How Alligator Immune Peptides Kill Gram-Negative Bacteria: A Lipid-Scrambling, Squeezing, and Extracting Mechanism Revealed by Theoretical Simulations.

Alligator sinensis cathelicidins (As-CATHs) are antimicrobial peptides extracted from alligators that enable alligators to cope with diseases caused by bacterial infections. This study assessed the damaging effects of sequence-truncated and residue-substituted variants of As-CATH4, AS4-1, AS4-5, and AS4-9 (with decreasing charges but increasing hydrophobicity) on the membranes of Gram-negative bacteria at the molecular level by using coarse-grained molecular dynamics simulations. The simulations predicted that all the variants disrupt the structures of the inner membrane of Gram-negative bacte…

Antimicrobial Treatment Options for Difficult-to-Treat Resistant Gram-Negative Bacteria Causing Cystitis, Pyelonephritis, and Prostatitis: A Narrative Review.

Urinary tract infections, including cystitis, acute pyelonephritis, and prostatitis, are among the most common diagnoses prompting antibiotic prescribing. The rise in antimicrobial resistance over the past decades has led to the increasing challenge of urinary tract infections because of multidrug-resistant and "difficult-to-treat resistance" among Gram-negative bacteria. Recent advances in pharmacotherapy and medical microbiology are modernizing how these urinary tract infections are treated. Advances in pharmacotherapy have included not only the development and approval of novel antibiotics,…

Circumventing colistin resistance by combining colistin and antimicrobial peptides to kill colistin-resistant and multidrug-resistant Gram-negative bacteria.

OBJECTIVES: Colistin is a 'last-line' antibiotic used to treat multidrug-resistant Gram-negative bacteria, but colistin resistance has emerged. Colistin normally binds to the lipid A moiety on the bacterial outer membrane, where it then destroys the bacterial membrane. Mobilize colistin resistance (MCR, encoded by mcr-1 and others) is a phosphoethanolamine transferase that modifies lipid A, preventing colistin binding. We hypothesized that combining pore-forming AMPs and colistin will circumvent this mechanism and reduce the minimum inhibitory concentration (MIC) of colistin for both colistin-…

Extended Spectrum Beta-Lactamase-Producing Gram-Negative Bacteria Recovered From an Amazonian Lake Near the City of Belém, Brazil.

Aquatic systems have been described as antibiotic resistance reservoirs, where water may act as a vehicle for the spread of resistant bacteria and resistance genes. We evaluated the occurrence and diversity of third generation cephalosporin-resistant gram-negative bacteria in a lake in the Amazonia region. This water is used for human activities, including consumption after appropriate treatment. Eighteen samples were obtained from six sites in October 2014. Water quality parameters were generally within the legislation limits. Thirty-three bacterial isolates were identified as Escherichia (n …

Engineering multiple genomic deletions in Gram-negative bacteria: analysis of the multi-resistant antibiotic profile of Pseudomonas putida KT2440.

The genome of the soil bacterium Pseudomonas putida strain KT2440 has been erased of various determinants of resistance to antibiotics encoded in its extant chromosome. To this end, we employed a coherent genetic platform that allowed the precise deletion of multiple genomic segments in a large variety of Gram-negative bacteria including (but not limited to) P. putida. The method is based on the obligatory recombination between free-ended homologous DNA sequences that are released as linear fragments generated upon the cleavage of the chromosome with unique I-SceI sites, added to the segment o…

RND efflux pump inhibitor-like natural products as antibiotic adjuvants against multidrug-resistant gram-negative bacteria: a graded-evidence review.

Multidrug-resistant (MDR) Gram-negative bacteria-particularly carbapenem-resistant Enterobacterales, Pseudomonas aeruginosa and Acinetobacter baumannii-remain among the hardest infections to treat, and Resistance-Nodulation-Division (RND) efflux pumps are central to that resistance, making them a recurring target for antibiotic-adjuvant strategies. Natural products offer chemically diverse starting points for efflux-modulating adjuvants, but a fall in MIC is hard to attribute to pump inhibition: membrane permeabilisation, proton-motive-force (PMF) dissipation, efflux-gene downregulation, intri…

Design, Synthesis, and Biological Evaluation of Pyrimido[4,5-b]indole Derivatives with Potent Activity against Multidrug-Resistant Gram-Negative Bacteria.

The escalating crisis of antimicrobial resistance (AMR), particularly multidrug-resistant (MDR) Gram-negative bacterial infections, presents a grave threat to global public health. This urgent unmet medical need underscores the demand for novel antibiotics capable of overcoming existing resistance mechanisms. The ATP-binding sites of bacterial type II topoisomerases represent promising therapeutic targets against MDR Gram-negative pathogens. Pyrimido[4,5-b]indole-based compounds (GP-1) are the first reported ATPase inhibitors of DNA gyrase with potent and broad-spectrum antibacterial activity …

FIRL potentiates antibiotic activity against multidrug-resistant Gram-negative bacteria: Selective BamA-BamD interface perturbation induces functional outer membrane vulnerability.

BACKGROUND: The outer membrane (OM) of Gram-negative bacteria limits antibiotic entry and contributes substantially to multidrug resistance. OM integrity depends on β-barrel outer membrane protein (OMP) biogenesis mediated by the β-barrel assembly machinery (BAM) complex, making BAM an attractive target for antibiotic adjuvant development. OBJECTIVES: To define the molecular mechanism by which the previously described BamD-derived peptide FIRL functions as a BAM-targeting antibiotic adjuvant, with particular emphasis on its activity at the BamA-BamD interface. METHODS: Antibiotic potentiatio…
