Purification, crystallization and X-ray diffraction analysis of Trypanosoma congolense insect-stage surface antigen (TcCISSA).

Trypanosoma congolense is a major contributor to the vast socioeconomic devastation in sub-Saharan Africa caused by animal African trypanosomiasis. These protozoan parasites are transmitted between mammalian hosts by tsetse-fly vectors. A lack of understanding of the molecular basis of tsetse-trypanosome interactions stands as a barrier to the development of improved control strategies. Recently, a stage-specific T. congolense protein, T. congolense insect-stage surface antigen (TcCISSA), was identified that shows considerable sequence identity (>60%) to a previously identified T. brucei insec

A recombinant AeDNA containing the insect-specific toxin, BmK IT1, displayed an increasing pathogenicity on Aedes albopictus.

The Aedes aegypti densovirus (AeDNV) has previously shown potential in mosquito control. To improve its efficacy as a biopesticide, the gene for an excitatory insect-specific toxin from Buthus martensii Karsch (BmK IT1) was inserted into the AeDNV genome and cloned into pUCA plasmid. The coding sequence for green fluorescent protein was ligated to the C-terminus of the BmK IT1 gene as a screening marker. Recombinant and helper plasmids were cotransfected into C6/36 cells; wild-type viruses were the controls. The recombinant viruses were identified and quantified by real-time polymerase chain r

The salivary gland-specific apyrase of the mosquito Aedes aegypti is a member of the 5'-nucleotidase family.

The saliva of hematophagous insects contains a variety of pharmacologically active substances that counteract the normal hemostatic response to injury in vertebrate hosts. The yellow-fever mosquito, Aedes aegypti, secretes an apyrase that inhibits ADP-dependent platelet aggregation. Apyrase was purified as an active enzyme from adult female salivary glands and subjected to tryptic digestion, and the resulting peptides were sequenced. The amino acid sequences obtained match the conceptual translation product of a cDNA clone isolated from an adult female salivary gland library. Sequence comparis

The zig-zag model of molecular defense and counterdefense in plant-herbivore interactions.

Plant-herbivore interactions follow zig-zag-like cycles in which herbivore- and damage-derived cues activate conserved defense signaling hubs, while insect effectors suppress or redirect these pathways, creating recurrent molecular bottlenecks that shape plant resistance. Plant-herbivore interactions are orchestrated through a dynamic molecular dialogue at the feeding interface, wherein insect attack triggers the generation of damage-derived signals from plant tissues and herbivore-derived signals from oral or salivary secretions. These cues can activate plant defense by sensing the damage- an

Structure and expression of Rhodnius prolixus GH18 chitinases and chitinase-like proteins: Characterization of the physiological role of RpCht7, a gene from subgroup VIII, in vector fitness and reproduction.

Chitinases are enzymes responsible for the hydrolysis of glycosidic linkages within chitin chains. In insects, chitinases are typically members of the multigenic glycoside hydrolase family 18 (GH18). They participate in the relocation of chitin during development and molt, and in digestion in detritivores and predatory insects, and they control the peritrophic membrane thickness. Chitin metabolism is a promising target for developing vector control strategies, and knowledge of the roles of chitinases may reveal new targets and illuminate unique aspects of their physiology and interaction with

A Bacterium Derived from the Ovary of the Black Soldier Fly (Hermetia illucens) Attract Oviposition of the Host.

The black soldier fly, BSF (Hermetia illucens) has extensive applications in insect protein production and organic waste conversion, serving as a crucial resource insect. However, large-scale breeding faces challenges such as low adult mating rates, unstable oviposition, and inefficient egg collection, which significantly hinder the industrial application of BSF. In this study, we aimed to enhance the oviposition efficiency of BSF by utilizing the microbes within it. We isolated a strain of Serratia marcescens from the ovaries of the BSF, which can attract BSF to lay eggs by producing dimethyl

A cluster of candidate odorant receptors from the malaria vector mosquito, Anopheles gambiae.

Olfaction is critical to the host preference selection behavior of many disease-transmitting insects, including the mosquito Anopheles gambiae sensu stricto (hereafter A. gambiae), one of the major vectors for human malaria. In order to more fully understand the molecular biology of olfaction in this insect, we have previously identified several members member of a family of candidate odorant receptor proteins from A. gambiae (AgORs). Here we report the cloning and characterization of an additional AgOR gene, denoted as AgOr5, which shows significant similarity to putative odorant receptors in

Genome sequence of the endocellular obligate symbiont of tsetse flies, Wigglesworthia glossinidia.

Many insects that rely on a single food source throughout their developmental cycle harbor beneficial microbes that provide nutrients absent from their restricted diet. Tsetse flies, the vectors of African trypanosomes, feed exclusively on blood and rely on one such intracellular microbe for nutritional provisioning and fecundity. As a result of co-evolution with hosts over millions of years, these mutualists have lost the ability to survive outside the sheltered environment of their host insect cells. We present the complete annotated genome of Wigglesworthia glossinidia brevipalpis, which is

Characterization and Seasonal Expression Profiling of a Takeout/Juvenile Hormone-Binding Protein-Family Gene Associated With Diapause in Eurygaster integriceps.

Diapause in the Sunn pest, Eurygaster integriceps, is a key seasonal adaptation associated with migration, overwintering survival, and reproductive timing. However, the molecular components associated with this physiological state remain poorly characterized in this hemipteran pest. In this study, we identified and characterized a takeout-family transcript from E. integriceps (EiTO1; GenBank OP913374.1) and combined evolutionary, structural, and expression analyses to examine its association with seasonal physiological states. EiTO1 contained a 738-bp open reading frame encoding a 245-amino ac

Heat shock response during development of the malaria vector Anopheles stephensi (Culicidae: Diptera).

The pattern of synthesis of heat shock proteins (HSP) and thermotolerance to elevated temperatures during the development of the malaria vector Anopheles stephensi normally reared at 28 +/- 2 degrees C was studied using SDS-PAGE. In total twelve heat shock proteins (i.e. 31, 33, 38, 43, 44, 51, 57, 62, 69, 71, 113 and 121 kD were induced by heat shock during various stages of development. Eight polypeptides (HSP during one or other of the instars) appeared during normal development of the adult, which showed very little response towards heat shock. Only two polypeptides (57 and 69 kD) were ind

Vasodilatory properties of recombinant maxadilan.

Maxadilan is a peptide from the salivary gland of the sand fly Lutzomyia longipalpis, a vector for leishmaniasis. Cutaneous injection of femtomolar quantities of maxadilan produces long-lasting erythema, making it the most potent vasodilator known. Isolated rabbit thoracic and abdominal aorta, carotid artery, and iliac artery demonstrated dose-dependent arterial relaxation in response to maxadilan with a mean effective concentration (EC50) of 2.7 +/- 1.5, 2.1 +/- 0.5, 2.6 +/- 0.4, and 1.9 +/- 0.5 nM, respectively. Maxadilan proved to be at least sevenfold more potent than nitroglycerin in each

Immunogold detection of two neurohormones: the locust ovary maturing parsin and neuroparsin.

The cellular localization of two neurohormones of the locust pars intercerebralis-corpora cardiaca system: the ovary maturing parsin and neuroparsin, was investigated using electron microscopic immunocytochemistry (post-embedding immunogold labelling). The ovary maturing parsin and neuroparsin containing cells were first identified in semithin sections treated by combined histochemical- and immunostaining. The neuroparsin cells were paraldehyde fuchsin positive (A-type cells) and the ovary maturing parsin cells were paraldehyde fuchsin negative when semithin sections were stained with paraldeh

Wolbachia Endosymbionts Modify Drosophila Ovary Protein Levels in a Context-Dependent Manner.

UNLABELLED: Endosymbiosis is a unique form of interaction between organisms, with one organism dwelling inside the other. One of the most widespread endosymbionts is Wolbachia pipientis, a maternally transmitted bacterium carried by insects, crustaceans, mites, and filarial nematodes. Although candidate proteins that contribute to maternal transmission have been identified, the molecular basis for maternal Wolbachia transmission remains largely unknown. To investigate transmission-related processes in response to Wolbachia infection, ovarian proteomes were analyzed from Wolbachia-infected Dros

The consequences of deglycosylation of recombinant intra-melanosomal domain of human tyrosinase.

Tyrosinase, a melanosomal glycoenzyme, catalyzes initial steps of the melanin biosynthesis. While glycosylation was previously studied in vivo, we present three recombinant mutant variants of human tyrosinase, which were obtained using multiple site-directed mutagenesis, expressed in insect larvae, purified and characterized biochemically. The mutagenesis demonstrated the reduced protein expression and enzymatic activity due to possible loss of protein stability and protein degradation. However, the complete deglycosylation of asparagine residues in vitro, including the residue in position 371

Gustatory function of Aedes aegypti opsins and TRPA1 in avoiding a natural insect repellent.

The mosquito, Aedes aegypti, spreads viruses that cause dengue, yellow fever, and other devastating diseases. To identify candidate repellents that suppress blood feeding by Aedes aegypti, we conducted an in vitro screen of natural chemicals for activators of two visual opsins, Op1 and Op2, which we found are expressed at very high levels in gustatory organs. We found that Op1 and Op2 are chemical sensors since they are activated by flavonoids, including catechin. We devised an artificial membrane that mimics multiple properties of skin (VectaDerm), which we used on blood feeders, leading to t

Omics Tools Applied to the Study of Chagas Disease Vectors: Proteomics and Metabolomics.

Recent omics studies (cytogenomics, genomics, and transcriptomics) have provided extensive information on Chagas disease (CD) vectors. Here, we conduct a review of proteomic and metabolomic studies on these insect vectors. We address proteomic studies in triatomines, focusing on the identification and characterization of proteins in their saliva, midgut, and salivary glands, emphasizing their role in blood feeding by providing vasodilatory and anticoagulant substances, along with anti-inflammatory and antiplatelet activities. In addition, the review investigates protein diversity in different

Chagas disease vector blood meal sources identified by protein mass spectrometry.

Chagas disease is a complex vector borne parasitic disease involving blood feeding Triatominae (Hemiptera: Reduviidae) insects, also known as kissing bugs, and the vertebrates they feed on. This disease has tremendous impacts on millions of people and is a global health problem. The etiological agent of Chagas disease, Trypanosoma cruzi (Kinetoplastea: Trypanosomatida: Trypanosomatidae), is deposited on the mammalian host in the insect's feces during a blood meal, and enters the host's blood stream through mucous membranes or a break in the skin. Identifying the blood meal sources of triatomin

Silencing of a chemosensory membrane protein gene, the odorant receptor co-receptor (Orco), induces a significant disruption in olfactory-driven behaviors of the pumpkin fruit fly, Bactrocera tau (Walker) (Diptera: Tephritidae).

Bactrocera tau is an economically important invasive pest damaging many vegetables and fruits. Due to the hidden characteristics of its larval infestation, a strategy for regulating B. tau adults' olfactory-driven behaviors to control the pest is desirable. The olfactory co-receptor (Orco) is a key chemosensory membrane protein in insect olfactory systems. However, the function of Orco in the olfactory-driven behaviors of this pest remains under explored. In this study, we characterized the Orco gene from B. tau, named BtauOrco. The complete open reading frame (ORF) of BtauOrco is 1422 bp, whi

Two heat shock cognate 70 genes involved in spermatogenesis regulate the male fertility of Zeugodacus cucurbitae, as potential targets for pest control.

The melon fly Zeugodacus cucurbitae Coquillett (Diptera: Tephritidae) is an agricultural quarantine pest threatening fruit and vegetable production. Heat shock cognate 70 (Hsc70), which is a homolog of the heat shock protein 70 (Hsp70), was first discovered in mice testes and plays an important role in spermatogenesis. In this study, we identified and cloned five Hsc70 genes from melon fly, namely ZcHsc70_1/2/3/4/5. Phylogenetic analysis showed that these proteins are closely related to Hsc70s from other Diptera insects. Spatiotemporal expression analysis showed that ZcHsc70_1 and ZcHsc70_2 ar
