Gut Microbiota Serves as a Crucial Independent Biomarker in Inflammatory Bowel Disease (IBD).

Inflammatory bowel disease (IBD), encompassing Crohn's disease (CD), ulcerative colitis (UC), and IBD unclassified (IBD-U), is a complex intestinal disorder influenced by genetic, environmental, and microbial factors. Recent evidence highlights the gut microbiota as a pivotal biomarker and modulator in IBD pathogenesis. Dysbiosis, characterized by reduced microbial diversity and altered composition, is a hallmark of IBD. A consistent decrease in anti-inflammatory bacteria, such as Faecalibacterium prausnitzii, and an increase in pro-inflammatory species, including Escherichia coli, have been o

The Role of Viral and Bacterial Infections in the Etiology of Behçet's Disease.

BACKGROUND: Behçet's disease (BD) is a complex systemic vasculitis with a poorly understood etiology that involves genetic, environmental, and immunological factors. Increasing evidence suggests that viral and bacterial infections may trigger or exacerbate BD through immune-mediated pathways. This review aims to clarify how different infectious agents may contribute to BD pathogenesis. METHODS: For this review, articles addressing microbial involvement in BD were collected from established databases such as PubMed, Scopus, and Web of Science. Priority was given to studies evaluating classical

Impact of Physical Exercise on Gut Microbiome, Inflammation, and the Pathobiology of Metabolic Disorders.

BACKGROUND: The gastrointestinal tract (GIT) harbors a complex and diverse microbial composition that outnumbers our own body cells and their gene contents. These microbes play a significant role in host metabolism and energy homeostasis. Emerging evidence suggests that the GIT microbiome significantly contributes to host health and that impairments in the microbiome may cause the development of metabolic diseases. The microbiome architecture is shaped by several genetic and environmental factors, including nutrition and physical activity. Physical exercise has preventive or therapeutic effect

Efficient insertional mutagenesis system for the dimorphic pathogenic fungus Sporothrix schenckii using Agrobacterium tumefaciens.

Sporothrix schenckii is a dimorphic pathogenic fungus that causes human and animal sporotrichosis globally. Here we developed and optimized an Agrobacterium tumefaciens-mediated transformation (ATMT) system of S. schenckii for insertional mutagenesis. The transformation efficiency reached more than 600 transformants per 10(6) conidia. Using this protocol enabled us to obtain a large number of T-DNA insertional mutants within a short experimental period. Several mutants with altered phenotypes were obtained during the transformation experiments. The mutants displayed mitotic stability. Transfer

Gut microbiota in poultry: Roles in nutrition, metabolism and production performance.

The gut microbiota plays an important role in nutrient utilization, intestinal homeostasis, immune regulation, and production performance in poultry. Its composition and functional activity are shaped by complex interactions among host species, developmental stage, intestinal segment, diet, genetic background, and rearing environment. This review summarizes current knowledge of the poultry gut microbiota, with particular emphasis on its roles in nutrition, metabolism, and production performance. We first examine microbial composition and spatial distribution across major poultry species, inclu

Intestinal microbiota profiles of captive-bred cynomolgus macaques reveal influence of biogeography and age.

BACKGROUND: Age-associated changes to the intestinal microbiome may be linked to inflammageing and the development of age-related chronic diseases. Cynomolgus macaques, a common animal model in biomedical research, have strong genetic physiological similarities to humans and may serve as beneficial models for the effect of age on the human microbiome. However, age-associated changes to their intestinal microbiome have previously only been investigated in faecal samples. Here, we have characterised and investigated the effects of age in the cynomolgus macaque intestinal tract in luminal samples

Microecological characteristics of water bodies/sediments and microbial remediation strategies after 50 years of pollution exposure in ammunition destruction sites in China.

The effects of long-term ammunition pollution on microecological characteristics were analyzed to formulate microbial remediation strategies. Specifically, the response of enzyme systems, N/O stable isotopes, ion networks, and microbial community structure/function levels were analyzed in long-term (50 years) ammunition-contaminated water/sediments from a contamination site, and a compound bacterial agent capable of efficiently degrading trinitrotoluene (TNT) while tolerating many heavy metals was selected to remediate the ammunition-contaminated soil. The basic physical and chemical propertie

Antibiotic-induced microbial depletion enhances murine small intestinal epithelial growth in a serotonin-dependent manner.

Regulation of small intestinal epithelial growth by endogenous and environmental factors is critical for intestinal homeostasis and recovery from insults. Depletion of the intestinal microbiome increases epithelial proliferation in small intestinal crypts, similar to the effects observed in animal models of serotonin potentiation. Based on prior evidence that the microbiome modulates serotonin activity, we hypothesized that microbial depletion-induced epithelial proliferation is dependent on host serotonin activity. A mouse model of antibiotic-induced microbial depletion (AIMD) was employed. S

The hologenomic stress legacy: A framework for soil as a transgenerational memory bank of plant immunity.

Plants pass transgenerational inheritance through genetic and epigenetic information; however, the soil's role as the host environment in this memory transfer remains largely unexplored. Research within this field has largely been domain-specific, with only 117 publications in 24 years integrating fields of soil persistent metabolites (soil chemistry), stress-induced microbial consortia (soil microbiology), and plant molecular science (plant epigenetics). In this perspective article, we synthesize evidence and propose a testable hypothesis, the Hologenomic Stress Legacy (HSL) hypothesis. It st

Host Range Breadth Correlates with Genic Diversity in Honeybee Phages.

Bacteriophages can evolve rapidly. Mutation and recombination via horizontal gene transfer allow them to counter adaptive responses by microbial hosts. However, little is known about the genomic processes underlying phage evolution within an ecological context-especially within natural microbial communities. This is due in part to the difficulty in resolving aspects of phage ecology, such as host range. To better understand the interplay of phage ecology and evolution within natural microbial communities, we combined measures of phage host range in vivo with measures of genome evolution in ord

Systemic Interventions Targeting Epidermal Barrier Function: Translational Insights from Oral Probiotics.

The epidermal barrier is increasingly recognized as a dynamic and pharmacologically targetable system that extends beyond its traditional role as a passive structural defense. In inflammatory skin diseases such as atopic dermatitis and psoriasis, barrier dysfunction contributes not only to increased permeability and environmental susceptibility but also to complex bidirectional interactions involving immune dysregulation, microbial imbalance, oxidative stress, and systemic inflammation. Emerging evidence further supports the concept of the gut-skin axis as a systemic regulatory network linking

Adaptive modification of antiviral defense systems in microbial community under Cr-induced stress.

BACKGROUND: The prokaryotic antiviral defense systems are crucial for mediating prokaryote-virus interactions that influence microbiome functioning and evolutionary dynamics. Despite the prevalence and significance of prokaryotic antiviral defense systems, their responses to abiotic stress and ecological consequences remain poorly understood in soil ecosystems. We established microcosm systems with varying concentrations of hexavalent chromium (Cr(VI)) to investigate the adaptive modifications of prokaryotic antiviral defense systems under abiotic stress. RESULTS: Utilizing hybrid metagenomic

Investigative modalities in infectious keratitis.

Standard recommended guidelines for diagnosis of infectious keratitis do exist. Based on an extensive Medline literature search, the various investigative modalities available for aiding the diagnosis of microbial keratitis have been reviewed and described briefly. Preferred practice patterns have been outlined and the importance of routine pre-treatment cultures in the primary management of infectious keratitis has been highlighted. Corneal scraping, tear samples and corneal biopsy are few of the specimens needed to carry out the investigative procedures for diagnosis and for initiating thera

The challenges of defining the human nasopharyngeal resistome.

The nasopharynx is an important microbial reservoir for the emergence and spread of antibiotic-resistant organisms. The nasopharyngeal resistome is an extensive, adaptable reservoir of antibiotic-resistance genes (ARGs) within this niche. Metagenomic sequencing decodes the genetic material of all organisms within a sample using next-generation technologies, permitting unbiased discovery of novel ARGs and associated mobile genetic elements (MGEs). The challenges of sequencing a low-biomass bacterial sample have limited exploration of the nasopharyngeal resistome. Here, we explore the current un

Extended-spectrum beta-lactamases in clinical isolates of enterobacteria in Mexico.

Resistance to extended-spectrum cephalosporins within members of the family Enterobacteriaceae occurs virtually world-wide. Nevertheless, nothing was known about this problem among isolates from Mexico. To address this issue, we studied oximino-cephalosporin resistant isolates of Klebsiella pneumoniae (13), Escherichia coli (7), and Enterobacter cloacae (23) recovered from patients in Mexico City hospitals during 1990 to 1992. In the presence of clavulanic acid, these strains increased susceptibility to cefotaxime and ceftazidime (MIC90 64 and >256 microg/ml, respectively). The ability of thes

The oral microbiota in oral squamous cell carcinoma: unravelling mechanisms and clinical potential.

Originating in the mucosal lining of the mouth, oral squamous cell carcinoma is the most common malignancy of the head and neck regions. Its pathogenesis is multifactorial, involving environmental exposures, genetic susceptibility, and lifestyle-related risk factors. Increasing evidence indicates that oral microbial dysbiosis contributes to the initiation and progression of OSCC. Under healthy conditions, the oral cavity harbors a diverse and functionally balanced microbial ecosystem that maintains mucosal integrity, supports immune homeostasis, and prevents colonization by pathogenic species.

Brief Report: Dialister as a Microbial Marker of Disease Activity in Spondyloarthritis.

OBJECTIVE: Dysbiosis of the intestinal microbiota has been widely established in inflammatory bowel disease (IBD). There is significant clinical and genetic overlap between spondyloarthritis (SpA) and IBD, and up to 50% of all patients with SpA exhibit microscopic signs of bowel inflammation, often bearing particular resemblance to early Crohn's disease, a subtype of IBD. This study was undertaken to assess the relationship between intestinal microbial composition, gut histology, and disease activity markers in SpA. METHODS: Gene analysis by 16S ribosomal RNA amplicon sequencing was used to c

A Conditional Plasmid System for Markerless Gene Deletion in Genetically Recalcitrant Fusobacterium nucleatum subsp. animalis.

Fusobacterium nucleatum subsp. animalis (FNA) plays a prominent role in oral microbial ecology and is increasingly implicated in systemic diseases such as colorectal cancer. However, its genetic intractability has significantly hindered functional studies to understand its pathogenic mechanisms. To address this challenge, we developed a conditional plasmid system that enables efficient, markerless gene deletion in FNA strains. This system features inducible control of plasmid replication via a theophylline-responsive riboswitch regulating repA, and counterselection via the MazF toxin under the
