Assessment of Immunological Response and Impacts on Fertility Following Intrauterine Vaccination Delivered to Swine in an Artificial Insemination Dose.

To protect the health of sows and gilts, significant investments are directed toward the development of vaccines against infectious agents that impact reproduction. We developed an intrauterine vaccine that can be delivered with semen during artificial insemination to induce mucosal immunity in the reproductive tract. An in vitro culture of uterine epithelial cells was used to select an adjuvant combination capable of recruiting antigen-presenting cells into the uterus. Adjuvant polyinosinic:polycytidylic acid (poly I:C), alone or in combination, induced expression of interferon gamma, tumor n

A review of simulation analyses of economics and genetics for the use of in-vitro produced embryos and artificial insemination in dairy herds.

The use of in-vitro produced (IVP) embryo transfer (ET) in dairy herds is growing fast. Much of this growth is on dairy farms where the focus is on milk production and not on selling breeding stock. The value of implementing IVP-ET in a dairy herd arises from a higher genetic merit of the IVP-embryo, but the cost to produce a pregnancy with an IVP embryo is greater than the cost of artificial insemination (AI). The first objective of this study was to review estimates of the net benefit of using IVP-ET over AI in dairy herds using existing literature. Another objective was to show how much IVP

Relationship between the Length of Sperm Tail Mitochondrial Sheath and Fertility Traits in Boars Used for Artificial Insemination.

The length of sperm tail midpiece, occupied by the mitochondrial sheath (MS), has been correlated with reproductive traits of mice, fish, and birds; however, it is not known whether such a correlation exists in higher order species such as domestic pigs. As the mitochondria provide for sperm motility and generate the fertility-affecting reactive oxygen species (ROS), we hypothesized that MS length correlates with boar semen parameters and artificial insemination (AI) fertility. Sperm samples collected from 57 boars and used for single sire AI were labeled with ProteoStat Aggresome probe (AGG;

Use of injectable progesterone and hCG for fixed-time artificial insemination during the non-breeding season in goats.

The objective of this study was to determine the estrous response and the moment of ovulation and fertility after a progesterone (P4) priming plus human chorionic gonadotropin (hCG) administration in multiparous and nulliparous goats. Therefore, two experiments were conducted during non-breeding season (April and May, 26° N) and all the animals received a single injection of 20 mg of P4 and 100 IU of hCG 24 h later. In Experiment 1, 13 multiparous and 9 nulliparous goats were subjected to estrus detection twice a day from P4 administration, and their ovaries were scanned by transrectal ultraso

Economic evaluation of artificial insemination of sex-sorted semen on a Brown Swiss dairy farm-A case study.

Artificial insemination using sex-sorted semen is employed to efficiently increase the number of female dairy calves born. Previous studies have determined that using sex-sorted semen is beneficial to improve the management, but the mechanism by which it increases cattle numbers through objective indices of breeding remains unclear. This study focused on a Brown Swiss cattle herd in which frozen female sex-sorted semen was systematically employed to increase the number of cattle. We analyzed the correlation between the increase in the number of cattle and the screening accuracy of sex-sorted s

Relationship of the conception rate and the side (left or right) of preovulatory follicle location at artificial insemination in dairy heifers.

In this study, we examined the locational effect (left or right ovary) of the preovulatory follicle (PF) on fertility in dairy heifers. In total, 1,111 artificial inseminations (AI) were analyzed. At AI, PF locations were examined using rectal palpation, and heifers were divided into two groups on their PF locations: (i) the PF located in the left ovary (L-PF); and (ii) the PF located in the right ovary (R-PF). Pregnancy was diagnosed by rectal palpation 60 days after AI. The conception rate was 50.7% in all heifers. Conception rate was significantly higher in the L-PF (60.1%) than in the R-PF

Puncture and drainage of the subordinate follicles at timed artificial insemination prevents the risk of twin pregnancy in dairy cows.

The objective of this study was to determine whether fluid drainage from the subordinate follicles by ovum pickup procedures prevents the risk of twin pregnancy without reducing the fertility of the cow. Lactating dairy cows with at least two follicles over 12 mm diameter located one on each ovary and selected from synchronized groups for fixed-time insemination were assigned to a Control (n = 49) or Drainage (n = 49) group. The largest follicle was considered as the dominant follicle. Draining of all subordinate follicles (≥10 mm) was performed in the Drainage group. All drained follicles dev

A comparison of timed artificial insemination and automated activity monitoring with hormone intervention in 3 commercial dairy herds.

The objective of this study was to compare the reproductive performance of cows inseminated based on automated activity monitoring with hormone intervention (AAM) to cows from the same herds inseminated using only an intensive timed artificial insemination (TAI) program. Cows (n=523) from 3 commercial dairy herds participated in this study. To be considered eligible for participation, cows must have been classified with a body condition score of at least 2.50, but no more than 3.50, passed a reproductive tract examination, and experienced no incidences of clinical, recorded metabolic diseases

Serum profile of cytokines interferon gamma and interleukin-10 in ewes subjected to artificial insemination by cervical retraction.

This study evaluated the influence of artificial insemination (AI) by cervical retraction (CRI) on serum levels of interferon gamma (IFNγ) and interleukin-10 (IL-10) in ewes. Synchronized pluriparous Santa Inês ewes were subjected to natural mating (NM, n = 8) and AI, which was performed for a fixed time (55 ± 1 hour) by CRI (n = 8) or laparoscopy (n = 8). Ewes were classified as pregnant, with return to estrus (RE) or with embryonic loss (EL). Blood samples were collected on Day 0, Day 3, Day 5, Day 12, and Day 17 (Day 0 = AI/NM) for progesterone dosage and cytokines were quantified from Day

Relationship between electrical resistance of cervical mucus and ovarian steroid concentration at the time of artificial insemination in ewes.

The purpose of this study was to investigate whether fertile or non-fertile inseminations (AI) in synchronized ewes are correlated with the electrical resistance of cervical mucus (ERCM) and the ovarian steroid concentration. AIs were performed either at fixed-time (group A) or after estrus detection (group B). Retrospective analysis revealed that at AI, pregnant ewes had lower ERCM values and progesterone concentrations than non-pregnant ones (p<0.05). It appears that ERCM may be used as an additional index for fertility enhancement of inseminated ewes.

A minimum number of motile spermatozoa are required for successful fertilisation through artificial intrauterine insemination with husband's spermatozoa.

Intrauterine insemination (IUI) is an effective, noninvasive, relatively simple and cheap method of infertility treatment. Many factors that affect IUI outcomes have been studied. However, there is no consensus about the optimal number of motile spermatozoa inseminated (NMSI) required for a reasonable chance of pregnancy after IUI. In this retrospective study, we aimed to assess the relationship between NMSI and the pregnancy rate after IUI with husband's spermatozoa. Couples who had either primary or secondary infertility for more than one year were recruited from the Department of Reproducti

Re-use of intravaginal progesterone devices associated with the Short-term Protocol for timed artificial insemination in goats.

Because intravaginal devices impregnated with 0.3 g of progesterone (i.e., CIDR-G) contain remaining hormone after their use in a Short-term Protocol (5 to 7 d of treatment), the reuse of these devices is proposed in goats. Two experiments were designed to study the effects of the reutilization of CIDR-G, establishing serum progesterone concentrations, follicular development, ovulatory response, and fertility. Experiment 1: Thirty dairy goats received a Short-term Protocol for 5 d using CIDR-G of first use (new devices, n = 10), second use (previously used for 5 d, n = 10), or third use (previ

Fixed-time artificial insemination with estradiol and progesterone for Bos indicus cows I: basis for development of protocols.

Five experiments were conducted on commercial farms in Brazil aiming to develop a fixed-time artificial insemination (TAI) protocol that achieved pregnancy rates between 40% and 55% in Bos indicus cows. These studies resulted in the development of the following protocol: insertion of an intravaginal device containing 1.9 g of progesterone (CIDR) plus 2.0mg im estradiol benzoate on Day 0; 12.5mg im dinoprost tromethamine on Day 7 in cycling cows or on Day 9 in anestrous cows; CIDR withdrawal plus 0.5mg im estradiol cypionate plus temporary calf removal on Day 9; TAI (48h after CIDR withdrawal)

The effect of progesterone and oestradiol benzoate on fertility of artificially inseminated repeat-breeder dairy cows during summer.

An experiment was conducted to examine the effect of progesterone (P(4)) and oestradiol benzoate (ODB) on fertility of repeat-breeder lactating dairy cows during summer. One hundred repeat-breeder lactating dairy cows were randomly allocated to four groups (Tr1, Tr2, Tr3 and C) in a study conducted at a private dairy farm. All cows were injected with 2 mg ODB (day 0), which were at random stages of their oestrous cycles. Cows in Tr1, Tr2 and Tr3 were administered with intravaginal progesterone-releasing devices (controlled internal drug-releasing, CIDR) at the time of ODB injection for 7 days

Effect of sire fertility and timing of artificial insemination in a Presynch + Ovsynch protocol on first-service pregnancy rates.

Sire fertility may influence pregnancy rate (PR) by differences in sperm survival in the female reproductive system and time required for capacitation and transport of sperm to site of fertilization. A predicted fertility index, Estimated Relative Conception Rate, was used to select 3 high-fertility artificial insemination (AI) sires (+3) and 3 average AI sires (-1). Ovulation can be predicted to occur at approximately 28 h following GnRH administration when used in an Ovsynch protocol. The objective of this study was to determine if AI at 2 times, 0 or 24 h after GnRH administration, in a Pre

Breeding soundness and libido examination of Belgian Blue and Holstein Friesian artificial insemination bulls in Belgium and The Netherlands.

Data on breeding soundness and libido evaluations in Belgian Blue (BB) bulls are scarce. The present study compared results of breeding soundness and libido evaluations of young BB bulls to young Holstein Friesian (HF) bulls prior to acceptance into an AI program. Breed differences for breeding soundness exist between BB and HF bulls, as 93.7% of the young BB bulls failed the breeding soundness examination (BSE) compared to 59.3% of the HF bulls (P=0.0005). Within the BB breed, differences were present between bulls of different ages, and bull selection for better fertility with increasing age
