BACKGROUND: In Mvoua, a village situated in a forested area of Cameroon, recent studies have reported high prevalence of Plasmodium falciparum infection among the population. In order to understand factors that can sustain such a high malaria transmission, we investigated the biology of Anopheles vectors and its susceptibility to insecticides, as well as long-lasting insecticidal net (LLIN) coverage, use and bio-efficacy.
METHODS: A longitudinal entomological survey was conducted from July 2018 to April 2019. Adult mosquitoes were collected using the human landing catch (HLC) method and ident

In response to rising insecticide resistance among malaria vector species, countries are implementing long-lasting insecticide treated nets (LLINs) coformulated with piperonyl butoxide (PBO), which can restore susceptibility to pyrethroid-based nets. In mid-2021, Kenya National Malaria Program rolled out PBO-LLINs in Bungoma County. We evaluated the susceptibility of wild-collected vectors to new versus used PBO-LLINs using the WHO cone bioassays. Results were confirmed with the WHO susceptibility assay. A total of 2,020 female Anopheles were tested, of which 1,480 were wild-collected and 540 …

Limited studies have highlighted the importance of incorporating behavioural assessments into insecticide efficacy evaluations for wireworm pest control. "For this study, video tracking technology combined with a soil bioassay arena was employed to analyse the behaviour of Agriotes obscurus wireworms before, during, and after exposure to wheat seeds treated with the neonicotinoid insecticides thiamethoxam and imidacloprid at field-relevant concentrations. The analysis identified a set of behavioural key metrics for assessing the effects of these insecticides on wireworms. The results showed th

Equine pediculosis is a significant health and welfare issue, particularly in elderly and chronically debilitated animals. Currently infestation is controlled predominantly using topically applied pyrethroid insecticides, allowing limited scope for the rotation of drugs and increasing the risk of selection for resistance. Here the insecticidal efficacies of two pyrethroid-based products against the louse Bovicola (Werneckiella) ocellatus collected from donkeys were examined in vitro. The products were cypermethrin (Deosect™, Pfizer Ltd., 5% (w/v) cypermethrin, cutaneous spray) and permethrin (

BACKGROUND: Insecticide resistance among invasive tephritid fruit flies poses a great risk to national food security and has the potential to disrupt quarantine and eradication programs, which rely on the efficacy of Spinosad to prevent widespread establishment in North America. During 2022 to 2023 we surveyed the extent of Spinosad resistance of two key species, oriental fruit fly Bactrocera dorsalis, and melon fly Zeugodacus cucurbitae, from 20 sites across five Hawaiian Islands including Kaua'i, O'ahu, Maui, Molokai and the "Big Island" (Hawai'i).

BACKGROUND: Insect RDL (resistant to dieldrin) receptors are essential pentameric ligand-gated chloride channels that mediate the neuroinhibitory effect of GABA, the chief inhibitory neurotransmitter in the central nervous system. These receptors serve as primary targets for various insecticides, including noncompetitive antagonists (NCAs) such as cyclodiene organochlorines and phenylpyrazoles, as well as allosteric modulators like meta-diamides and isoxazolines. This study focuses on a newly discovered A2'N mutation within the RDL receptors, identified in fipronil-resistant planthoppers. Desp

BACKGROUND: Cyproflanilide is a novel chemical that is already undergoing insecticide registration in China and has been categorized as a member of group 30 by the IRAC. Since it was first detected in 2019, the fall armyworm (FAW), Spodoptera frugiperda, has become a serious pest in China. Our laboratory and field efficacy trials indicated that cyproflanilide exhibits high larvicidal activity against FAW. However, the effect of cyproflanilide against FAW remains unknown. And it is worth exploring further before the cyproflanilide becomes commercially available.
RESULTS: We found larvae expose

Although esterase-mediated spinosad resistance has been proposed for several insects, the associated molecular mechanism remains poorly understood. In this study, we investigated the mechanism of esterase-based spinosad resistance in house flies using a susceptible strain (SSS) and a spinosad-resistant, near-isogenic line (N-SRS). Combined with the synergistic effect of DEF on spinosad in the N-SRS strain, decreased ali-esterase activity in the spinosad-resistant strain has implicated the involvement of mutant esterase in spinosad resistance in house flies. Examination of the carboxylesterase

BACKGROUND: The clustered regularly interspaced short palindromic repeats (CRISPR)/Cas9 system provides some advantages over other reverse genetic techniques to investigate the causal relationship between insecticide resistance phenotype and candidate gene. Several studies published to date point to the nicotinic acetylcholine receptor (nAChR) α6 subunit strongly associated with spinosyns resistance in insects, including Plutella xylostella. However, reverse genetic verification of the P. xylostella nAChRα6 has not yet been achieved via an in vivo approach.
RESULTS: Here, we successfully cons…

Scabies has been diagnosed surprisingly frequently in Germany in recent years, and the use of acaricides has risen markedly. Present figures indicate an increase in the prevalence/incidence of scabies, but do not prove or quantify it for the following reasons: (a) scabies is not a notifiable disease in Germany; (b) the diagnosis is not always confirmed lege artis by means of light microscopy or dermatoscopy (which may lead to a comparatively high proportion of false‐positive diagnoses due to the low overall prevalence of scabies); (c) repeated treatments of the same patient and treatment of co

Temephos is a major organophosphate (OP) larvicide that has been used extensively for the control of Aedes albopictus and Aedes aegypti, the major vectors for viral diseases, such as dengue fever, zika and chikungunya. Resistance to temephos has been recently detected and associated with the upregulation of carboxylesterases (CCEs) through gene amplification, in both species. Here, we expressed the CCEae3a genes which showed the most striking up-regulation in resistant Aedes strains, using the baculovirus system. All CCEae3a variants encoded functional enzymes, with high activity and preferenc

Abamectin resistance was selected in the vegetable leafminer, Liriomyza sativae (Blanchard) (Diptera: Agromyzidae) under laboratory conditions, and cross-resistance patterns and possible resistance mechanisms in the abamectin-resistant strains (AL-R, AF-R) were investigated. Compared with the susceptible strain (SS), strain AL-R displayed 39-fold resistance to abamectin after 20 selection cycles during 25 generations, and strain AF-R exhibited 59-fold resistance to abamectin after 16 selection cycles during 22 generations. No cross-resistance to cyromazine was found in both abamectin-resistant

A southern house mosquito Culex quinquefasciatus Say colony was established from surviving late instars and pupae from a semifield evaluation on Natular XRG (a granular formulation containing 2.5% spinosad). The initial lethal levels ofNatular XRG against this colony were determined in the laboratory for the first-generation progeny (designated as F1). Selection pressure was applied at LC70-90 levels to 10,000-15,000 late third- and early fourth-instar larvae of each generation with Natular XRG. Susceptibility changes in response to selection were determined every other generation, where a gra

BACKGROUND: Long-lasting insecticidal nets (LLINs) have contributed to the reduction of malaria in sub-Saharan Africa, including Tanzania. However, they rely on daily user behaviour and high coverage which is difficult to maintain. Also, insecticide resistance among malaria vector mosquitoes is contributing to reduced efficacy of control tools. To overcome these problems, we propose to evaluate a new tool for house modification, the insecticide-treated eave nets (ITENs) in combination with insecticide-treated window screens (ITWS) incorporated with dual active ingredient (dual AI) for the con…

BACKGROUND: Understanding the molecular basis of insecticide resistance is key to improve the surveillance and monitoring of malaria vector populations under control. In the major malaria vector Anopheles funestus, little is currently known about the role of the knockdown resistance (kdr) mechanism. Here, we investigated the presence and contribution of knockdown resistance (kdr) to pyrethroids/DDT resistance observed in Anopheles funestus across Africa.
RESULTS: Pyrosequencing genotyping and sequencing of the voltage gated sodium channel (VGSC) gene did not detect the common L1014F mutation …
