Ingested microplastics and the gut-brain axis: A systematic review of mechanistic pathways and neurobiological impacts in human and preclinical models.

Miroplastic (MP) pollution which affects both humans and animals, has emerged as a global health concern. Evidence from in vivo rodent studies, in vitro human cell models, and multi-omics analyses indicates that ingested and inhaled MPs can accumulate in the gastrointestinal tract, cross epithelial barriers, and alter host-microbe interactions. Nanoplastics (NPs, <1 μm), owing to their small size and high surface-area-to-volume ratio, may exhibit greater cellular uptake, tissue translocation, and neurotoxic potential than larger MPs. These processes contribute to systemic effects, including im

Natural recreational waters and the risk that exposure to antibiotic resistant bacteria poses to human health.

Antimicrobial resistance (AMR) is widely recognised as a considerable threat to human health, wellbeing and prosperity. Many clinically important antibiotic resistance genes are understood to have originated in the natural environment. However, the complex interactions between humans, animals and the environment makes the health implications of environmental AMR difficult to quantify. This narrative review focuses on the current state of knowledge regarding antibiotic resistant bacteria (ARB) in natural bathing waters and implications for human health. It considers the latest research focusing

Functional Coupling of Human Microphysiology Systems: Intestine, Liver, Kidney Proximal Tubule, Blood-Brain Barrier and Skeletal Muscle.

Organ interactions resulting from drug, metabolite or xenobiotic transport between organs are key components of human metabolism that impact therapeutic action and toxic side effects. Preclinical animal testing often fails to predict adverse outcomes arising from sequential, multi-organ metabolism of drugs and xenobiotics. Human microphysiological systems (MPS) can model these interactions and are predicted to dramatically improve the efficiency of the drug development process. In this study, five human MPS models were evaluated for functional coupling, defined as the determination of organ in

A Proposed Oral-Liver-Pancreas Axis in Type 2 Diabetes: Linking Oral Dysbiosis, Hepatic Insulin Resistance, and Pancreatic Beta-Cell Stress.

Type 2 diabetes mellitus develops through the interaction of metabolic, inflammatory, and microbial processes that extend beyond the pancreas. Although the gut microbiota has received substantial attention, the oral microbiota may offer an additional, modifiable source of microbial and inflammatory signals. Periodontitis repeatedly exposes the circulation and gastrointestinal tract to bacteria, lipopolysaccharide, outer membrane vesicles, and host mediators. This narrative perspective asks whether oral dysbiosis may amplify pancreatic beta-cell stress through hepatic immunometabolic dysfunctio

Genomic factors that shape craniofacial outcome and neural crest vulnerability in FASD.

Prenatal alcohol exposure (PAE) causes distinctive facial characteristics in some pregnancies and not others; genetic factors may contribute to this differential vulnerability. Ethanol disrupts multiple events of neural crest development, including induction, survival, migration, and differentiation. Animal models and genomic approaches have substantially advanced our understanding of the mechanisms underlying these facial changes. PAE during gastrulation produces craniofacial changes corresponding with human fetal alcohol syndrome. These result because PAE reduces prechordal plate extension a

Neuroligin-3-deficient mice: model of a monogenic heritable form of autism with an olfactory deficit.

Autism spectrum disorder (ASD) is a frequent neurodevelopmental disorder characterized by variable clinical severity. Core symptoms are qualitatively impaired communication and social behavior, highly restricted interests and repetitive behaviors. Although recent work on genetic mutations in ASD has shed light on the pathophysiology of the disease, classifying it essentially as a synaptopathy, no treatments are available to date. To develop and test novel ASD treatment approaches, validated and informative animal models are required. Of particular interest, in this context are loss-of-function

The COVID-19 Vaccine Landscape.

The history of vaccine development spans centuries. At first, whole pathogens were used as vaccine agents, either inactivated or attenuated, to reduce virulence in humans. Safety and tolerability were increased by including only specific proteins as antigens and using cell culture methods, while novel vaccine strategies, like nucleic acid- or vector-based vaccines, hold high promise for the future. Vaccines have generally not been employed as the primary tools in outbreak response, but this might change since advances in medical technology in the last decades have made the concept of developin

Olive Oil Nutraceuticals in the Prevention and Management of Diabetes: From Molecules to Lifestyle.

Lifestyle is the primary prevention of diabetes, especially type-2 diabetes (T2D). Nutritional intake of olive oil (OO), the key Mediterranean diet component has been associated with the prevention and management of many chronic diseases including T2D. Several OO bioactive compounds such as monounsaturated fatty acids, and key biophenols including hydroxytyrosol and oleuropein, have been associated with preventing inflammation and cytokine-induced oxidative damage, glucose lowering, reducing carbohydrate absorption, and increasing insulin sensitivity and related gene expression. However, resea

Highly differentiated human airway epithelial cells: a model to study host cell-parasite interactions in pertussis.

BACKGROUND: Bordetella pertussis colonizes the human respiratory mucosa. Most studies on B. pertussis adherence have relied on cultured mammalian cells that lack key features present in differentiated human airway cells or on animal models that are not natural hosts of B. pertussis. The objectives of this work were to evaluate B. pertussis infection in highly differentiated human airway cells in vitro and to show the role of B. pertussis fimbriae in cell adherence. METHODS: Primary human airway epithelial (PHAE) cells from human bronchi and a human bronchial epithelial (HBE) cell line were gr

A human monoclonal IgG1 potently neutralizing the pro-inflammatory cytokine GM-CSF.

The pro-inflammatory cytokine GM-CSF is aberrantly produced in many autoimmune and chronic inflammatory human diseases. GM-CSF neutralization by antibodies has been shown to have a profound therapeutic effect in animal models of rheumatoid arthritis, inflammatory lung diseases, psoriasis and multiple sclerosis. Moreover, the absence of GM-CSF in null mutant mice ameliorates or prevents certain of these diseases. Here we describe the biophysical and biological properties of a human anti-GM-CSF IgG1 antibody designated MT203, which was derived by phage display guided selection. MT203 bound with

Interactions of human microglia cells with Japanese encephalitis virus.

BACKGROUND: Japanese encephalitis virus (JEV) is a neurotropic flavivirus causing mortality and morbidity in humans. Severe Japanese encephalitis cases display strong inflammatory responses in the central nervous system and an accumulation of viral particles in specific brain regions. Microglia cells are the unique brain-resident immune cell population with potent migratory functions and have been proposed to act as a viral reservoir for JEV. Animal models suggest that the targeting of microglia by JEV is partially responsible for inflammatory reactions in the brain. Nevertheless, the interact

Rapid quantitative immunohistochemical assessment of human peripheral neuropathies using a monoclonal antibody against nerve growth factor receptor.

An analysis of nerve growth factor (NGF) receptor expression and density in human sural nerve biopsies was performed by immunocytochemistry with a murine monoclonal antibody against the human NGF receptor. Quantitative assessment of immunostaining density was made by histospectrophotometry on frozen sections. Although there was enhanced expression of NGF receptor within endoneurium in all patients with clinical neuropathies, expression was highest in nerves with axonal disease, consistent with the proposal that disruption of axon-Schwann cell interactions triggers the re-expression of the NGF

Application of human liver cell models in drug development and toxicity studies: The role of membrane transporters.

The critical involvement of hepatic transporters in both physiological and pathological processes, including metabolic diseases, drug metabolism, and toxicity, represents a key issue in pharmacology. The expression and localization of transporters involved in hepatic functions are especially important in new drug development and toxicology studies. Because of major species differences in transporter expression, localization, and substrate recognition, the value of animal experiments is limited in this regard, and human liver cell preparations are already available to allow relevant in vitro st

Human-bat Competition on Cultivated Fruit Resources Promotes Bat-borne Pathogens Spillover to Humans and Domestic Animals in Bangladesh: An Exploratory Qualitative Study.

BACKGROUND: Nipah virus (NiV) outbreaks have recurred nearly annually in Bangladesh since 2001, with a fatality >70%. These outbreaks are primarily linked to the consumption of raw date palm sap (RDPS), which is contaminated by Indian flying fox bats. Due to the rapidly changing ecology, bats increasingly depend on cultivated fruits. To address this, we observed how humans and domestic animals compete and interact with bats for cultivated fruits facilitate pathogen transmission to humans and animals. METHODS: We conducted an exploratory qualitative study in 4 NiV outbreak districts of Banglad

The influence of various metals on magnesium concentration in cellular subfractions of certain rat tissues.

There are interactions between magnesium and other elements in human and animal organisms. Changes in homeostasis of this macro-element may be induced by intoxication of lead, chromium and the other metals. The purpose of our work was to analyze the influence of various metals (lithium, aluminium, lead and chromium) on magnesium concentrations of certain rat tissues. It was shown that the changes in magnesium concentration were dependent on the kind of metal, tissue and cellular subfraction.

The role of the microbiome in the neurobiology of social behaviour.

Microbes colonise all multicellular life, and the gut microbiome has been shown to influence a range of host physiological and behavioural phenotypes. One of the most intriguing and least understood of these influences lies in the domain of the microbiome's interactions with host social behaviour, with new evidence revealing that the gut microbiome makes important contributions to animal sociality. However, little is known about the biological processes through which the microbiome might influence host social behaviour. Here, we synthesise evidence of the gut microbiome's interactions with var

Insights on ADAMTS proteases and ADAMTS-like proteins from mammalian genetics.

The mammalian ADAMTS superfamily comprises 19 secreted metalloproteinases and 7 ADAMTS-like proteins, each the product of a distinct gene. Thus far, all appear to be relevant to extracellular matrix function or to cell-matrix interactions. Most ADAMTS functions first emerged from analysis of spontaneous human and animal mutations and genetically engineered animals. The clinical manifestations of Mendelian disorders resulting from mutations in ADAMTS2, ADAMTS10, ADAMTS13, ADAMTS17, ADAMTSL2 and ADAMTSL4 identified essential roles for each gene, but also suggested potential cooperative functions

Assessing the extent and public health impact of bat predation by domestic animals using data from a rabies passive surveillance program.

Domestic animals can serve as consequential conveyors of zoonotic pathogens across wildlife-human interfaces. Still, there has been little study on how different domestic species and their behaviors influence the zoonotic risk to humans. In this study, we examined patterns of bat encounters with domestic animals that resulted in submission for testing at the rabies laboratories of the Canadian Food Inspection Agency (CFIA) during 2014-2020. Our goals were specifically to examine how the number of bats submitted and the number of rabies positive bats varied by the type of domestic animal exposu

A systematic review on the effects of clozapine on B-lymphocytes and immunoglobulins.

BACKGROUND: Treatment-resistant schizophrenia (TRS) patients have been reported to exhibit distinct immunological characteristics compared with treatment-responsive patients, which have been hypothesised to relate to differential illness course or treatment response. Clozapine treatment has been associated with an increased risk of secondary antibody deficiency and a recent prospective observational study found reductions in immunoglobulin concentrations after clozapine initiation in individuals with treatment resistant schizophrenia. Researchers have hypothesised that hypogammaglobulinaemia m
