Wolframin expression induces novel ion channel activity in endoplasmic reticulum membranes and increases intracellular calcium.

Wolfram syndrome is an autosomal recessive neuro-degenerative disorder associated with juvenile onset non-autoimmune diabetes mellitus and p … ctopic expression in Xenopus oocytes. Reconstitution of wolframin from oocyte membranes into planar lipid bilayers demonstrated that the protein induced a large cation-selective ion channel that was blocked by Mg2+ or Ca2+. Inositol triphosphate was capable of activating channels in the fused bilayers that were similar to channel components induced by wolframin …

Molecular characterization of CTNS mutations in Tunisian patients with ocular cystinosis.

BACKGROUND: Ocular cystinosis is a rare autosomal recessive disorder characterized by intralysosomal cystine accumulation in renal, ophthalmic (cornea, conjunctiva), and other organ abnormalities. Patients with ocular cystinosis are mostly asymptomatic and typically experience mild photophobia due to cystine crystals in the cornea observed accidently during a routine ocular examination. The ocular cystinosis is associated with different mutations in CTNS gene. Cysteamine therapy mostly corrects the organ abnormalities. METHODS: This study was performed in collaboration with the department of

Overcoming the Barrier Treatment of Ichthyosis: A Combination-therapy Approach.

Ichthyosis vulgaris is an inherited disorder of keratinization that results in asteatotic scales on extensor surfaces of the arm, legs, and trunk. A combination-therapy approach with a physiological lipid-based barrier repair topical emulsion and ammonium lactate 12% lotion applied topically was shown to be effective at four-week follow up without any untoward side effects. This combination therapy addresses the importance of caring for both the corneocytes ("bricks") and the intercellular lipid bilayer ("mortar") for optimal benefit.

Reversible tuning of membrane sterol levels by cyclodextrin in a dialysis setting.

Large unilamellar vesicles are popular membrane models for studying the impact of lipids and bilayer properties on the structure and function of transmembrane proteins. However, the functional reconstitution of transmembrane proteins in liposomes can be challenging, especially if the hydrophobic thickness of the protein does not match the thickness of the lipid bilayer. Such hydrophobic mismatch causes protein aggregation and low yields during the reconstitution procedure, which are exacerbated in sterol-rich membranes featuring low membrane compressibility. Here, we explore new approaches to

Monoclonal antibodies can uncouple the main alpha-latrotoxin effects: toxin-induced Ca2+ influx and stimulated neurotransmitter release.

A panel of monoclonal antibodies has been produced against alpha-latrotoxin using black widow spider venom. Five of them were characterized relative to their affinity for alpha-latrotoxin and ability to modify the main toxin effects--to increase calcium permeability of synaptosomes, to stimulate the neurotransmitter release and to form the ion channels in artificial lipid membrane. The results reported here show that: (i) the monoclonal antibodies do not alter the alpha-latrotoxin affinity for the membrane acceptor; (ii) two monoclonal antibodies, A6 and A24, can simultaneously inhibit the alp

Lipid-Mediated Aggregation of Influenza Fusion Peptide.

The fusion peptide (FP), a highly conserved domain within the influenza A spike protein hemagglutinin, is the only portion of the virus that interacts directly with the target membrane. FP clusters within the membrane, is necessary for viral fusion to occur, and can induce membrane poration. Here, the aggregation of FP was investigated through molecular dynamics simulations in four different membrane compositions to examine the lipid-mediated clustering and poration free energy. The formation of highly stable antiparallel FP dimers was confirmed, found to be primarily stabilized by peptide-pep

Beyond the standard model of solubilization: Non-ionic surfactants induce collapse of lipid vesicles into rippled bilamellar nanodiscs.

HYPOTHESIS: Although solubilization of lipid membranes has been studied extensively, questions remain regarding the structural pathways and metastable structures involved. This study investigated whether the non-ionic detergent Triton X-100 follows the classical solubilization pathway or if intermediate nanostructures are formed. EXPERIMENTS: Small angle X-ray and neutron scattering (SAXS/SANS) was used in combination with transmission electron cryo-microscopy and cryo-tomography to deduce the structure of mixtures of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) vesicles and Triton X-10

[Is fluidization of the phospholipid bilayer by general anesthetics a reason for general anesthesia?].

It was found that chlorpromazine and pipolfene potentiating general anaesthesia increase viscosity of phospholipid membrane in the region of polar headgroups and in the hydrophobic area near glycerol sceleton. These results (independently of potentiating mechanism) confuse a mechanism of anaesthetics action as increased fluidity of membrane phospholipids.

Tailoring Pectin-PLA Bilayer Film for Optimal Properties as a Food Pouch Material.

This study focuses on developing a biodegradable film using a novel hybrid citrus peel pectin. A bilayer approach with PLA was proposed and optimized using Response Surface Methodology (RSM) to complement pectin films' mechanical and barrier property limitations. The optimized film composition (2.90 g PLA and 1.96 g pectin) showed enhanced mechanical strength with a tensile strength (TS) of 7.04 MPa and an elongation at break (EAB) of 462.63%. In addition, it demonstrated lower water vapor (1.45 × 10-10 g/msPa), oxygen (2.79 × 10-7 g/ms) permeability, and solubility (23.53%). Compared to singl

Liposome-Supplemented Cell-free Synthesis of a Peptide that Forms β-barrel Nanopores.

Rational design of nanopore-forming proteins and peptides is a promising approach for single-molecule analysis by using custom-designed nanopores tailored to specific target molecules. One challenge in this approach is the expression or synthesis of the designed nanopores, due to their high hydrophobicity. We sought to streamline the synthesis of an artificial and highly aggregative nanopore-forming peptide using liposome-supplemented cell-free protein synthesis. By optimizing the liposomal lipid composition, we recovered yields of the expressed aggregative peptide that were suitable for funct

Membrane Mimetics Control Internal Hydration and Catalytic Activity of the Copper ATPase LpCopA from Legionella pneumophila.

Membrane protein function is strongly influenced by the surrounding lipid bilayer through specific lipid-protein interactions and collective membrane properties. Here, we investigated how the membrane environment modulates internal hydration and catalytic activity of the P1B-type copper ATPase LpCopA. LpCopA variants labeled with 6-bromoacetyl-2-dimethylaminonaphthalene (BADAN) were reconstituted in Escherichia coli lipid-doped mixed micelles, membrane scaffold protein nanodiscs, and diisobutylene-maleic acid lipid particles (DIBMALPs). Local hydration near the conserved metal-binding cysteine

Membrane interactions and pore stability of aurein 1.2 and LLAA in single- and multi-peptide systems revealed by coarse-grained molecular dynamics simulations.

CONTEXT: Aurein 1.2 is a short cationic antimicrobial peptide secreted by the Australian tree frog Litoria aurea. In this study, we investigated the membrane interactions of aurein 1.2 and its analog LLAA in single- and multi-peptide systems using model bacterial-like and eukaryotic-like lipid bilayers. LLAA exhibited stronger membrane interactions and deeper insertion than aurein, particularly in the bacterial-like membrane models. The simulations further indicated that peptide dissociation from aggregates was generally associated with increased membrane insertion. In pre-formed pore simulati…

Effects of divalent cations on diffusion dynamics of biological water confined between lipid membranes.

Biological water is an ionic solution containing both monovalent and divalent ions. However, the effects of divalent ions on the dynamics of biological water remain largely unknown. Here, we investigate how the transport dynamics of water molecules nanoconfined between lipid membranes depends on the concentration of calcium (Ca2+) and magnesium (Mg2+) ions by using molecular dynamics simulations and the generalized transport equation for biological water. We find that the diffusion coefficient of biological water monotonically increases with Ca2+ ion concentration but exhibits a largely opposi

Targeting of Acid Sphingomyelinase, a Lysosomal Membrane Protein, Using Apolipoprotein E3-containing Nanodiscs.

We developed a nanodisc (ND)-based platform with recombinant acid sphingomyelinase (ASM) embedded in a bilayer of phospholipids that are circumscribed by apolipoprotein E3 (apoE3) for targeted delivery to lysosomes. We designed a fusion protein comprising essential catalytic segments of ASM with a transmembrane helix at one end (TM-ASMcat). Purified TM-ASMcat was reconstituted with phospholipids and the N-terminal (NT) domain of apoE3 to promote the formation of NDs (TM-ASMcat-ND). Physicochemical characterization revealed the presence of both TM-ASMcat (35 kDa) and apoE3NT (24 kDa) in the pre

Let's Stalk about Membranes: Committor-Based Enhanced Sampling of Stalk Formation.

Membrane fusion is essential for cellular communication and function, and understanding how two lipid bilayers merge is the key to informing therapeutic strategies. Functionalized nanoparticles have recently emerged as synthetic fusogens, but the molecular mechanisms driving this process remain unclear, partly because fusion involves transitions over high free-energy barriers, which are difficult to capture in molecular simulations. While enhanced sampling methods can address this problem, they also rely on the definition of collective variables, which are especially hard to define for fusion …

Tumor Derived Extracellular Vesicles Modulate Gene Expression in T cells.

Extracellular vesicles (EVs) are excreted from all cells in the human body and are heterogeneous lipid bilayer particles containing proteins, RNA, DNA, and other cargo. T cells are primary players of the adaptive immune system and have a significant role in anti-cancer immunotherapies. Tumor derived EVs have diverse effects on host cells including modulating the immune response. In this study, we investigate the role acute myeloid leukemia (AML) derived EVs have on modulation of gene expression levels in three different T cell populations (CD8+, CD4+, and CD4 + CD39 + T cells). AML derived EVs

Macromolecular crowding in solution alters huntingtin interaction and aggregation at interfaces.

Huntington's disease (HD) is a fatal neurodegenerative disease caused by an extended polyglutamine (polyQ) domain within the first exon of the huntingtin protein (htt). PolyQ expansion directly invokes the formation of a heterogenous mixture of toxic htt aggregates, including fibrils and oligomers. While htt is a cytosolic protein, it also associates with numerous membranous surfaces within the cell, leading to altered organelle morphology and dysfunction. Here, the impact of macromolecular crowding on htt aggregation in bulk solution and at solid/liquid or membrane/liquid interfaces was inves

Production of a human mitochondrial ABC transporter in E. coli.

Membrane proteins play important roles in health and disease. Despite their importance, the study of membrane proteins has been significantly limited by the difficulties inherent to their successful expression, purification, and stabilization once they have been extracted from the cell membrane. In addition, expression of human membrane proteins commonly requires the use of expensive and/or time-consuming eukaryotic systems, hence their successful expression in bacteria will be obviously beneficial for experimental research. Furthermore, since lipids can have critical effects on the activity o

Partition of common anesthetic molecules in the liquid disordered phase domain of a composite multicomponent membrane.

Despite a vast clinical application of anesthetics, the molecular level of understanding of general anesthesia is far from our reach. Using atomistic molecular dynamics simulation, we study the effects of common anesthetics: ethanol, chloroform, and methanol in the fully hydrated symmetric multicomponent lipid bilayer membrane comprised of an unsaturated palmitoyl-oleoyl-phosphatidyl-choline (POPC), a saturated palmitoyl-sphingomyelin, and cholesterol, which exhibits phase coexistence of liquid-ordered (l_{o})-liquid-disordered (l_{d}) phase domains. We find that the mechanical and physical pr

Clostridium difficile toxins A and B: Receptors, pores, and translocation into cells.

The most potent toxins secreted by pathogenic bacteria contain enzymatic moieties that must reach the cytosol of target cells to exert their full toxicity. Toxins such as anthrax, diphtheria, and botulinum toxin all use three well-defined functional domains to intoxicate cells: a receptor-binding moiety that triggers endocytosis into acidified vesicles by binding to a specific host-cell receptor, a translocation domain that forms pores across the endosomal membrane in response to acidic pH, and an enzyme that translocates through these pores to catalytically inactivate an essential host cytoso
