed to monitor hydrogel degradation during cartilage regeneration. The physicochemical characterization and biocompatibility of the hydrogel were evaluated in vitro. The in vivo hydrogel degradation and

Anterior cruciate ligament reconstruction in a rabbit model using silk-collagen scaffold and comparison with autograft.

The objective of the present study was to perform an in vivo assessment of a novel silk-collagen scaffold for anterior cruciate ligament (ACL) reconstruction. First, a silk-collagen scaffold was fabricated by combining sericin-extracted knitted silk fibroin mesh and type I collagen to mimic the components of the ligament. Scaffolds were electron-beam sterilized and rolled up to replace the ACL in 20 rabbits in the scaffold group, and autologous semitendinosus tendons were used to reconstruct the ACL in the autograft control group. At 4 and 16 weeks after surgery, grafts were retrieved and anal

Skin repair and infection control in diabetic, obese mice using bioactive laser-activated sealants.

Conventional wound approximation devices, including sutures, staples, and glues, are widely used but risk of wound dehiscence, local infection, and scarring can be exacerbated in these approaches, including in diabetic and obese individuals. This study reports the efficacy and quality of tissue repair upon photothermal sealing of full-thickness incisional skin wounds using silk fibroin-based laser-activated sealants (LASEs) containing copper chloride salt (Cu-LASE) or silver nanoprisms (AgNPr-LASE), which absorb and convert near-infrared (NIR) laser energy to heat. LASE application results in

Harnessing wireless electrical stimulation and silk-based conductive hydrogels to boost iPSC-derived astrocytes neuroprotection and guide macrophage polarisation in vitro for spinal cord repair.

Spinal cord injury (SCI) remains a major clinical challenge due to its complex pathophysiology and lack of effective treatments. While electrical stimulation (ES) offers therapeutic potential for promoting neural repair, its clinical translation is limited by the invasiveness of conventional systems. Here, we report a non-invasive wireless electrical stimulation (WES) platform based on charge polarisation-induced capacitive coupling, enabled by a conductive silk fibroin/PEDOT:PSS (SF/PEDOT) hydrogel. SF/PEDOT hydrogels exhibit a tuneable mechanical stiffness (2-120 kPa), injectable delivery (<

Biomimetic SilMA patches with smart suction and macrophage immunoregulation for cascading skin regeneration.

Hydrogel-based biomaterials are promising for diabetic wound closure, facilitating exudate absorption and cell proliferation. However, conventional hydrogel dressings suffer from unsatisfactory adhesion performance. Insufficient interfacial adhesion easily leads to dressing shedding and treatment interruption. By contrast, excessive adhesion induced by chemically bonded adhesive groups may trigger secondary wound damage during dressing replacement. Moreover, macrophage immunoregulation fails to induce its pro-healing phenotype, posing a challenge for diabetic skin regeneration. Inspired by the

Recent concepts of autoimmune pancreatitis and IgG4-related disease.

Recent studies suggested the existence of two subtypes of autoimmune pancreatitis (AIP): type 1 related with IgG4 (lymphoplasmacytic sclerosing pancreatitis; LPSP) and type 2 related with a granulocytic epithelial lesion (idiopathic duct-centric chronic pancreatitis; IDCP). Apart from type 2 AIP, the pathological features of type 1 AIP with increased serum IgG4/IgE levels, abundant infiltration of IgG4+ plasmacytes and lymphocytes, fibrosis, and steroid responsiveness are suggestive of abnormal immunity such as allergy or autoimmunity. Moreover, the patients with type 1 AIP often have extrapan

Recombinant Spider Silk Gels Derived from Aqueous-Organic Solvents as Depots for Drugs.

Hydrogels are widely used in various biomedical applications, as they cannot only serve as materials for biofabrication but also as depots for the administration of drugs. However, the possibilities of formulation of water-insoluble drugs in hydrogels are rather limited. Herein, we assembled recombinant spider silk gels using a new processing route with aqueous-organic co-solvents, and the properties of these gels could be controlled by the choice of the co-solvent. The presence of the organic co-solvent further enabled the incorporation of hydrophobic drugs as exemplarily shown for 6-mercapto

The effect of four types of artificial nerve graft structures on the repair of 10-mm rat sciatic nerve gap.

Investigating the effect of four types of artificial nerve graft (ANG) structures on rat sciatic nerve defect repair will aid future ANG designs. In this study, fibroin fibers and polylactic acid were used to prepare four ANGs with differing structures: nerve conduit with micron-sized pores (Conduit with pore group), nerve conduit without micron-sized pores (Conduit group), nerve scaffold comprising Conduit with pore group material plus silk fibers (Scaffold with pore group), and nerve scaffold comprising Conduit group material plus silk fibers (Scaffold group). Research on GM1 bound bMSCs loaded with SF hydrogel for spinal cord injury repair.

BACKGROUND: Monosialotetrahexosylganglioside (GM1), a major ganglioside component, is expressed in the spinal cords of mammals. Gangliosides have been shown to play important roles in various physiological processes, including neuronal protection, neurorepair, and stem cell differentiation. Bone marrow-derived mesenchymal stem cells (bMSCs) were expected to be a good source of cells for clinical transplantation and repair due to their abundance, easy accessibility and lack of ethical restrictions. They can also be induced to differentiate into neurons. However, there is still no ideal delivery

A smart bilayered scaffold supporting keratinocytes and muscle cells in micro/nano-scale for urethral reconstruction.

Rationale: In urethral tissue engineering, the currently available reconstructive procedures are insufficient due to a lack of appropriate scaffolds that would support the needs of various cell types. To address this problem, we developed a bilayer scaffold comprising a microporous network of silk fibroin (SF) and a nanoporous bacterial cellulose (BC) scaffold and evaluated its feasibility and potential for long-segment urethral regeneration in a dog model. Methods: The freeze-drying and self-assembling method was used to fabricate the bilayer scaffold by stationary cultivation G. xylinus usin

Effects of electrospinning and solution casting protocols on the secondary structure of a genetically engineered dragline spider silk analogue investigated via Fourier transform Raman spectroscopy.

Micrometer and submicrometer diameter fibers of recombinant dragline spider silk analogues, synthesized via protein engineering strategies, have been electrospun from 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) and compared with cast films via Raman spectroscopy in order to assess changes in protein conformation that may result from the electrospinning process. Although the solvent casting process was shown to result in predominantly beta-sheet conformation similar to that observed in the bulk, the electrospinning process causes a major change in conformation from beta-sheet to alpha-helix. Microstructural Anisotropy in Bone-Mimetic Silk Constructs Promotes Self-Mineralization.

Understanding how organic matrix architecture regulates biomineralization remains a central challenge in mineralized tissue regeneration. Emerging evidence highlights the importance of coupled phenomena, including nanoconfinement, ionic and pH regulation, and hierarchical structural remodeling, in directing mineral formation. Silk fibroin (SF) is a versatile structural protein widely used in biomaterials due to its tunable mechanics, scalable processing into micro- and nanoscale constructs, controllable biodegradation, and ability to support cell infiltration and tissue integration, including

Highly Deformable Organic Heterojunctions Empower Piezoelectric-Conductive Hydrogels for Functional Volumetric Muscle Regeneration.

Volumetric muscle loss (VML) results in irreversible muscle impairment, necessitating tissue-matched electrostimulation scaffolds for clinical repair. Here, we report a biointegrated hydrogel that leverages a piezoelectric-conductive organic heterojunction to deliver self-powered electrical cues directly to the injury site. The heterojunction comprises a piezoelectric metal-organic framework (MOF) coated with conductive polypyrrole (PPy), forming a stable p-n organic heterojunction with an intrinsic built-in electric field. Unlike inorganic piezoelectrics, the organic MOF-PPy features an ultra

A Sprayable Multifunctional Film Based on EGFR-Targeting DNA Nanoactivators for Burn Wound Healing.

Burn treatment remains a tough clinical hurdle, as traditional epidermal growth factor (EGF) treatments suffer from rapid proteolytic degradation and poor skin permeation. Herein, a sprayable, multifunctional film (STAR) based on EGFR-targeting DNA nanoactivators is reported for efficient burn wound healing. The STAR platform combines tannic acid (TA), silk fibroin (SF), and tetrahedral framework nucleic acids (tFNA) modified with tetravalent EGFR-stimulating aptamers. This design leverages the nanostructure of tFNA to realize multivalent cooperativity, inducing potent and sustained EGFR activ

Bioinspired silk-catechol-enzyme co-assemblies as jigsaw-designed conformation-locking bioactive-biocatalytic platforms.

Immobilized enzymes offer a sustainable, cost-effective alternative to soluble catalysis, but an adaptable platform preserving native conformation, stability, and broad enzyme/material compatibility remains elusive. We report a facile, general, jigsaw-designed platform via co-assembling silk fibroin (SF), protocatechualdehyde (PA), and enzymes. Mussel-inspired PA remodels SF into β-sheet-rich "nano-pockets" to conformationally lock bioenzymes, while its aldehyde and catechol moieties anchor enzymes and surfaces, yielding structurally and functionally adaptable, bioactive-biocatalytic self-asse
