Polycatechol-based iron predators disrupt fungal iron homeostasis to drive selective antifungal action.

Fungal infections pose a growing global health challenge, exacerbated by a scarcity of effective treatments and rising drug resistance. Although cationic polymers emerge as promising antifungal candidates owing to structural tunability, design flexibility, and resistance to proteolytic degradation, their clinical utility has been hampered by nonselective membrane-disruption mechanisms. Herein, we develop a class of polycatechols- termed fungal iron predators (FIPs), exhibit exceptional fungicidal activity and markedly low cytotoxicity. These FIPs can efficiently infiltrate fungal cells, select

Synthesis and antifungal activity of substituted 2,4,6-pyrimidinetrione carbaldehyde hydrazones.

Opportunistic fungal infections caused by the Candida spp. are the most common human fungal infections, often resulting in severe systemic infections-a significant cause of morbidity and mortality in at-risk populations. Azole antifungals remain the mainstay of antifungal treatment for candidiasis, however development of clinical resistance to azoles by Candida spp. limits the drugs' efficacy and highlights the need for discovery of novel therapeutics. Recently, it has been reported that simple hydrazone derivatives have the capability to potentiate antifungal activities in vitro. Similarly, p

Xyloglucan structure-dependent cell wall remodelling elicited by the exopolysaccharide of Hymenoscyphus fraxineus, the causal agent of ash dieback.

The plant cell wall is a dynamic matrix integrating mechanical, biochemical and immune signals during plant-microbe interactions. Although fungal exopolysaccharides (EPS) modulate these interactions, their structures and roles in cell wall remodelling remain unclear. Here we characterize the EPS secreted by Hymenoscyphus fraxineus, the causal agent of ash dieback, and demonstrate its biological activity in Fraxinus excelsior and Arabidopsis thaliana. Biochemical analyses revealed that the EPS is enriched in mixed linkage β-glucans (MLG). EPS exposure triggered substantial remodelling of cell w

Diorcinol L, a natural pentenyl diphenyl ether isolated from Aspergillus sp. MA6037, exhibits cytotoxic activity by inhibiting KIF11.

Fungi are important sources for the discovery and production of bioactive metabolites, many of which exhibit promising cytotoxicity against colorectal cancer (CRC). Among these, human gut-derived fungi have emerged as a valuable reservoir of natural products with cytotoxic properties. Diphenyl ethers (DEs) constitute a structurally diverse class of fungal secondary metabolites. In this study, a series of DEs, including the rarely reported prenylated diphenyl ethers (PDEs), were isolated and identified from the human gut-derived fungus Aspergillus sp. MA 6037. Using chromatographic and spectros

Global Stress Responses Identify the Functionally Divergent Regulators Required for Candida auris Commensalism and Pathogenicity.

Given its global distribution and high transmissibility in the environment, Candida auris poses a serious threat to global public health. However, the underlying mechanisms of its adaptive strategies remain poorly understood. Here we delineate the pan-genome structures of 1,306 representative C. auris isolates collected from 28 countries. In addition to the clade-related genetic diversity and highly variable pan-genomes, we identify the key regulatory modules and genes specific to C. auris in response to 32 different host microenvironment-mimicking stresses. Through comparative analysis with e

Fungal Glycosidases in Sporothrix Species and Candida albicans.

Glycoside hydrolases (GHs) are enzymes that participate in many biological processes of fungi and other organisms by hydrolyzing glycosidic linkages in glycosides. They play fundamental roles in the degradation of carbohydrates and the assembly of glycoproteins and are important subjects of studies in molecular biology and biochemistry. Based on amino acid sequence similarities and 3-dimensional structures in the carbohydrate-active enzyme (CAZy), they have been classified in 171 families. Members of some of these families also exhibit the activity of trans-glycosydase or glycosyl transferase

Significant tipping points of sediment microeukaryotes forewarn increasing antibiotic pollution.

Antibiotic pollution imposes urgent threats to public health and microbial-mediated ecological processes. Existing studies have primarily focused on bacterial responses to antibiotic pollution, but they ignored the microeukaryotic counterpart, though microeukaryotes are functionally important (e.g., predators and saprophytes) in microbial ecology. Herein, we explored how the assembly of sediment microeukaryotes was affected by increasing antibiotic pollution at the inlet (control) and across the outlet sites along a shrimp wastewater discharge channel. The structures of sediment microeukaryoti

Cell-wall dyes interfere with Cryptococcus neoformans melanin deposition.

Melanization is an intrinsic characteristic of many fungal species, but details of this process are poorly understood because melanins are notoriously difficult pigments to study. While studying the binding of cell-wall dyes, Eosin Y or Uvitex, to melanized and non-melanized Cryptococcus neoformans cells we noted that melanization leads to reduced fluorescence intensity, suggesting that melanin interfered with dye binding to the cell wall. The growth of C. neoformans in melanizing conditions with either of the cell-wall dyes resulted in an increase in supernatant-associated melanin, consistent

Leveraging the Attributes of Mucor hiemalis-Derived Silver Nanoparticles for a Synergistic Broad-Spectrum Antimicrobial Platform.

Driven by the need to engineer robust surface coatings for medical devices to prevent infection and sepsis, incorporation of nanoparticles has surfaced as a promising avenue to enhance non-fouling efficacy. Microbial synthesis of such nanoscale metallic structures is of substantive interest as this can offer an eco-friendly, cost-effective, and sustainable route for further development. Here we present a Mucor hiemalis-derived fungal route for synthesis of silver nanoparticles, which display significant antimicrobial properties when tested against six pathological bacterial strains (Klebsiella

[Wounds in the genital and oral region].

Wounds mean disruption of normal continuity of structures. Ulcers are excavations of the skin that reach at least to the depth of the dermis. More superficial defects are called erosions. Erosions and ulcerations are seen in a wide variety of conditions. Although many urogenital wounds have similar clinical appearances, their etiologies encompass many disorders. General categories of the causes of wounds are different types of trauma, viral, bacterial and fungal infections, neoplasms, autoimmune blistering diseases, immune and systemic diseases, and neoplasms. Many drugs are capable of causing

Biological function and molecular mapping of M antigen in yeast phase of Histoplasma capsulatum.

Histoplasmosis, due to the intracellular fungus Histoplasma capsulatum, can be diagnosed by demonstrating the presence of antibodies specific to the immunodominant M antigen. However, the role of this protein in the pathogenesis of histoplasmosis has not been elucidated. We sought to structurally and immunologically characterize the protein, determine yeast cell surface expression, and confirm catalase activity. A 3D-rendering of the M antigen by homology modeling revealed that the structures and domains closely resemble characterized fungal catalases. We generated monoclonal antibodies (mAbs)

Giant transposons promote strain heterogeneity in a major fungal pathogen.

UNLABELLED: Fungal infections are difficult to prevent and treat in large part due to strain heterogeneity which confounds diagnostic predictability. Yet the genetic mechanisms driving strain-to-strain variation remain poorly understood. Here, we determined the extent to which Starships-giant transposons capable of mobilizing numerous fungal genes-generate genetic and phenotypic variability in the opportunistic human pathogen Aspergillus fumigatus. We analyzed 519 diverse strains, including 11 newly sequenced with long-read technology and multiple isolates of the same reference strain, to reve

Antifungal activity of redox-active benzaldehydes that target cellular antioxidation.

BACKGROUND: Disruption of cellular antioxidation systems should be an effective method for control of fungal pathogens. Such disruption can be achieved with redox-active compounds. Natural phenolic compounds can serve as potent redox cyclers that inhibit microbial growth through destabilization of cellular redox homeostasis and/or antioxidation systems. The aim of this study was to identify benzaldehydes that disrupt the fungal antioxidation system. These compounds could then function as chemosensitizing agents in concert with conventional drugs or fungicides to improve antifungal efficacy. M

Characterization of active site variants of xanthine hydroxylase from Aspergillus nidulans.

Xanthine/alpha-ketoglutarate (alphaKG) dioxygenase (XanA) is a non-heme mononuclear Fe(II) enzyme that decarboxylates alphaKG to succinate and CO(2) while hydroxylating xanthine to generate uric acid. In the absence of a XanA crystal structure, a homology model was used to target several putative active site residues for mutagenesis. Wild-type XanA and ten enzyme variants were purified from recombinant Escherichia coli cells and characterized. The H149A and D151A variants were inactive and the H340A variant exhibited only 0.17% of the wild-type enzyme activity, consistent with the proposed rol

The skin as a reservoir for Candidozyma auris: colonization dynamics, host defense, and decolonization challenges.

Candidozyma auris has emerged as one of the most formidable fungal pathogens of the modern era, designated a critical priority pathogen by the World Health Organization due to its multidrug resistance, thermotolerance, and capacity to cause prolonged outbreaks in healthcare settings. Unlike most fungal pathogens, C. auris exhibits a remarkable tropism for human skin, where it persists asymptomatically, forms biofilm-like structures resistant to conventional antiseptics, and serves as a reservoir for nosocomial transmission. Invasive infections, particularly candidemia, carry mortality rates of
