Analysis of cholesterol in mouse brain by HPLC with UV detection.

We describe a sensitive high performance liquid chromatography (HPLC)-based method for the determination of cholesterol in brain tissue. The method does not require the derivatization of the analyte and uses separation and quantification by reversed-phase HPLC coupled to UV detection. Lipids were methanol/chloroform extracted following the method of Bligh and Dyer, and separated using isopropanol/acetonitrile/water (60/30/10, v/v/v) as mobile phase. We observed lineal detection in a wide range of concentrations, from 62.5 to 2000 ng/μL, and were able to detect a significant increase in the bra

Fibroblast Growth Factor Binding Protein 3 (FGFBP3) impacts carbohydrate and lipid metabolism.

Secreted FGF binding proteins (FGFBP) mobilize locally-acting paracrine FGFs from their extracellular storage. Here, we report that FGFBP3 (BP3) modulates fat and glucose metabolism in mouse models of metabolic syndrome. BP3 knockout mice exhibited altered lipid metabolism pathways with reduced hepatic and serum triglycerides. In obese mice the expression of exogenous BP3 reduced hyperglycemia, hepatosteatosis and weight gain, blunted de novo lipogenesis in liver and adipose tissues, increased circulating adiponectin and decreased NEFA. The BP3 protein interacts with endocrine FGFs through its

Effects of aquatic exercise on improving body composition and muscle strength in the older adults: a systematic review and meta-analysis of randomized controlled trials.

BACKGROUND: Due to the challenges posed by aging such as decreased physical abilities and higher susceptibility to chronic illnesses, effective exercise interventions are crucial for older individuals. Despite the potential benefits of aquatic exercise, there is a lack of robust evidence supporting their efficacy. To address this gap, a systematic review and meta-analysis were conducted in this study to comprehensively assess the impact of aquatic exercise on muscle strength, body composition, and related physical (flexibility, mobility) and metabolic indicators (lipid profiles) in the older a

Future perspectives for glycolipid research in medicine.

Medical interest in glycolipids has been mainly directed to the rare and complex glycosphingolipid storage disorders that are principally caused by unitary deficiencies of lysosomal acid hydrolases. However, glycolipids are critical components of cell membranes and occur within newly described membrane domains known as lipid rafts. Glycolipids are components of important antigen systems and membrane receptors; they participate in intracellular signalling mechanisms and may be presented to the immune system in the context of the novel CD1 molecules present on T lymphocytes. A knowledge of their

Resolving liquid-to-glass transitions of water under soft nanoconfinement.

Water has challenged scientists with numerous anomalies and confounding problems since the very early times. The kinetic glass transition of bulk water is a pristine example, as its location remains mostly inaccessible to experimentalists in an area below about -41 °C, where bulk water spontaneously crystallises. Confining water within nanometre-sized domains is an intriguing approach inspired by the cellular environment in nature to completely prevent crystallisation. Here we probe the existence of liquid-to-glass transitions of water nanoconfined between non-freezing lipidic bilayers and fin

Metabolic Signature of Ethanol-Induced Hepatotoxicity in HepaRG Cells by Liquid Chromatography-Mass Spectrometry-Based Untargeted Metabolomics.

Alcoholic liver disease is highly prevalent but poorly identified and characterized, leading to knowledge gaps, which impairs early diagnosis. Excessive alcohol consumption is known to alter lipid metabolism, followed by progressive intracellular lipid accumulation, resulting in alcoholic fatty liver disease. In this study, HepaRG cells were exposed to ethanol at IC10 and 1/10 IC10 for 24 and 48 h. Metabolic alterations were investigated intra- and extracellularly with liquid chromatography-high-resolution mass spectrometry. Ion mobility was added as an extra separation dimension for untargete

Quality by design-based development and validation of an HPLC method for simultaneous estimation of pregabalin and piperine in dual drug-loaded liposomes.

The current research work describes the development of a rapid HPLC method for the concurrent detection of pregabalin and piperine in dual drug-loaded nanoformulations. The primary goal was to recognize the chromatographic conditions wherein propitious segregation of the integrants with quality peaks can be attained. An attempt to expound the target analytical profile was made to accomplish this goal, and critical method attributes (CMAs), viz. percentage acetonitrile content, injection volume and pH, which affect critical quality attributes (CQAs), were identified using systemic risk analysis

Premorbid obesity, but not nutrition, prevents critical illness-induced muscle wasting and weakness.

BACKGROUND: The 'obesity paradox' of critical illness refers to better survival with a higher body mass index. We hypothesized that fat mobilized from excess adipose tissue during critical illness provides energy more efficiently than exogenous macronutrients and could prevent lean tissue wasting. METHODS: In lean and premorbidly obese mice, the effect of 5 days of sepsis-induced critical illness on body weight and composition, muscle wasting, and weakness was assessed, each with fasting and parenteral feeding. Also, in lean and overweight/obese prolonged critically ill patients, markers of m

Development and validation of TLC-densitometric method for determination of lipid A adjuvant as a bulk and in solid fat nanoemulsions.

A simple, sensitive, selective and precise high-performance thin-layer chromatographic method was developed for determination of lipid A (MPLA) adjuvant as a bulk and in solid fat nanoemulsions. Chromatographic separations were performed on thin-layer chromatography aluminum plates precoated with silica gel 60 F-254 as stationary phase and chloroform-methanol-ethyl acetate solution (10:2:4, v/v/v) as mobile phase. With this solvent system, compact spots for MPLA at Rf value 0.80 ± 0.02 were obtained. Densitometric analysis of MPLA was carried out in absorbance mode at 357 nm. Linear regression

Synthetic Cell-Based Artificial Stem Cell Niches for Hematopoietic Stem Cell Differentiation.

Hematopoietic stem cells (HSCs) receive a combination of biochemical and biomechanical signals within the bone marrow that guide their differentiation process. These include soluble factor signaling with cytokines, cellular confinement in the stem cell niche, and contact-dependent receptor-ligand interactions with stromal cells. Recreating this complex microenvironment in vitro is a principal engineering challenge for regenerative therapies and tissue engineering. While cytokines can be easily supplemented in vitro, and several systems for confined HSC culture have been developed, integrating

Analysis of flavor formation and metabolite changes during production of Double-Layer Steamed Milk Custard made from buffalo milk.

Double-Layer Steamed Milk Custard (DLSMC) is a famous traditional Chinese dessert. This study aimed to analyze the flavor and the changes in metabolites during different stages of DLSMC preparation, including raw buffalo milk, thermo-processing, first and second-layer milk skin formation. Electronic nose and electronic tongue were employed to preliminarily assess the overall flavor characteristics between these stages. The results indicated that DLSMC demonstrated increased sweetness, saltiness, and umami compared to raw buffalo milk, along with heightened levels of nitrogen oxides, methane, a

Predator-Induced Changes in Prey Behavior Alters the Lipid and Protein Content in Both Prey and Their Resources.

AbstractThe general stress paradigm predicts that when prey are exposed to predation risk they will mobilize and divert energy toward risk responses and away from costly somatic growth. However, there is mixed evidence to support this paradigm likely because of the different behavioral strategies prey employ to cope with increased predation risk. Furthermore, this paradigm has rarely been tested in marine systems, where both the prey and their resources can be expected to alter their energetics because of top-down pressure. We performed a laboratory mesocosm experiment using a rocky intertidal

Skin Hydration by Natural Moisturizing Factors, a Story of H-Bond Networking.

Dry skin is a common condition that is experienced by many. Besides being particularly present during the cold season, various diseases exist all year round, leading to localized xerosis. To prevent it, the skin is provided with natural moisturizing factors (NMFs). They are small amino acids or derivatives found in the outermost layer of the skin, the stratum corneum (SC). They are often claimed to be highly efficient humectants, increasing the water content to maintain the fluidity of the skin. However, alternative mechanisms have been proposed, suggesting that NMFs themselves may act as lipi

Affinity Purification and Single-Molecule Analysis of Integral Membrane Proteins from Crude Cell-Membrane Preparations.

The function of integral membrane proteins is critically dependent on their naturally surrounding lipid membrane. Detergent-solubilized and purified membrane proteins are therefore often reconstituted into cell-membrane mimics and analyzed for their function with single-molecule microscopy. Expansion of this approach toward a broad range of pharmaceutically interesting drug targets and biomarkers however remains hampered by the fact that these proteins have low expression levels, and that detergent solubilization and reconstitution often cause protein conformational changes and loss of membran

A hypothesized mechanism by which inhaled low-concentration ethanol vapor may bias early entry dynamics of enveloped respiratory viruses.

Enveloped respiratory viruses rely on conserved biophysical properties of their lipid envelopes for successful host-cell entry, including membrane fluidity, spike conformational mobility, and coordinated fusion activation. These properties impose variation-independent constraints that may serve as potential targets for sequence-agnostic antiviral modulation. Here, a mechanistic hypothesis is proposed in which, under physiologically humid respiratory system conditions, inhalation of low-concentration ethanol vapor may generate a transient, ethanol-enriched microenvironment at airway surface liq

Tailoring strategies for dyslipidemia management and atherosclerotic cardiovascular disease prevention in muscular dystrophy patients.

OBJECTIVES: Patients with muscular dystrophies are at increased cardiometabolic risk due to reduced mobility, increased adiposity, glucose intolerance, and dyslipidemia. While statins remain the cornerstone of cardiovascular disease prevention in the general population, their use in muscular dystrophies is complicated by the presence of underlying muscle symptoms, which can be difficult to distinguish from statin-associated myopathy. This overlap contributes to heightened concerns about statin tolerance and may impair long-term adherence. The emergence of nonstatin lipid-lowering therapies pre

Lipodystrophy Due to Adipose Tissue-Specific Insulin Receptor Knockout Results in Progressive NAFLD.

Ectopic lipid accumulation in the liver is an almost universal feature of human and rodent models of generalized lipodystrophy and is also a common feature of type 2 diabetes, obesity, and metabolic syndrome. Here we explore the progression of fatty liver disease using a mouse model of lipodystrophy created by a fat-specific knockout of the insulin receptor (F-IRKO) or both IR and insulin-like growth factor 1 receptor (F-IR/IGFRKO). These mice develop severe lipodystrophy, diabetes, hyperlipidemia, and fatty liver disease within the first weeks of life. By 12 weeks of age, liver demonstrated i

Liquid extraction surface analysis (LESA) of hydrophobic TLC plates coupled to chip-based nanoelectrospray high-resolution mass spectrometry.

Direct identification and structural characterization of analyte spots on TLC plates have always been of great interest and the development of interfaces that allow TLC to be combined with MS is making steady progress. The recently introduced liquid extraction surface analysis (LESA) approach has the potential to hyphenate TLC with MS. A mixture of lipid standards was separated on HPTLC RP-18 glass plates using chloroform:methanol :acetonitrile 2:1:1 (v:v:v) as mobile phase. After visualization with primuline dye (0.02% in acetone:water 8:2 (v:v)), LESA was performed, followed by a chip-based

Effects of sexual maturation and feeding level on fatty acid metabolism gene expression in muscle, liver, and visceral adipose tissue of diploid and triploid rainbow trout, Oncorhynchus mykiss.

In many cultured fish species, such as salmonids, gonadal development occurs at the expense of stored energy and nutrients, including lipids. However, mechanisms regulating nutrient repartitioning during sexual maturation are not well understood. This study compared sexually maturing diploid (2N) and sterile triploid (3N) female rainbow trout to investigate effects of sexual maturation on expression of 35 genes involved in fatty acid metabolism, including genes within fatty acid synthesis, β-oxidation, and cofactors of the mTOR and PPAR signaling pathways, in liver, white muscle, and visceral

Modular PEG coatings for engineering a biomimetic glycocalyx on lipid membranes.

Engineering synthetic cells with biomimetic surface architectures requires precise control over the spatial organization and mechanical properties of the membrane surface. Inspired by the protective and regulatory functions of the natural glycocalyx, we present a modular platform for constructing hierarchical, crosslinked PEG networks on fluid lipid membranes. Through spatially controlled, sequential strain-promoted azide-alkyne cycloaddition (SPAAC), we assemble a tunable, multilayered PEG mesh that mimics the dynamic, dense, and mechanically resilient architecture of the glycocalyx.
