An unusual adverse effect of interferon: hypertrichosis of the eyelashes.

Hypertrichosis of the eyelashes is a rare adverse effect of interferon-alpha treatment. We present a 21-year-old man with chronic renal failure and hepatitis B virus (HBV) infection who developed hypertrichosis of the eyelashes as a complication of IFN-alpha therapy. The patient was a candidate for living related renal transplantation and was given IFN-alpha 15 million units per week for HBV DNA positivity. After 6 months of therapy, HBV DNA positivity persisted, and the dose of IFN was increased to 30 million units per week. At the end of the first half of the second 6 months of therapy, the

Effect of early versus delayed postoperative injection of 5-fluorouracil plus interferon-alpha on colonic healing.

The purpose of this study was to determine whether delayed, postoperative, intraperitoneal treatment with 5-fluorouracil (5-FU) plus interferon-alpha-2a (IFN) has adverse effects on colonic healing, as does early treatment. Seventy male Wistar rats underwent colonic anastomoses. The rats were randomized to one of four groups. Early intraperitoneal injection was given to groups 1 and 2 which was repeated once daily for the first 3 postoperative days. Treatment was delayed in groups 3 and 4, from the 4th to the 7th postoperative day. A 0.9% NaCl solution was injected in the rats of control group

Clinical stable systemic mastocytosis with interferon alpha-2b therapy.

Systemic mastocytosis (SM) with progressive infiltration of various organs by mast cells is a rare disease with a mean survival of 2 years. So far there is no established therapy. We describe a patient with progressive mastocytosis who had a clinical response to interferon alpha-2b 3 x 5 Mio U s.c. three times a week. The response is stable 3 years after initiation of therapy and includes reduction of abdominal lymphadenopathy, ascites, decrease of serum and urinary histamine levels as well as pulmonary reticulonodular markings.

Acute renal failure in kidney transplant patients treated with interferon alpha 2b for chronic hepatitis C.

Sixteen kidney transplant (KT) patients (10 men, 6 women, aged 49 +/- 10 years) with chronic hepatitis C alpha-interferon (IFN-alpha) therapy (Intron A, Schering Plough) at a dose of 3 x 10(6) units subcutaneously 3 times a week. The treatment was scheduled for 24 consecutive weeks. Each patient had had stable renal function for at least 12 months prior to IFN-alpha therapy (mean serum creatinine, SCr, 121 +/- 38 mmol/l). Fourteen patients were receiving cyclosporin-A (CsA)-based immunosuppression and 2 patients were on conventional therapy. The patients' SCr was checked every 2 weeks while on

Interferon therapy for Behçet's disease.

BACKGROUND: Behçet's disease was first described in 1937 as a trisymptom complex. No standard therapy has been established. Its cause is presumed to be viral or immunologic. OBJECTIVE: We sought to determine whether patients with Behçet's disease benefit from interferon, which has antiviral, immunomodulatory, antiproliferative, and antitumoral properties. METHODS: Fourteen patients were initially given 3 MU interferon alfa-2a subcutaneously three times per week. The doses were gradually increased to 12 MU. Each patient received a total dose of 216 MU during the 2-month study. Therapy with 9

Recurrent clinical exacerbations of liver disease during the course of infection with hepatitis C virus.

Symptomatic clinical relapses during the course of chronic hepatitis C virus (HCV) infection are uncommon. Furthermore, acute liver dysfunction with elevated bilirubin during alpha-interferon therapy without other apparent coexisting diagnoses is rare. The case of a 31-yr-old man with three clinical exacerbations of HCV infection over an 18-month period is described. The third episode was characterized by rising serum aminotransferase levels on alpha-interferon therapy. The precise cause of this patient's flares is unknown. An immunologically mediated clearance of the hepatitis C virus, mutati

The in vitro and in vivo effect of recombinant interferon alpha-2a on circulating haemopoietic progenitors in polycythaemia vera.

In four patients with polycythaemia vera (PV) who received interferon alpha (IFN-alpha) (3 MU subcutaneously three times a week) for 5 months, peripheral blood levels of granulocyte-macrophage colony-forming units and erythroid burst-forming units were assessed monthly. Circulating progenitors significantly decreased throughout the treatment period. Moreover, we observed an inhibitory activity of IFN-alpha on haemopoietic progenitor cells (HPC) from patients with PV in vitro. The data presented confirm previous research which has shown an inhibitory effect of IFN-alpha on HPC from patients wit

Combined zidovudine and interferon-alpha treatment in patients with AIDS-associated Kaposi's sarcoma.

The effectiveness of addition of interferon-alpha (IFN-alpha) to zidovudine in patients with AIDS-associated Kaposi's sarcoma was assessed in a non-randomized, phase II clinical trial. Twenty-one patients were treated with oral zidovudine (600 mg daily) and IFN-alpha was increased to 18 MU daily for another 4 weeks. Only one of the 20 evaluable patients achieved a partial response at 8 weeks, that lasted for 3 months. Despite IFN-alpha dose escalation in six patients, no further responses were seen. While myelotoxicity was mild, fatigue was the dose-limiting side-effect that prevented dose esc

Intralesional interferon therapy for basal cell carcinoma.

In a clinical trial of 172 patients at four medical centers, interferon alfa-2b (1.5 x 10(6) IU) or a placebo was injected directly into biopsy-proved noduloulcerative or superficial basal cell carcinomas three times weekly for 3 weeks, for a cumulative dose of 13.5 million IU. Efficacy of treatment was determined at 16 to 20 weeks by examination of biopsy specimens that demonstrated cure of lesions in 86% of interferon-treated patients and in only 29% of placebo-treated patients. During the treatment course and follow-up, an initial inflammatory response was observed at the treatment sites, f

Treatment of hepatocellular carcinoma with major portal vein thrombosis by combined therapy with subcutaneous interferon-alpha and intra-arterial 5-fluorouracil; role of type 1 interferon receptor expression.

We previously reported the beneficial effects of combination therapy of interferon (IFN)-alpha/5-fluorouracil (FU) for advanced hepatocellular carcinoma (HCC) with tumour thrombi in the major portal branches. This report describes the results of longer follow-up and includes more than double the number of patients relative to the original report, and evaluates the role of IFN-alpha/type 2 interferon receptor (IFNAR2) expression on the response to the combination therapy. The study subjects were 55 patients with advanced HCC and tumour thrombi in the major branches of the portal vein (Vp3 or 4)

Treatment with interferon-α2b and ribavirin improves outcome in MERS-CoV-infected rhesus macaques.

The emergence of Middle East respiratory syndrome coronavirus (MERS-CoV) is of global concern: the virus has caused severe respiratory illness, with 111 confirmed cases and 52 deaths at the time of this article's publication. Therapeutic interventions have not been evaluated in vivo; thus, patient management relies exclusively on supportive care, which, given the high case-fatality rate, is not highly effective. The rhesus macaque is the only known model organism for MERS-CoV infection, developing an acute localized to widespread pneumonia with transient clinical disease that recapitulates mil

[Systemic mastocytosis--new therapeutic strategies].

Systemic mastocytosis is characterized by an excessive proliferation of mast cells and their accumulation in different organs. Avoidance of trigger factors leading to anaphylaxis is a general measure valid for all forms of mastocytosis. A premedication is necessary in case of surgery, anesthesia or administration of radiocontrast agents. Symptomatic treatment comprises antihistamines, anti-leukotrienes, proton pump inhibitors and topical corticosteroids. Indolent mastocytosis with refractory symptoms, the rare cases of aggressive mastocytosis with organ dysfunction and the even rarer mast cell

[Treatment of chronic hepatitis viral C: new associations].

The current treatment of patients with chronic HCV infection since ten years is pegylated interferon combined with ribavirin. This association allows a virological eradication in 55% of patients, all genotype and 40% of those infected with genotype 1, the most prevalent. Two protease inhibitors (telaprevir and boceprevir) were approved in 2011, in combination with pegylated interferon and ribavirin in both naïve and non-responder patients infected with HCV genotype 1. These new drugs allow obtaining a viral eradication in 70% of cases. Other direct acting antiviral drugs are also currently bei

Generalized flare of pustular psoriasis induced by PEGylated interferon-α2b therapy for chronic hepatitis C.

New onset or exacerbation of psoriasis vulgaris has been reported in a small number of patients after interferon (IFN)-α therapy. Herein, we report a case of generalized flare of pustular psoriasis induced by PEGylated IFN-α2b (PEG-IFN-α2b) in a 59-year-old woman with a 15-year history of pustular psoriasis and chronic hepatitis C. Interferon-α therapy was discontinued and the rash resolved after treatment with cyclosporin and systemic methylprednisolone. The potential side effect of PEG-IFN-α2b in inducing or exacerbating psoriasis should be kept in mind when treating patients with a history
