An Ultrasensitive Biosensing Platform Employing Acetylcholinesterase and Gold Nanoparticles.

Enzyme-linked immunosorbent assay (ELISA) is a well-known strategy for biomarker detection with a color change, which can be seen by the naked eyes. However, the moderate sensitivity of conventional ELISA limits its applications in many cases where the concentrations of biomarker are very low, such as cancer diagnosis. Here we describe an ultrasensitive colorimetric assay based on acetylcholinesterase (AChE)-catalyzed hydrolysis reaction, whose products trigger the aggregation of gold nanoparticles (AuNPs), causing a distinct color change of the solution from red to purple. This enhanced color

Abatacept: a T-cell co-stimulation modulator for the treatment of rheumatoid arthritis.

Rheumatoid arthritis (RA) is a debilitating autoimmune disease that has traditionally been treated with disease-modifying anti-rheumatic drugs (DMARDs). In the European Union (EU), patients who fail to respond to traditional DMARDs may receive tumor necrosis factor-alpha (TNF-alpha) antagonists. However, approximately one-third of patients fail TNF-alpha antagonists due to adverse effects or lack of efficacy, and there are limited treatment options available to these patients. As knowledge of the underlying immunopathology of RA evolves, new strategies for inhibiting the inflammatory process h

Radioimmunotherapeutic strategies in autologous hematopoietic stem-cell transplantation for malignant lymphoma.

High-dose therapy followed by autologous hematopoietic stem-cell transplantation is the preferred therapy for relapsed chemotherapy-sensitive aggressive non-Hodgkin lymphoma, and may play a role in the treatment of high-risk first-remission aggressive lymphomas, mantle-cell lymphomas and relapsed follicular lymphomas. The primary cause of failure of this approach is disease recurrence despite initial responses. Traditional high-dose regimens have relied upon myeloablative combinations of chemotherapy with or without total body irradiation. In the Western world, over 90% of lymphomas are of B-c

Prodrug and antedrug: two diametrical approaches in designing safer drugs.

The prodrug and antedrug concepts, which were developed to overcome the physical and pharmacological shortcomings of various therapeutic classes of agents, employ diametrically different metabolic transformations. The prodrug undergoes a predictable metabolic activation prior to exhibiting its pharmacological effects in a target tissue while the antedrug undergoes metabolic deactivation in the systemic circulation upon leaving a target tissue. An increased therapeutic index is the aspiration for both approaches in designing as well as evaluation criteria. The recent research endeavors of prodr

Antibody-drug conjugates in advanced small-cell lung cancer: a systematic review and single-arm meta-analysis of later-line monotherapy.

OBJECTIVE: Antibody-drug conjugates (ADCs) are being investigated in advanced small-cell lung cancer (SCLC), but the evidence is dominated by early-phase studies conducted in non-exchangeable treatment settings. We evaluated the activity and toxicity of ADCs while separating later-line monotherapy from frontline combination therapy, later-line combination therapy, and post-induction maintenance. METHODS: PubMed, Embase, the Cochrane Library, and Web of Science Core Collection were searched from inception and in 28 August 2026. HEV-Targeted Antibody-Drug Conjugate Promotes Long-Term Cardiac Allograft Acceptance.

BACKGROUND: The entrance of naive T cells into lymph nodes (LNs) is a crucial step for induction of heart transplant acceptance under costimulatory blockade. Specialized blood vessels within the LN known as high endothelial venules (HEVs) mediate this process. HEVs express a key glycoprotein containing 6-sulfo sialyl Lewis X, the binding site for L-selectin on the membrane of naive T cells. The proper formation of this carbohydrate requires sulfation by N-acetylglucosamine 6-O-sulfotransferases, particularly CHST4. Given the critical role of CHST4 in HEVs and transplant immunity, we aimed to d

Topoisomerase I inhibitor antibody-drug conjugates in breast cancer: the relevance of the DNA damage response to resistance, response, and combination treatment strategies.

INTRODUCTION: Antibody drug conjugates (ADCs) have revolutionized cancer treatment. ADCs that contain topoisomerase I inhibitors have been especially important in breast cancer treatment. Despite the substantial progress brought about by these ADCs, there remains a critical need to explore combination treatment strategies to overcome resistance and enhance the efficacy of these agents. Topoisomerase I inhibitors induce DNA damage and thus activate the DNA damage response (DDR). DDR elements have been examined in terms of their role in resistance and response to topoisomerase I inhibitors, and

HER2 ultralow in metastatic breast cancer: Diagnostic boundaries in the era of antibody-drug conjugates.

The introduction of antibody-drug conjugates (ADCs) such as trastuzumab deruxtecan (T-DXd) has challenged the traditional binary classification of HER2 in breast cancer. Evidence from the DESTINY-Breast04/06 and DAISY trials demonstrates clinically meaningful antitumour activity across a continuum of HER2 expression, including HER2-low and HER2-ultralow categories, while efficacy in HER2-null tumours remains uncertain. Despite this graded biological response, regulatory criteria remain categorical, requiring detectable membrane staining for treatment eligibility and excluding IHC 0 tumours. Pa

Research on precision treatment of pancreatic cancer targeted by antibody-drug conjugates.

Pancreatic cancer, and especially pancreatic ductal adenocarcinoma (PDAC), is extremely difficult to treat due to early asymptomatic stage, molecular heterogeneity, and resistance to conventional treatments, with a 5-year survival rate of less than 10%. Antibody-drug conjugates (ADCs), as an emerging precision therapy, show the potential to treat PDAC through the synergy of antibody targeting and cytotoxic drugs. Multiple targets (such as uPAR, Mesothelin, CLDN18.2, and TROP2) are highly expressed in PDAC, which has become the key direction of ADC development. However, the matrix barrier restr

Toxicity and efficacy of antibody-drug conjugates in advanced solid tumors: a retrospective single-center analysis of clinical trials.

BACKGROUND: Antibody-drug conjugates (ADCs) combine targeted monoclonal antibodies with cytotoxic payloads and are an emerging modality in systemic cancer therapy. Thirteen ADCs are Food and Drug Administration approved, with many more in development. However, design and use remain challenging, with issues including on/off-target toxicity, resistance from prior exposure to payload classes, and optimal target/payload selection. PATIENTS AND METHODS: This pooled analysis included patients treated on 19 clinical trials (9 phase I, 10 phase II/III) of 14 novel ADCs at Sarah Cannon Research Instit

[Expert consensus on diagnosis and treatment of advanced non-small cell lung cancer with HER-2 alterations (2025 edition)].

Mutations in the human epidermal growth factor receptor 2 (HER-2) gene are recognized as significant but relatively rare driver alterations in non-small cell lung cancer (NSCLC). These mutations predominantly manifest as gene mutation, amplification, and protein overexpression, with an estimated prevalence from 2.8% to 15.4% among NSCLC patients in China. Research indicates that HER-2 mutations, particularly exon 20 insertions (ex20ins), are strongly correlated with aggressive tumor biology, poor prognosis, and limited responsiveness to immunotherapy, thereby exhibiting characteristics of "col

Characteristics and utilization of the three BCMA-targeted therapies in multiple myeloma.

Recent advancements in the treatment of multiple myeloma have significantly improved patient outcomes, primarily due to the introduction of various therapeutic modalities. This review focuses on three BCMA-targeted therapies: anti-BCMA CAR-T cell therapy, bispecific antibody (BsAb) therapy, and antibody-drug conjugate (ADC) therapy. BCMA, a membrane-bound protein predominantly expressed on myeloma cells, presents a promising target due to its limited expression in normal tissues, minimizing off-target toxicity. We explore the characteristics, efficacy, and safety profiles of these therapies, h

Case Report: Possible association between dermatomyositis and Trastuzumab Deruxtecan therapy of triple-negative breast cancer patient.

INTRODUCTION: we present the case of the first documented occurrence of concurrent dermatomyositis and interstitial pneumonia associated with Trastuzumab Deruxtecan (T-DXd) therapy. We hypothesize that T-DXd likely induced an autoimmune response through tumor antigen release, resulting in multi-system involvement of skin, muscle, and pulmonary tissues. The shared pathogenesis of dermatomyositis and interstitial pneumonia involves aberrant activation of the type I interferon pathway, NF-κB signaling, and TGF-β cascade, collectively driving inflammatory and fibrotic processes. Notably, progressi

Comprehensive analysis of 177Lu-lilotomab satetraxetan in lymphoma cell lines: Implications for precision radioimmunotherapy and combination schemes.

177Lu-lilotomab satetraxetan (Betalutin) is an anti-CD37 radioimmunoconjugate evaluated as a single administration therapy for the treatment of patients with relapsed/refractory follicular lymphoma or diffuse large B-cell lymphoma (DLBCL). 177Lu-lilotomab satetraxetan treatment is well tolerated and shows consistent activity in most of the patients evaluated so far. Herein, we investigated the activity of 177Lu-lilotomab satetraxetan in a panel of 55 lymphoma cell lines of B- and T-cell origin. CD37-targeted radioimmunotherapy was more effective in CD37-positive B-cell lymphomas (n = 46) than

Antibody-Drug Conjugate Made of Zoledronic Acid and the Anti-CD30 Brentuximab-Vedotin Exert Anti-Lymphoma and Immunostimulating Effects.

Relevant advances have been made in the management of relapsed/refractory (r/r) Hodgkin Lymphomas (HL) with the use of the anti-CD30 antibody-drug conjugate (ADC) brentuximab-vedotin (Bre-Ved). Unfortunately, most patients eventually progress despite the excellent response rates and tolerability. In this report, we describe an ADC composed of the aminobisphosphonate zoledronic acid (ZA) conjugated to Bre-Ved by binding the free amino groups of this antibody with the phosphoric group of ZA. [Effectiveness and tolerability of sacituzumab govitecan in elderly patient with advanced triple negative breast cancer.].

The triple-negative breast carcinoma (TNBC), despite representing a minority of all breast carcinoma cases, poses a significant problem due to its biological aggressiveness and the resulting unfavorable prognosis for affected patients. Until recently, the only therapeutic strategy available was chemotherapy. However, new therapeutic approaches have recently led to an improvement in the natural history of this disease. Among these, the monoclonal antibody-drug conjugate sacituzumab govitecan has been shown to double the survival of the patients under examination. The use of this drug is associa

PLL-g-PEG Polymer Inhibits Antibody-Drug Conjugate Uptake into Human Corneal Epithelial Cells In Vitro.

Purpose: Antibody-drug conjugates (ADCs) are a relatively recent advance in the delivery of chemotherapeutics that improve targeting of cytotoxic agents. However, despite their antitumor activity, severe ocular adverse effects, including vision loss, have been reported for several ADCs. The nonspecific uptake of ADCs into human corneal epithelial cells (HCECs) and their precursors via macropinocytosis has been proposed to be the primary mechanism of ocular toxicity. In this study, we evaluated the ability of a novel polymer, poly(l-lysine)-graft-poly(ethylene glycol) (PLL-g-PEG), to decrease t

Prospective effect of linkers type on the anticancer activity of pemetrexed-monoclonal antibody (atezolizumab) conjugates.

BACKGROUND: Conventional chemotherapy results in severe toxic side effects due to affecting normal and cancer cells. The conjugation of chemotherapy with mAb will improve the chemotherapy selectivity towards cancer cells and at the same time will potentiate immune system to detect and kill cancer cells. The aim of the study was to prepare atezolizumab-pemetrexed conjugate using two types of linkers (linker conjugated with -NH2 of lysine amino acid in the mAb). METHODS: This study utilizes (for the first time) the mAb atezolizumab (AtZ) to prepare a new, selective conjugate carrier for pemetre
