Preeclampsia at term: evidence of disease heterogeneity based on the profile of circulating cytokines and angiogenic factors.

BACKGROUND: Intravascular inflammation and an antiangiogenic state have been implicated in the pathophysiology of preeclampsia. On the basis of the profiles of their angiogenic/antiangiogenic factors, women with preeclampsia at term may be classified into 2 subgroups with different characteristics and prevalence of adverse outcomes. This study was undertaken to examine whether these 2 subgroups of preeclampsia at term also show differences in their profiles of intravascular inflammation. OBJECTIVE: This study aimed to determine the plasma profiles of cytokines and chemokines in women with pre

Interleukin-18 as a diagnostic marker of adult-onset Still's disease in older patients: a case report and review of the literature.

BACKGROUND: Adult onset Still's disease is a systemic auto-inflammatory condition of unknown etiology characterized by intermittent spiking high fever, an evanescent salmon-pink or erythematous maculopapular skin rash, arthralgia or arthritis, and leukocytosis. Recently, a high level of interleukin-18 has been reported as a new characteristic marker. On the other hand no reports have been published on high interleukin-18 as a marker in older patients. We report a case of adult onset Still's disease in an older patient successfully treated with steroids in which interleukin-18 was a useful mark

Peripheral blood mononuclear cells in patients with nonleukemic cutaneous T-cell lymphoma. Reduced proliferation and preferential secretion of a T helper-2-like cytokine pattern on stimulation.

BACKGROUND AND DESIGN: Since patients with cutaneous T-cell lymphoma (CTCL) demonstrate several immunologic abnormalities such as elevated serum immunoglobulin levels or reduced natural killer cell activity, we analyzed functional properties of the peripheral blood mononuclear cells in patients with nonleukemic CTCL and healthy donors. RESULTS: After priming with phytohemagglutinin for 3 days, a reduced proliferation in the presence of interleukin 2 and interleukin 4 was found. Limiting dilution technique revealed a diminished number of peripheral blood mononuclear cells from patients with CT

Interleukin-6, but not relaxin, predicts outcome of rescue cerclage in women with cervical incompetence.

OBJECTIVE: We investigated the potential roles of relaxin and subclinical intra-amniotic inflammation by quantitating amniotic fluid relaxin and interleukin-6 concentrations for the prediction of outcome of rescue cerclage in women with cervical incompetence. STUDY DESIGN: Cervical incompetence was diagnosed when cervical dilatation exceeded 2 cm with intact but bulging membranes and no detectable uterine activity. Each woman underwent amniocentesis to facilitate the performance of a rescue cerclage between 15 and 27 weeks of gestation (n=40 women). Forty-five additional women who underwent a

Initial exploration of the health effects on Qinghai-Tibetan Plateau yaks following short-term exposure to polystyrene microplastics: Analysis of rumen microbiota, host metabolism, antioxidant function and inflammatory responses.

Microplastics (MPs) are ubiquitous across environments including the Qinghai-Tibet Plateau. Most existing MPs studies focus on aquatic animals and rodents, while MPs influences on yaks (Bos grunniens) remain poorly understood. Using yaks as animal models, we combined metagenomics and metabolomics to explore short-term polystyrene-MPs (PS-MPs) impacts on ruminal microbiota, metabolism, antioxidant capacity and inflammation. Seven-day PS-MPs exposure reshaped rumen microbiota and elevated β-diversity. Four KEGG pathways (peptidoglycan synthesis, vitamin B6/riboflavin metabolism, terpenoid backbo

METTL3-mediated m6A modification exacerbates vascular endothelial inflammation in coronary artery disease by regulating DNMT1 mRNA stability.

Chronic vascular endothelial inflammation plays a pivotal role in the pathogenesis of coronary artery disease (CAD). N6-methyladenosine (m6A), the predominant epitranscriptomic modification in messenger RNA (mRNA), regulates pro-atherogenic inflammatory responses in coronary endothelial cells. This study explores the role of the m6A methyltransferase METTL3 in CAD-associated endothelial inflammation through its regulation of DNA methyltransferase 1 (DNMT1) mRNA stability. Using cultured human coronary artery endothelial cells (HCAECs), an apolipoprotein E-deficient (ApoE-/-) mouse model of ath

Immune markers for pulmonary aspergillosis in patients with chronic obstructive pulmonary disease: a narrative review.

COPD complicated by pulmonary aspergillosis (COPD-PA) encompasses invasive, chronic, and allergic phenotypes and is increasingly recognized as a high-burden comorbidity. Clinical recognition is often hindered by nonspecific manifestations, corticosteroid-suppressed inflammatory signs, and the suboptimal performance of serum galactomannan in non-neutropenic hosts. To define the translational utility of immune biomarkers in this population, evidence was synthesized from international guidelines and contemporary studies in COPD-enriched cohorts, and performance was appraised across diagnostic, mo

Efficacy of anakinra for refractory acute calcium pyrophosphate crystal arthritis.

Acute synovitis induced by deposition of calcium pyrophosphate (CPP) and monosodium urate crystals involves interleukin-1β production and activation. The efficacy of blocking interleukin-1β activity (with an interleukin-1 receptor antagonist [anakinra] or interleukin-1β antibody) is well documented for gout attacks but has only been reported in two single-case reports of CPP crystal-induced acute arthritis. Here we report on five cases (four males, mean age 71±27) of CPP crystal-induced inflammatory arthritis refractory and/or intolerant to usual drug therapy and efficiently treated with anaki

Survival-factor-induced phosphorylation of Bad results in its dissociation from Bcl-x(L) but not Bcl-2.

The pro-apoptotic Bcl-2-family protein Bad heterodimerizes with Bcl-2 and Bcl-x(L) in the outer mitochondrial membranes, nullifying their anti-apoptotic activities and promoting cell death. We report that interleukin-3 (IL-3) stimulation induces Bad phosphorylation and triggers its translocation from mitochondria to cytoplasm in cells expressing Bcl-x(L) but not Bcl-2. Overexpression of Bad sensitized Bcl-x(L)-expressing FL5.12 cells to apoptosis induced by IL-3 deprivation, but had no effect on the viability of cells expressing Bcl-2. IL-3 stimulation induced Bad phosphorylation at Ser-112, i…

Four-modality therapy in malignant pleural mesothelioma: a phase II study.

BACKGROUND: Treatment approaches in malignant pleural mesothelioma (MPM) patients range from mere palliation to aggressive anticancer therapy, and there is currently no consensus on the optimal therapeutic strategy. In 1999, we began a phase II study to investigate four-modality treatment of advanced stage MPM. METHODS: From 1999 to 2004, 49 patients with International Mesothelioma Interest Group stage II-III MPM underwent four-modality treatment with intrapleural preoperative interleukin-2 (18 x 10(6) UI/day for 3 days), pleurectomy/decortication, intrapleural postoperative epidoxorubicin (2

Contribution of IL-18 to atopic-dermatitis-like skin inflammation induced by Staphylococcus aureus product in mice.

Atopic dermatitis (AD) is a common inflammatory skin disease of unknown etiology. Cutaneous infection with microbes such as Staphylococcus aureus and/or skin cleansing with detergent exacerbates clinical AD. Here, we generated an AD animal model by destroying skin barrier function with detergent and subsequent topical application of protein A from S. aureus (SpA). NC/Nga mice, which genetically have reduced skin barrier function, and BALB/c mice having intact skin barrier function, were susceptible to this combination and developed severe and moderate AD, respectively, associated with dermal a

Interleukin-27 promotes oral squamous cell carcinoma pathogenesis via FSIP1-mediated activation of the PI3K-Akt signaling pathway.

OBJECTIVE: This study aimed to examine the mechanisms by which Interleukin-27 (IL-27) contributes to the pathogenesis of oral squamous cell carcinoma (OSCC) through focal adhesion-induced stemness protein 1 (FSIP1)-mediated activation of the phosphatidylinositol 3-kinase (PI3K)-protein kinase B (Akt) signaling pathway. METHODS: The effects of IL-27 on cellular proliferation, apoptosis, and migration were examined in human OSCC cell lines [squamous cell carcinoma cell line-27 (CAL-27) and squamous cell carcinoma-4 (SCC-4)] using Cell Counting Kit-8 (CCK-8) assays, flow cytometry, and wound hea

A γδ T cell-IL-3 axis controls allergic responses through sensory neurons.

In naive individuals, sensory neurons directly detect and respond to allergens, leading to both the sensation of itch and the activation of local innate immune cells, which initiate the allergic immune response1,2. In the setting of chronic allergic inflammation, immune factors prime sensory neurons, causing pathologic itch3-7. Although these bidirectional neuroimmune circuits drive responses to allergens, whether immune cells regulate the set-point for neuronal activation by allergens in the naive state is unknown. Here we describe a γδ T cell-IL-3 signalling axis that controls the allergen r

CD123 thioaptamer protects against sepsis via the blockade between IL-3/CD123 in a cecal ligation and puncture rat model.

Sepsis is one of the most common causes of death in ICU and especially is a harmful and a life-threatened disease to pediatrics in the world. It has been demonstrated that IL-3 plays an essential role in the processing of sepsis and the inhibition of IL-3 may alleviate sepsis progress. In our previous study, we selected a novel CD123 aptamer successfully which could inhibit the interaction of CD123 and IL-3. The aim of this study is to explore the protection ability of the first thioaptamer SS30 against sepsis in a cecal ligation and puncture (CLP) rat model. Serum IL-3 level of sepsis patient

Suppressor of Cytokine Signaling 3 Expression Induced by Varicella-Zoster Virus Infection Results in the Modulation of Virus Replication.

Varicella-zoster virus (VZV) is an important viral pathogen that is responsible for causing varicella (chickenpox) and herpes zoster (shingles). VZV has been shown to suppress early anti-viral innate immune responses, but the exact mechanisms are not yet well understood. Here we demonstrate that host control of VZV is impaired by the expression of suppressor of cytokine signaling (SOCS)3. We used three different cell types to characterize VZV-induced anti-viral and inflammatory responses. Infection of human fibroblasts (MRC-5) and human macrophages (THP-1) with VZV triggered upregulation of an

Evaluation of immunological and gene expression responses induced by Galectin-3 and whole-sonicated Candida krusei antigen in rats experimentally infected with Candida krusei.

BACKGROUND: Candida krusei is an emerging non-Candida albicans species increasingly associated with antifungal resistance and invasive systemic infections. Galectin-3 (GAL-3), a β-galactosidase-binding lectin, and the surfactant protein D gene are important components of innate antifungal immunity. The chemokine (C-X-C motif) ligand-1 gene plays a central role in neutrophil recruitment, whereas whole-sonicated C. krusei antigen may act as a broad fungal immunogen. AIM: To evaluate the immunological and pulmonary gene expression responses induced by GAL-3, whole-sonicated C. krusei antigen, an
