CONTEXT: More than a quarter of the human population is likely infected with soil-transmitted helminths (Ascaris lumbricoides, hookworm, and Trichuris trichiura) in highly endemic areas. The development of immunity against Ancylostoma caninum larval reinfection has been investigated in mice after the primary infection has been treated with an effective anthelmintics. Experimentally A, caninum larvae infected mice treated with levamisole (5 x 40 mg/kg), thiabendazole (5 x 200 mg/kg), oxfendazole (5 x 100 mg/kg), or albendazole (5 x 100 mg/kg) when challenged with A. caninum larvae showed 47.77 to 55.81% larval reduction from the challenge dose indicating thereby the development of partial immunity.

A 46-year-old female, otherwise healthy, developed erythematous, pseudotumoral nodules, mainly on the hands; biopsies led to the diagnosis of Woringer-Kolopp disease. Anti-anergic chemotherapy with levamisole proved hazardous but, possibly, beneficial. Classification into benign localized, fatal generalized and intermediate disease is briefly commented upon. Differential diagnosis and tumor origin are discussed; while the Merkel cell hypothesis has been recently advanced strong analogies and slight discrepancies with mycosis fungoides advocate the T cell lymphoma theory.

Pyoderma gangrenosum developed in a 45-year-old caucasian woman who had an 18 year past history of seronegative polyarthritis without any visceral or haematologic associated abnormalities. A literature review revealed only 12 reported cases. Extensive laboratory evaluation failed to detect alterations of the immune functions. The presence of a vasculitis with C3 deposits suggested that levamisole may have played a role in eliciting the lesion. Clofazimine, administered for a period of one month, was a relative failure.

An in vivo drug screen for identifying new compounds with activity against Onchocerca macrofilariae was developed using male Onchocerca gibsoni implanted subcutaneously in outbred mice. There were several similarities (Mel W, CGP 20376, CGP 6140, levamisole) and two differences (suramin, furapyrimidone) between levels of drug efficacy in this model and activity against natural infections of O. gibsoni and O. volvulus. There was considerable variation in the mouse reaction. This mouse model is a potentially useful primary screen for macrofilaricidal drugs against Onchocerca.
