Role for neurological and immunological resilience in the pathway of the aging muscle powerpenia: InCHIANTI study longitudinal results.

The neuromuscular junction shows several degenerative changes with aging, resulting in a reduction of transmission efficacy. These changes, paired with low-grade chronic inflammation, were considered triggers of the aging muscle processes. The main objective of this study is to assess the role of leukocyte count-derived ratios, nerve conduction velocity (NCV), and compound muscle action potential (CMAP) in determining time-dependent reduction in lower limb muscle explosive strength, a condition that has been defined as powerpenia. The InCHIANTI study enrolled a representative sample from the r

A multi-omic single-cell landscape of the aging mouse ovary.

The ovary is one of the first organs in humans to exhibit age-related functional impairments. As an organ composed of diverse heterogeneous cell types, the ovary exhibits cell-type-specific changes during the aging process, ultimately leading to a decline in female fertility. Investigating the molecular mechanisms of ovarian aging is crucial for understanding age-related fertility dysfunction in females. In this study, we combine scRNA-seq and scATAC-seq from mouse young/aged ovaries to characterize molecular features during ovarian aging. Using the single-cell multi-omic data, we revealed the

Circular RNAs as regulators and biomarkers of mammalian ovarian ageing.

Ageing is a prevalent characteristic of all cells, tissues, and organs; certain ones, such as the ovaries, undergo significant ageing and do so early in the lifespan of mammals. Despite the widely recognized hormonal regulation occurring in the ovaries, numerous mechanisms influencing ovarian ageing remain unclear. Recently, a long-overlooked consequence of splicing, small RNA molecules, characterized by their circular structure, have gained attention. Circular RNAs (circRNAs) have been identified as products of gene transcription and are categorized among other non-coding molecules. The focus

The road to a long lifespan in the Persian squirrel, a natural model for extended longevity: resisting free radical stress and healthy phospholipids.

Longevity is influenced by various factors, including fatty acid composition and free radical stress, which relate to the membrane pacemaker and rate of living hypotheses. While these aspects are well-documented in some long-lived species, they remain largely unexplored in tree squirrels. This study aimed to compare oxidative stress, antioxidant activity, nitrosative stress, and lipid composition between the long-lived Persian squirrel (Sciurus anomalus) and the short-lived Wistar rat across age cohorts (younger and older). Tissue homogenates from skin, liver, skeletal muscle, spleen, lung, an

Recent advances in treating age-related macular degeneration and diabetic retinopathy: Current therapies and emerging novel approaches.

The incidence of retinopathies is expected to increase globally as our population ages and diabetes becomes more prevalent. Neovascular age-related macular degeneration (nAMD) and proliferative diabetic retinopathy (PDR) are major causes of severe vision loss. These ophthalmological conditions are associated with neovascularization caused by excessive vascular endothelial growth factor (VEGF) secretion. This review focuses on the milestones achieved during the last few decades in treating retinopathies, presents a meta-analysis of key human clinical trials, and finally describes various novel

Reference values for handgrip strength in Europe: analysis of individual participant data from 27 countries.

We aimed to determine reference values for handgrip strength in Europe. Data from the Survey of Health, Ageing and Retirement in Europe were used, involving adults aged 50 + years. Reference values were expressed as weighted percentiles for absolute (kg) and relative (kg/m2) handgrip strength. For women, the highest absolute handgrip strength was observed among the 50-54-year-olds (standing: 5th percentile [P5] = 19 kg; 50th percentile [P50] = 29 kg; 95th percentile [P95] = 39 kg, sitting: P5 = 20 kg; P50 = 29 kg; P95 = 40 kg) and the lowest among the ≥ 90-year-olds (standing: P5 = 9 kg; P50 =

Association between blood nicotinamide adenine dinucleotide levels and blood laboratory parameters at baseline and after nicotinamide mononucleotide supplementation in middle-aged healthy individuals: post hoc analysis of a randomized, double-blinded, placebo-controlled clinical trial.

To assess associations between blood nicotinamide adenine dinucleotide (NAD) levels and laboratory parameters at baseline and after 60 days of nicotinamide mononucleotide (NMN) supplementation. Post hoc analysis of a randomized, double-blind, clinical trial of daily NMN (300, 600, or 900 mg) or placebo in healthy middle-aged participants. Among the 80 participants (49.4 ± 6.8 year), the baseline NAD was 7.21 [5.5, 10.6] nM. Every 1 nM higher baseline NAD level was associated with 0.24% (95%CI: 0.05-0.44) higher lymphocytes, - 0.36% (95% CI: - 0.57 to - 0.14) lower neutrophils, 0.023% (95% CI:

A scalable step count-based predictor of biological age: development and validation of MoveIt! Age in community-dwelling adults and geriatric rehabilitation inpatients.

Measuring biological age typically requires invasive and costly procedures. To address this, the MoveIt! Age Score was developed: a simple, scalable, and interpretable aging clock that predicts biological age using only wearable-derived steps data. MoveIt! Age was trained on steps data from the United States National Health and Nutrition Examination Survey (NHANES), using chronological age, maximum step count, and step count variability to predict PhenoAge, a blood biochemistry biological age score. MoveIt! Age performance was evaluated in two independent cohorts: Mitochondria and Muscle Healt

Glucagon receptor signaling is indispensable for the healthspan effects of caloric restriction in aging male mice.

Obesity and type 2 diabetes mellitus accelerate aging, shortening the duration of healthspan. Conversely, chronic calorie restriction (CR) extends healthspan. Research aimed at understanding the mechanism by which CR slows aging has focused heavily on insulin and downstream signaling cascades. Glucagon, a hormone that counter-regulates insulin, is commonly affected by these same interventions. To investigate the role of glucagon in aging, we used dietary manipulation, global and liver-specific glucagon receptor knockout, and pharmacological glucagon receptor activation. We found that globally

Age, glucose tolerance, and cognitive performance in female vervet monkeys.

This study assessed relationships between age, glucose and insulin metabolism, and cognitive performance in vervet monkeys (Chlorocebus aethiops sabaeus) a valuable model for aging research. Executive function, working memory, body mass index (BMI), and glucose and insulin responses during intravenous glucose tolerance tests (IVGTTs) were assessed in 41 middle-aged to older (9.1-29.5 years), socially housed, female vervets. Cluster analyses yielded four distinct IVGTT response patterns relevant to type 2 diabetes development: (I) normal insulin response and euglycemia; (II) baseline euglycemia

Blockade of A2AR improved brain perfusion and cognitive function in a mouse model of Alzheimer's disease.

Alzheimer's disease (AD) is a neurodegenerative disorder that affects more than 6.2 million Americans aged 65 and older, particularly women. Along with AD's main hallmarks (formation of β-amyloid plaques and tau neurofibrillary tangles), there are vascular alterations that occurs in AD pathology. Adenosine A2 receptor (A2AR) is one of the key factors of brain vascular autoregulation and is overexpressed in AD patients. Our previous findings suggest that protein arginine methyltransferase 4 (PRMT4) is overexpressed in AD, which leads to decrease in cerebral blood flow in aged female 3xTg mice.

A physically and mentally active lifestyle relates to younger brain and cognitive age.

Resistance to age-related pathological changes (brain maintenance), including Alzheimer's disease, cerebrovascular disease, and neurodegeneration may promote cognitive resilience in aging. However, how lifestyle and health profiles relate to successful cognitive and brain aging remains poorly understood. In a novel, deeply phenotyped cohort of 211 cognitively unimpaired older adults (age = 71.0 ± 7.4 years, 46% female), we characterized principal components of lifestyle and health using questionnaire, fitness, and blood data. We estimated cognitive age gap (CAG) based on comprehensive neuropsy

Subjective memory concern, negative affect, and cortical microstructural integrity in community-dwelling middle-aged men.

Concerns about memory often increase with age and have been suggested as a precursor to impending memory impairment or dementia. However, subjective memory concern (SMC) has also been shown to reflect an individual's trait-like tendency to worry about memory, which is more strongly linked to negative affect than to objective memory performance. Despite behavioral evidence supporting a trait-like dimension of SMC, its neuroanatomical underpinnings remain underexplored. In 477 community-dwelling dementia-free men (56-72 years old), we investigated the association between SMC and cortical mean di

Healthy diet and slower biological aging as protective factors against microvascular complications in type 2 diabetes.

This study aimed to evaluate the independent and joint effects of adherence to healthy dietary patterns and slower biological aging on the incidence of diabetic microvascular complications in individuals with type 2 diabetes mellitus (T2DM), and to assess the mediating role of biological aging. In a prospective cohort of 13,294 T2DM participants without baseline DMCs, dietary quality was assessed using a validated 10-point score, while biological aging was calculated from nine biomarkers and chronological age. Cox regression models were used to assess associations, and mediation analysis was p

Impacts of systemic milieu on cerebrovascular and brain aging: insights from heterochronic parabiosis, blood exchange, and plasma transfer experiments.

Aging is a complex biological process that detrimentally affects the brain and cerebrovascular system, contributing to the pathogenesis of age-related diseases like vascular cognitive impairment and dementia (VCID) and Alzheimer's disease (AD). While cell-autonomous mechanisms that occur within cells, independent of external signals from neighboring cells or systemic factors, account for some aspects of aging, they cannot explain the entire aging process. Non-autonomous, paracrine and endocrine, pathways also play a crucial role in orchestrating brain and vascular aging. The systemic milieu mo

Effects of electrical muscle stimulation on cognitive function and neuropathology in senescence-accelerated mouse (SAMP8) model of aging-associated cognitive decline.

The global increase in aging populations has heightened the urgency to develop effective interventions for age-related cognitive decline. Skeletal muscle has recently emerged as a potential modulator of brain health, particularly in the context of aging. This study investigates the effects of electrical muscle stimulation (EMS) on cognitive function and neuropathology in Senescence-Accelerated Mouse (SAMP8), a model of aging-associated cognitive decline. SAMP8 mice were divided into 3 groups: healthy controls (SAMR1), untreated SAMP8, and EMS-treated SAMP8. EMS was applied daily for 30 days, a

Exploring the power of MRI and clinical measures in predicting AD neuropathology.

Predicting Alzheimer's disease (AD) pathology prior to clinical diagnosis is important for identifying individuals at high risk of developing AD dementia. However, there remains a gap in leveraging MRI and clinical data to predict AD pathology. This study examines a novel machine learning approach that integrates the combined vascular (white matter hyperintensities, WMHs) and structural brain changes (gray matter, GM) with clinical factors (cognitive scores) to predict post-mortem neuropathology. Participants from the Alzheimer's Disease Neuroimaging Initiative dataset (ADNI) and National Alzh

Metabolic syndrome and risk of dementia and cognitive decline: a systematic review and meta-analysis of prospective cohort studies from 6,753,197 participants.

Previous findings on the link between metabolic syndrome (MetS) and the risk of all-cause dementia, Alzheimer's disease (AD), vascular dementia (VD), and cognitive decline are inconsistent. We systematically searched Embase, PubMed, Web of Science, the Cochrane Library, and Scopus up to June 2025 for prospective cohort studies conducted in community-based settings among adults aged 18 years or older that reported risk estimates (e.g., relative risks, hazard ratios, or odds ratios) for the association between MetS and the risk of dementia or cognitive decline. Risk of bias for studies were asse

Task-specific dynamical entropy variations in EEG as a biomarker for Parkinson's disease progression.

Uncovering the neuronal mechanisms un-derlying optimal behavioral performance is essential to understand how the brain dynamically adapts to changing conditions. In Parkinson's disease (PD), these neuronal mechanisms are disrupted and lead to impairments in motor coordination and higher-order cognitive functions. This study investigates neuronal dynamics during a lower-limb pedaling task by analyzing the dynamical entropy of EEG signals in healthy controls (HC), PD patients, and PD patients with freezing of gait (PDFOG). We examined both average entropy changes and entropy variability across t

Effects of aerobic exercise of different intensities on the social, emotional, and financial functioning of healthy older adults: results from a 16-week exercise randomized control trial.

Better social, emotional, and financial functioning are associated with improved health outcomes in older adults. While the literature supports a positive relationship between physical activity and increased functional ability in older adulthood, the intensity of physical activity necessary to achieve these gains remains uncertain. To address this gap, the current analysis uses data collected as part of a larger randomized controlled trial to examine the effects of a supervised exercise intervention on measures of social, emotional, and financial functioning.
