Dietary supplementation of Citrus bioflavonoids improves lactation performance in buffaloes during hot weather by regulating antioxidant capacity, immune function, and rumen microbes.

Citrus bioflavonoids (CB) are well recognized for their antioxidant, anti-inflammatory, and digestion-promoting properties. This study aimed to evaluate the effects of dietary CB supplementation on apparent nutrient digestibility, lactation performance, blood biochemical/immunological parameters, and the ruminal microbial community of dairy buffaloes during hot weather. A 35-day trial was conducted using 20 Mediterranean dairy buffaloes, which were randomly assigned to two groups: a control group (CON) fed a basal diet, and a CB-supplemented group (CB) fed the basal diet plus 20 g/d of CB. Augmented Fibroblast Growth Factor-23 Secretion in Bone Locally Contributes to Impaired Bone Mineralization in Chronic Kidney Disease in Mice.

Chronic kidney disease-mineral and bone disorder (CKD-MBD) is a systemic disorder of mineral and bone metabolism caused by CKD. Impaired bone mineralization together with increased bony secretion of fibroblast growth factor-23 (FGF23) are hallmarks of CKD-MBD. We recently showed that FGF23 suppresses the expression of tissue nonspecific alkaline phosphatase (TNAP) in bone cells by a Klotho-independent, FGF receptor-3-mediated signaling axis, leading to the accumulation of the mineralization inhibitor pyrophosphate. Therefore, we hypothesized that excessive FGF23 secretion may locally impair bo

Vitamin D, secondary hyperparathyroidism, and preeclampsia.

BACKGROUND: Secondary hyperparathyroidism, which is defined by a high concentration of intact parathyroid hormone when circulating 25-hydroxyvitamin D [25(OH)D] is low, is a functional indicator of vitamin D insufficiency and a sign of impaired calcium metabolism. Two large randomized controlled trials examined effects of calcium supplementation on preeclampsia but did not consider the vitamin D status of mothers. OBJECTIVE: We examined the association of secondary hyperparathyroidism with risk of preeclampsia. DESIGN: Circulating maternal 25-hydroxyvitamin D [25(OH)D] and intact parathyroid

Determination of lactose and xylose malabsorption in preruminant diarrheic calves.

In preliminary studies feeding the poorly absorbed carbohydrate sorbitol at 2.3 g/kg body weight as an indication of maximal fermentative capacity failed to produce the expected large increase in breath hydrogen excretion but did produce a transient diarrhea in five out of six control calves. Twelve healthy control and eighteen diarrheic calves were fed lactose or D-xylose on consecutive days at 1.15 g/kg body weight and a concentration of 46 g/L. Breath and blood samples were collected at 1 h intervals from 0 to 7 h. After administration of lactose, there was a significant increase in breath

Irritable bowel syndrome and food interaction.

Irritable bowel syndrome (IBS) is one of the most common gastrointestinal disorders in Western countries. Despite the high prevalence of this disorders, the therapeutic management of these patients is often unsatisfactory. A number of factors have been suggested to be involved in the pathogenesis of IBS, including impaired motility and sensitivity, increased permeability, changes in the gut microbiome and alterations in the brain-gut axis. Also food seems to play a critical role: the most of IBS patients report the onset or the exacerbation of their symptoms after the meals. Recently, an incre

Maternal perinatal undernutrition modifies lactose and serotranferrin in milk: relevance to the programming of metabolic diseases?

A close link between intrauterine growth restriction and development of chronic adult diseases such as obesity, diabetes, and hypertension has been established both in humans and animals. Modification of growth velocity during the early postnatal period (i.e., lactation) may also sensitize to the development of metabolic syndrome in adulthood. This suggests that milk composition may have long-lasting programming/deprogramming metabolic effects in the offspring. We therefore assess the effects of maternal perinatal denutrition on breast milk composition in a food-restricted 50% (FR50) rat model

Supply of a histidine-rich bypass protein source improves productive performance and N efficiency independently of metabolizable Met and Lys.

The milk protein equation of the Nutrient Requirements of Dairy Cattle 8th Revised Edition includes additive effects of the metabolizable (m-) supply of Met, Lys, Leu, Ile, and His. However, additive effects of those AA in dietary studies have not been reported. This study aimed to evaluate the relationship between dietary level of mHis and of mMet, mLys, and mLeu as a group (MKL) on lactation performance, N efficiency, and metabolism, with a 2 × 2 factorial arrangement of treatments. The 4 diets consisted of low (L) or high (H) mHis (HIS; 2.2 vs 2.7% MP) and L or H levels of mMet (2.4 vs 2.8%

Highly efficient transactivation by the yeast Kluyveromyces lactis transcription factor LAC9 and its inhibition by the negative regulator GAL80 in mammalian cells.

Expression of the LAC9 gene from the yeast Kluyveromyces lactis in HepG2 human hepatoblastoma cells efficiently induced luciferase expression from reporter plasmids containing the four LAC9 binding sites from the K. lactis GAL1-GAL10 gene linked to a basal promoter. Induction was approximately 100fold and was dependent on the presence of the UAS sequence and an intact reading frame in the LAC9 gene. Additional cotransfection of constructs expressing the K. lactis GAL80 gene reduced luciferase activity by up to 98%. This inhibition was not affected by addition of 14mM galactose to the medium. N

Genetic predictors of celiac disease, lactose intolerance, and vitamin D function and presence of peptide morphins in urine of children with neurodevelopmental disorders.

Gastrointestinal disturbances, nutritional deficiencies, and food intolerances are frequently observed in children with neurodevelopmental disorders (NDD). To reveal possible association of celiac disease risk variants (HLA-DQ), lactose intolerance associated variant (LCT-13910C>T) as well as variant associated with vitamin D function (VDR FokI) with NDD, polymerase chain reaction-based methodology was used. Additionally, intestinal peptide permeability was estimated in NDD patients and healthy children by measuring the level of peptides in urine using high-performance liquid chromatography. L

Milk secretion in the rabbit: changes during lactation and the mechanism of ion transport.

Changes in the yield and composition of milk and in the permeability of the mammary epithelium to labelled disaccharides and ions have been studied during lactation in rabbits of the Dutch breed. 2. Milk yield increased to reach a peak on day 20 of lactation and then declined, but by 30-32 days the yield was still relatively high. Milk [protein] and [fat] increased in late lactation; [Na] and [Cl] decreased from early to established lactation (11-14 days) and then increased, whereas milk [K] and [lactose] showed an inverse pattern to that displayed by[Na] and [Cl]. 3. At all stages of lactatio

Rotavirus regulates the gut microhabitat immune response through the IL-22/pSTAT3/RegIIIγ signaling pathway, leading to lactose intolerance.

Lactose intolerance (LI) is a significant contributor to diarrhea in infants and young children. Recent studies suggest a potential link between rotavirus (RV) infection and alterations in gut microbiota, which may play a role in the mechanism of LI. However, the precise underlying mechanism remains unclear and requires urgent clarification. In this study, we combined insights from immunology and gut microbiome research to propose that the activation of the IL-22/pSTAT3/RegIIIγ signaling pathway, triggered by gut microbiota, may be involved. To investigate this, we established a LI animal mode
